SummaryThe activity of renal 25‐hydroxycholecalciferol‐1‐hydroxylase was studied inin vitropreparations of normal healthy and rachitic pigs. The rachitic pigs suffered from a hereditary form of vitamin‐D dependant rickets. Renal cortex homogenates and/or mitochondrial suspensions from the two groups were incubated at 37 °C for 15 to 20 min in tris‐acetate buffer at pH 7.4 in the presence of 25‐(26,27‐3H)‐hydroxycholecalciferol (25‐HCC). The 1 + 1 (V/V) methanol/chloroform extract of the incubation mixture was chromatographed on Sephadex LH 20 using a chloroform/hexane mixture 40 + 60 (V/V) as eluant. Chromatography revealed that substances more polar than 25‐hydroxycholecalciferol were formed during incubation of kidney preparations from normal pigs. The substances co‐migrated on Sephadex LH 20 with 1,25‐dihydroxycholecalciferol (1,25‐DHCC). No labelled products of this kind were detected in extracts obtained from preparations of rachitic pigs. The findings indicate that renal production of 1,25‐dihydroxycholecalciferol from 25‐hydroxyvitamin D3is either insufficient or absent in rachitic pigs. The failure to be able to convert 25‐HCC into 1,25‐DHCC is likely to be responsible for the development of rachitic symptoms in the hereditary rachitic pig strain. In Präparationen von gesunden Schweinen traten im Eluat des LH 20‐Chromatogramms markierte Substanzen mit einer größeren Polarität als 25‐HCC auf. Diese polaren markierten Reaktionsprodukte hatten dieselbe Retention wie 1,25‐Dihydroxycholecalciferol. Sie traten nur bei Inkubation von Nieren gesunder Schweine auf und waren bei rachitischen Tieren nicht nachweisbar. Die Befunde deuten darauf hin, daß bei kranken Schweinen die renale Synthese von 1,25‐Dihydroxycholecalciferol entweder nicht stattfindet oder stark gestört ist. Dieser Defekt könnte bei den an erblicher Rachitis leidenden Schweinen für die Entwicklung der Rachitis verantwortlichsein. Ces produits de réaction marqués polairement ont présenté la même rétention que 1,25‐dihydroxycholecalciferol; ils ne sont apparus que lors d'incubation des reins de porcs sains et n'ont pas été mis en évidence chez des animaux rachitiques. Les résultats montrent que la synthèse rénale de 1,25‐dihydroxycholecalciferol n'a pas lieu ou est fortement perturbée chez des porcs malades. Cette déficience pourrait être responsable du développement du rachitisme chez les porcs atteints de rachitisme héréditaire. SummaryCreatine kinase (CK) isoenzyme activity was investigated in muscle and brain tissues and in serum from pigs. Electrophoretic separation of CK isoenzymes was performed in agarose gel and the isoenzymes were estimated by fluorometric quantitation.From brain tissue homogenates (grey matter) four different CK fractions were obtained: one anodal fraction CK‐1 (CK‐BB), one intermediary fraction CK‐2 (CK‐MB) and one cathodic migrating band CK‐3 (CK‐MM); a fourth fraction, referred to as CK‐4, having an electrophoretic mobility cathodic to the application site and probably of CK‐BB nature, was detected. In myocardial tissue, a percentage composition of 95% CK‐3 (MM) and 5% CK‐2 (MB) was obtained. In striated muscle tissue only CK‐3 (MM) was detected, whereas in the intestinal mucosa four different isoenzyme bands were registered: CK‐1 (BB), CK‐2 (MB), CK‐3 (MM) and the CK‐4 fraction. In kidneys, adrenals, liver, spleen and lung tissue extracts, CK‐1 (BB), CK‐2 (MB) and CK‐3 (MM) activities were detected.In serum from growing pigs (3–6 months of age), CK‐1 (BB) and CK‐3 (MM) were regularly recorded. Trace amounts of CK‐2 (MB) were seen in most samples. Die elektrophoretische Trennung der CK‐Isoenzyme erfolgte auf Agar‐Gel und die Isoenzyme wurden mittels Fluorometrie quantitativ erfaßt.Aus den Homogenaten der grauen Substanz des Gehirns wurden 4 verschiedene CK‐Fraktionen isoliert: Eine anodale Fraktion CK‐1 (CK‐BB), eine intermediäte Fraktion CK‐2 (CK‐MB) und eine katodische Fraktion CK‐3 (MM); darüber hinaus wurde eine vierte Fraktion, CK‐4, mit einer elekro‐phoretischen Mobilität in Richtung Katode und möglicherweise von der Art der CK‐BB entdeckt.Im Myocard entfielen 95% auf die CK‐3 (MM) und 