Previous in vitro studies have predicted that the type of chemical used to crosslink albumin-coated polyester arterial prostheses may influence the rate of bioerosion of the albumin layer in vivo. This study has confirmed that the healing process of this type of compound prosthesis does indeed depend on the nature and concentration of the cross-linking agent used. Four series of implantations in the thoracic aorta of dogs for scheduled periods for 4 h up to 6 months were conducted using 1.6% glutaraldehyde, 2.5% glutaraldehyde and 0.2 M carbodiimide as the alternative cross-linking agents plus a nonalbuminated preclotted polyester prosthesis which served as the control. The pathology of the explanted grafts revealed that in the short and medium term the rate of healing and the extent of tissue ingrowth was dependent initially on the presence of and later on the rate of bioerosion of the albumin layer. After 3 months in situ, the prostheses coated with albumin cross-linked with 1.6% glutaraldehyde and carbodiimide had healed more rapidly and were invaded by more extensive tissue ingrowth than the one cross-linked with 2.5% glutaraldehyde or the preclotted control. Moreover, the migration of cells over the carbodiimide-treated surface was the most fully developed and most regularly organized of all four series. Immunostaining revealed that the presence of glutaraldehyde induced an inflammatory response which failed to support the growth of normal luminal cells with the endothelial phenotype.

We performed an in vitro study to determine whether certain bacteria may lyse a commercially available fibrin seal (Beriplast®, Behringwerke, Marburg, FRG). Fibrinogen solution was mixed with actively growing bacterial cultures, and thrombin was added. During 20 days of incubation at 37 °C complete lysis was observed with a number of different bacteria, however, at different rates. Complete lysis within 1–5 days was observed for the following species (number of strains in parentheses): Streptococcus faecalis (3), Pseudomonas aeruginosa (2), Serratia marcescens (2), and Serratia liquefaciens (1). Lysis was observed after 6–10 days for Enterobacter cloacae (2), Morganella morganii (2), Escherichia coli (2), S. marcescens (1), and Klebsiella pneumoniae (2). Somatostatin, a 28-amino-acid inhibitory polypeptide has been advocated for the treatment of upper gastrointestinal bleeding and acute pancreatitis. This study examines the effect of somatostatin in acute hemorrhagic pancreatitis in piglets (n = 12), weighing 8–12 kg. Six animals served as controls, and received only fluid resuscitation (0.9%, NaCl, 20 ml/kg/h). Six animals received somatostatin treatment consisting of a 15 µg/kg bolus i.v. given simultaneously with the induction of pancreatitis, and treatment continued with an intravenous infusion (15 µg/kg/h) for 5 h. Cardiac output, heart rate, blood pressure, arterial pO2 hematocrit and serum amylase were recorded before and each hour during the experiment. Regional blood flow in the gastrointestinal area was measured using the microsphere method. The microspheres labelled with three different isotopes were administered before the experiment and at 2 and 5 h, respectively. There was a significant decrease in the cardiac output (p < 0.05) and an increase in systemic blood pressure in the somatostatin-treated group (p < 0.025). Pancreatic blood flow decreased by 43 % following somatostatin infusion. The decreases at 2 and 5 h were highly significant (p < 0.005). Blood flow to the mucosal but not muscular region of the stomach was decreased by somatostatin. This study suggests that somatostatin might be harmful in acute pancreatitis due to its adverse effects on pancreatic blood flow and cardiac output.
