Peripheral blood from 4 patients with progeria and their family members and 2 patients with Werner’s syndrome were analyzed for the proportion of total T cells and T cells with receptors for IgM (Tμ) or IgG (Tγ). In 3 patients proportions of Tμ cells were within the range for age- and sex-matched controls. 1 patient had significantly lower proportions of Tμ cells when compared to age- and sex-matched controls. The proportions of Tγ cells were increased in 2 and were within the range for normal controls in the remaining 2 patients. When data were analyzed with regard to Tμ/Tγ ratios, 3 of 4 patients exhibited ratios lower than the lowest limit observed in age- and sex-matched controls. Surprisingly, family members in all four families exhibited patterns of Tμ and Tγ cells similar to that seen in the patient. Thermal inactivation of hepatic succinic dehydrogenase (SDH) (EC 1.3.99.1) at 51 ± 1 °C resulted in a higher degree of inhibition in middle-aged and old than in young lizards. The oxaloacetate inhibition of SDH activity and that of oxygen uptake of liver homogenates after 1 h of incubation also showed a similar pattern of age dependency. With increase in time interval (from 15 to 60 min) the young and to some extent the middle-aged lizards showed a partial recovery from inhibition of oxygen consumption of liver homogenates, but not the old animal

Transfer RNAs and aminoacyl-tRNA synthetases were isolated from 5- and 20-day-old etiolated ageing soybean cotyledons. The relative concentrations of Mg2+ and ATP for optimal acylation (using homologous tRNAs and synthetases) for the 16 amino acids tested indicate that the amino acids can be grouped into 3 categories with a Mg:ATP ratio of 2:1, 5:1 and 10:1 (aspartic acid, isoleucine, leucine, serine, and valine). The total amino acid acceptance for 5-day-old cotyledons was 0.427 μmol/μmol tRNA and 0.349 μmol/μmol tRNA for 20-day-old cotyledons. No significant differences were found in either mitochondrial or total superoxide dismutase activity levels in two different wild-type strains of Drosophila melanogaster whose life spans differed by 40%. The strains used were Oregon R and Swedish C.

Alloxan diabetes in Swiss mice of both sexes accelerated the ageing of collagen in newly formed cotton pellet granuloma concomitant with its similar effect on dorsal skin. The degree of response of collagen to alloxanization was dependent on the duration of the treatment and the sex of the animal. It is suggested that the physiologically altered state resulting from alloxanization leads to the formation of metabolites that act as promotants of collagen cross-linking in both existing and newly formed connective tissues.

Thyrotrophin-releasing hormone and gonadotrophin-releasing hormone were measured in lumbar CSF from patients with idiopathic senile dementia, cerebral tumours and spinal disc lesions. Somatostatin was also measured in lumbar CSF from patients with dementia and patients with other neurological disorders, but the numbers involved were much smaller. The levels of these neuropeptides were significantly reduced in the patients with senile dementia. These results suggest a possible involvement of hypothalmic neuropeptides in idiopathic senile dementia.

Osmotic fragility and erythrocyte dimensions were measured in 40 normal males between the ages of 18 and 78 years. The principal independent variables correlating with osmotic fragility were mean cell haemoglobin concentration and the age of the donor. The age-related effect was shown to increase both the mean fragility of the cells and the variability of the fragilities of the cells within the individual blood sample. The physical basis for the change is an increase in the degree of isometric sphering of the erythrocytes at isotonicity. The change in shape does not appear sufficient to affect capillary perfusion significantly in normal subjects, but suggests that cell shape will increase the vulnerability of the elderly to abnormalities of the microvasculature.