5% auf die CK‐2 (MB). La séparation électrophorétique des isoenzymes CK a été faite sur gel d'agar et les isoenzymes ont été déterminées quantitativement au moyen fluorométrie.On a isolé 4 fractions CK différentes à partir de l'homogénat de la substance grise du cerveau: une fraction anodique CK‐1 (CK‐BB), une fraction intermédiate CK‐2 (CK‐MB) et une fraction cathodique CK‐3 (MM); on a trouvé en plus une quatrième fraction CK‐4 avec une mobilité électrophorétique en direction de la cathode et probablement du genre CK‐BB.On a trouvé dans le myocarde 95% de CK‐3 (MM) et 5% de CK‐2 (MB). SummaryIn porcine serum and tissue extracts an aberrant migrating CK isoenzyme fraction with an electrophoretic mobility cathodic to the normal CK‐MM position was investigated for complex formation with serum immunoglobulins. Specific antibodies to porcine immunoglobulins (IgG and IgM) were used. This aberrant CK band, referred to as CK‐4, is probably BB in nature and is only present in serum and tissue extracts where normally migrating CK‐BB can be detected after electrophoretic separation. No complex binding to lipoproteins seems to occur spontaneously, whereas the immunoinhibition experiments indicate a complex formation between the IgG fraction and the CK‐BB isoenzyme which probably contributes to the abnormal site of this isoenzyme.ZusammenfassungEine Isoenzym‐Variante der Kreatinkinase aus Gewebeextrakten und Serum des SchweinesEin elektrophoretisch von der normalen CK‐MM‐Position aberrant in Richtung Katode wanderndes CK‐Isoenzym wurde bezüglich Komplexbildung mit Serum‐Immunglobulinen untersucht. Es wurden spezifische Antikörper gegen IgG und IgM des Schweines verwendet. Diese aberrante CK‐Fraktion, die als CK‐4 bezeichnet wird, ist möglicherweise von der Art des BB und ist nur nachweisbar im Serum und in Gewebeextrakten, in denen sich nach elektrophoretischer Trennung eine normal wandernde BB‐Fraktion erkennen läßt. Mit den Lipoproteinen scheint es keine Komplexbildung zu geben. SummaryThe histochemical reaction for myofibrillar (Ca‐ATPase with and without acid pre‐incubation was performed in samples oflongissimus dorsiandgracilismuscles in neonatal (12–48 hr) piglets. Measurements of the two basic fibre types I and II show that type I fibres have a considerably greater diameter than type II, the ratio of D1: D2being high, but the coefficients of variation are more pronounced in the case of type II fibres. Three basic types of primary bundles were seen: 1. Bundles with a single central type I fibre and other fibres of type II A. 2. Bundles with one central type I fibre with 1–2 adjacent II C fibres and II A fibres in the periphery. 3. Bundles with 2–3 fibres of type I and sometimes also 1–2 IIC fibres in the centre and the remaining fibres around them of type IIA.These bundle types are considered to represent consecutive stages in muscle differentiation. According to the histometric and type‐differentiation trends the following criteria of advanced differentiation are formulated: a decreasing D1/D2ratio, the occurence of the II C type fibre and the presence of more than one central type I fibre in the primary bundles.ZusammenfassungMorphologische und histochemische Typen‐Differenzierung bei primären und sekundären fetalen Muskelfasern von neugeborenen FerkelnBei 12–48 Stunden alten Ferkeln wurden in Proben desM. longissimus dorsiund desM. gracilisdie histochemischen Reaktionen für myofibrillär Ca‐ATPase mit und ohne Saure‐Vorinkubation durchgeführt. Messungen an den beiden Fasertypen I und II ergaben, daß der Durchmesser der Typ‐I‐Fasern beträchtlich größer war als derjenige der Typ‐II‐Fasern, so daß daraus ein hoher Quotient D1: D2resultiere. Die Variationskoeffizienten waren bei den Typ‐II‐Fasern größer. Es ließen sich 3 grundsätzliche Anordnungen von Primärbündeln unterscheiden: 1. Bündel mit einer einzigen zentralen Typ‐I‐Faser und einem Kreis von II‐A‐Fasern. 2. Bündel mit einer zentralen Typ‐I‐Faser mit 1–2 benachbarten II‐C‐Fasern und einem Kreis von II‐A‐Fasern. 