Lymphocyte typing (DLA typing) will be indispensable in the future for the transplantation research in dogs. For reliable cytotoxicity testing, a nearly pure suspension of fully viable lymphocytes is the most important prerequisite. As none of the methods described up to now in connection with DLA typing fulfills this criterion and as many of them are complicated and time-consuming, a number of modifications of the Ficoll-Isopaque technique were investigated. Of all methods tested, the immediate mechanical defibrination of the blood followed by centrifugation over a Ficoll-Isopaque gradient (mixture 24:10, density 1.077 g/ml) gave the best result. This method results in a yield of 50% of the lymphocytes with 100% viability, with only a few granulocytes, and virtually without contamination with erythrocytes, thrombocytes, and fibrin. The isolation can easily be terminated within 90 min, and it allows for simple and reliable lymphocytotoxicity testing.

In 24 cats with permanent and temporary coronary artery occlusion of 45–120 min duration, the heart was subsequently completely freed from work for 120 min by a total cardiopulmonary bypass with the aid of a heart-lung machine designed for cats. Neither after permanent nor temporary coronary occlusion did the relief of the heart by cardiopulmonary bypass affect the extent of ischemic myocardial damage, recognizable after intravital fluorescent staining with acridine orange, by the estimation of the hydrogen-ion concentration in the tissue and by the macrotest with nitro-BT, without substrate addition. Two 3-m2 Landé-Edwards membrane oxygenators were linked in parallel and joined with a standard roller pump apparatus for total cardiopulmonary support during aortocoronary artery-vein bypass operations. Three 3-m2 membranes were used for patients weighing over 80 kg. Normal levels of oxygen, carbon dioxide and arterial pH were obtained when the membrane was used properly. Satisfactory platelet counts over 800,000/ml and plasma hemoglobin levels below 50–100 mg°/o documented the safe levels of blood destruction. Better postoperative pulmonary and cerebral function was noted after membrane support than with the use of the bubble oxygenator support. 12 consecutive patients, 41–103 kg, had successful partial and total cardiopulmonary support during elective aortocoronary artery-vein graft operations lasting 1 to 6.5 h. A simple technique to induce a reproducible functional stenosis of a coronary artery in acute experiments is described. The artery is narrowed by stepwise twisting of a thin copper wire until a zero flow occlusion of 10 sec is no longer followed by a period of diastolic reactive hyperemia. The degree of coronary stenosis is expressed as the diastolic hemodynamic resistance over the copper wire. This technique allows comparison from animal to animal and carries the possibility to follow changes in myocardial metabolism at a given degree of coronary stenosis.

Total hepatic blood flow and ammonia tolerance test have been measured in dogs before and after portacaval shunt. In spite of a very significant decrease of more than 50% in hepatic blood flow, the hepatic extraction efficiency remained normal. A good correlation was observed between maximal blood ammonia level during tolerance test and total hepatic blood flow. No correlation was found between blood ammonia level and hepatic extraction efficiency. In shunted dogs, the hyperammonemia during the ammonia tolerance test seemed to be determinated by the hepatic blood flow variation more than by the liver parenchymal damage.

Hepatic inflow occlusion was applied to five pigs for 15 min. Splanchnic congestion was avoided by a temporary portacaval anastomosis. Different coagulation factors were determined during and after the hepatic ischaemia. Transient increase in recalcification time and in partial thromboplastin time in siliconized glass and in untreated glass were found during the occlusion in two pigs and shortly after re-establishment of the circulation in two others. No pronounced changes in coagulation factors were noted. The possibility of a transient increase in anticoagulating substances, which might be due to either a release from the liver or a decreased clearing in the liver of anticoagulating substances during and after the ischaemic period, is discussed.

A system is described for automatic adjustment of pH in an isolated organ preservation circuit. The system incorporates a pH monitor and gasometric titrating device which has been shown to be superior to manual pH control under hyperbaric and ambient pressure conditions for periods of 12 h.

The distribution of blood flow to different areas of skin on the back of rabbits was determined by 131I-labelled macroaggregates of albumins, given intracardially in a single dose. The radioactivity was determined in 3 concentric standard samples of skin punched out of each skin area. Inflammation was induced in the areas on the right side of the back by intracutaneous injection of turpentine, the corresponding areas on the left side serving as controls. Turpentine injections were given at different intervals, up to 16 h, before determination of blood flow distribution. In normal skin, the blood flow was similar in corresponding areas on the right and left side of the back. However, it differed significantly from one area to another along the back. Turpentine inflammation was found to be associated with a diphasic increase of blood flow; an early phase completed in 2–3 h, followed by a late, more prolonged, phase. In the early phase, blood flow was markedly increased in the central zone (about 1 cm2), much less so in the intermediate and peripheral zones of the areas studied. In the late phase, blood flow was uniformly increased in the whole area (about 10 cm2). The vascular reaction to turpentine was found to be significantly influenced by the location of the inflamed area. Turpentine caused the highest blood flow in the areas which had the highest flow under normal condition
