Alzheimer neurofibrillary changes and senile plaques were found in a variety of distribution patterns and intensities in 50 brains of patients older than 73 years who had been carefully examined for signs of organic dementia in a geriatric unit. As noted by earlier observers, the severity of dementia usually corresponded to the intensity of morphologic changes. In 2 patients, clinically identical with other demented subjects, the neocortex was spared and changes were confined to the hippocampus. The same pattern was found in 2 patients with less severe dementia. No patients with isolated temporal lobe changes could be clinically identified as having so-called ‘simple senile dementia

In canine ceroid lipofuscinosis (one case studied), isocortical layer Illab pyramidal cells develop spindle-shaped enlargements of their proximal axon filled with lipopigment, a feature that can be observed in juvenile and adult type of human neuronal ceroid lipofuscinosis and in normal ageing of the human isocortex as well.

Observations in individuals have suggested that pancreatic functions may be diminished with aging. In order to explore this possibility, we measured pancreatic fluid, protein and amylase output as a function of aging in the rat. Surgical preparations of the rats included external drainage of pancreatic secretions without obstructing normal bile flow and complete diversion of gastric acid out of the duodenum to prevent endogenous secretin release. Pancreatic secretions were collected in the basal state in unanesthetized animals 24 h after surgery. Protein and amylase output were highest at 3 months of age and decreased to less than 20% of maximal values by 27 months of age. For example, amylase output decreased from 4,354 to 105 U/h/kg and protein output fell from 33.5 to 1.7 mg/h/kg between 3 and 27 months of age. Our findings indicate that aging in the rat is associated with diminished pancreatic exocrine functions. Further careful assessment of pancreatic functions in aging individuals is advisable.

Immunological studies were performed in aging C57BL/6 and BDF1 mice. In C57BL/6 mice the incidence of malignant tumor was high and their mean life span was 665 days in males and 649 days in females. In BDF1 mice, the incidence of malignant tumors was significantly lower, and their mean life span was 954 days in males and 846 days in females. Immunologically, either the onset of decline occurred earlier, the rate of the age-related decline in activity was more rapid, and/or the magnitude of decline was greater in the shorter-lived C57BL/6 than in the longer-lived BDF1 mice. The close relationship between (a) the susceptibility especially to malignant lymphoma; (b) the onset, rate and magnitude of age-related decline in T-cell-dependent immunologic activities, and (c) the life span would suggest that T-cell immunity may be playing an important role in the resistance against life-shortening malignant lymphoma in C57BL/6 mice.

Dry weight, total basic protein and myelin basic protein (MBP) in human corpus callosum tissue were found to be significantly reduced with age, confirming other reports. The heterogeneous patterns of the MBP samples were investigated and could be classified into three groups: a normal one with peak 1 larger than peak 3; an old-age pattern with peak 1 smaller than peak 3; and a pathological pattern with peak 1 greatly reduced. Because peak 1 is the only one which is significantly reduced with age, it is suggested that future investigations could be simplified. The physiological and pathophysiological role of MBP heterogeneity is discussed.

16 normal subjects, ranging in age from 24 to 79 years, underwent simultaneous central and regional (renal, hepatic and limb) hemodynamic measurements; the purpose of this investigation was to determine the relationship between these measurements and age. Cardiac index was inversely related to age, with a correlation coefficient of-0.53 (p < 0.05). Renal blood flow correlated inversely with age at an r value of-0.52 (p < 0.05). Although the correlation coefficients did not achieve statistical significance, several trends were noted with increasing age: systemic vascular resistance, renal, hepatic and limb vascular resistances, and mean systemic blood pressure increase and hepatic and limb flows decrease. In normal human subjects, certain central and regional hemodynamic parameters change with age; these changes may ultimately influence function and perhaps, drug disposition (liver and kidney) as well.

The reference intervals of 18 blood components were established for an elderly population in the 8th decade of life. The most significant findings were a broader range of values for most of the components, lower values of plasma folate and plasma potassium and higher values of erythrocyte sedimentation rate, plasma creatinine and serum cholesterol, the lattermost in women only, compared to younger people.

Using a modified sensitive and specific radioenzymatic method we have determined the normal range for plasma noradrenaline in healthy, elderly individuals. The effects of tilt on the cardiovascular system and on plasma noradrenaline responses were found to be different compared with healthy young controls. The elderly had higher resting plasma noradrenaline concentrations with an exaggerated response to head-up tilt. In spite of this higher sympathetic drive, the cardiovascular responses in the elderly were attenuated.

The foul smell of urine around patients with urine incontinence is thought to be due mainly to the production of ammonia from urea by bacterial ureases. The production of ammonia was thus measured by infrared spectrophotometry in control diapers and in test diapers impregnated with copper acetate (CA), after use by urine-incontinent patients with persistent bacteriuria. The CA-impregnated diapers produced significantly less ammonia than the control diapers. The CA-impregnated diapers were not bactericidal, however. The results are consistent with the hypothesis that the smell reduction of the CA-impregnated diapers is due to a bacteriostatic action or an inhibition of bacterial ureases by CA.