3. Bündel mit 2–3 Typ‐I‐Fasern und manchmal auch mit 1–2 Typ‐II‐C‐Fasern im Zentrum, umgeben von II‐A‐Fasern.Diese Bündeltypen werden als konsekutive Stadien der Muskeldifferenzierung betrachtet. Unter Berücksichtigung der morphologischen Veränderungen werden die folgenden Kriterien als Zeichen einer fortgeschrittenen Differenzierung postuliert: Abnehmender Quotient D1/D2; Auftreten von Typ‐II‐C‐Fasern und Vorkommen von mehr als einer Typ‐I‐Faser im Primärbündel.RésuméDifférenciation morphologique et histochimique des types de fibres musculaires foetales primaires et secondaires chez des porcelets nouveaux‐nésLes réactions histochimiques pour la Ca‐ATPase myofibrillaire avec ou sans préincubation acide ont été effectuées dans des échantillons deM. longissimus dorsietM. gracilischez des porcelets âgés de 12–48 heures. RésuméUne enzootie de cancers de la vulve observée dans un troupeau de brebis est décrite. 19 tumeurs de la région ano‐vulvaire ont été découvertes dans un troupeau de 340 brebis, elles semblent plus fréquentes chez les animaux âgés.Sur le plan histologique, elles correspondent à des épithéliomas spinocellulaires.Cette étude histologique est suivie d'une discussion relative à l'étiologie de ces tumeurs. Leurs analogies nombreuses avec d'autres cas rapportés dans la littérature, leur association fréquente avec des lésions non tumorales d'hyperkératose, conduisent les auteurs à considérer ces tumeurs comme des Actinocancers.ZusammenfassungAuftreten einer enzootischen Vulva‐Karzinomatose beim SchafDas Auftreten einer enzootischen Vulva‐Karzinomatose bei Schafen wird beschrieben. In einer Herde von 340 Mutterschafen wurden 19 Tumoren in der Anogenitalregion beobachtet, wobei vor allem ältere Tiere betroffen waren. Histologisch entsprachen die Veränderungen einem Stachelzellepitheliom.Die histologischen Untersuchungen werden im Hinblick auf die Ätiologie der Tumoren diskutiert. SummaryA mixture of dieldrin (1 mg./kg./day) and phosphamidon (3 mg./kg./day) given daily to 3 goats caused death in 10, 14 and 20 days, whereas the individual compounds caused no clinical changes when the same doses were given continuously for 227 days. Nach Verabreichung beider Verbindungen wurde ein Anstieg der GOT, des Ammoniums und des Harnstoffes im Serum sowie eine stärkere Reduktion der Cholinesteraseaktivität in Serum und Gewebe beobachtet. Eine mögliche Erklärung besteht darin, daß die Wirkung des Phosphamidons durch die Dieldrinbedingte Aktivierung der mikrosomalen Leberenzyme potenziert wurde.RésuméAction combinée de la dieldrine et du phosphamidon chez des chèvresL'application orale simultanée de dieldrine (1 mg/kg/jour) et de phosphamidon (3 mg/kg/jour) chez 3 chèvres a provoqué la mort des animaux après 10, 14 et 20 jours. L'application continue d'un seul de ces composants aux mêmes doses pendant 227 jours n'a provoqué aucun symptôme clinique. Après l'application des deux éléments, on a observé une augmentation de TGO, de l'ammonium et de l'urée dans le sérum ainsi qu'une plus forte diminution de l'activité de la cholestérase dans le sérum et les tissus. Une explication possible indique que l'action du phosphamidon serait potentialisée par une activation de l'enzyme microsomique hépatique due à la dieldrine.ResumenLa acción combinada de dieldrina y fosfamidón en cabrasLa administración oral de dieldrina (1 mg./kg./día) y de fosfamidón (3 mg./kg./día) a 3 cabras condujo al cabo de 10, 14 y 20 días a la muerte de los animales, mientras que la administración continuada de cada uno de los componentes, por separado, con las mismas dosificaciones durante 227 días no provocaba ningún síntoma clínico. Tras la administración de ambas combinaciones se observó un aumento en la actividad de la GOT, en la concentración de amoníaco y de urea en el suero sanguíneo así como una reducción muy fuerte de la actividad de la colinesterasa en el suero sanguíneo y en los tejidos. SummaryThe administration of carbon tetrachloride and chloroform to goats in which the hepatic microsomal enzyme activity had been increased by administration of dieldrin or phenobarbitone was followed by marked hepatotoxicity and rapid death. The histological and enzyme‐histochemical changes in the liver and the increase in the activity of GOT and in the concentrations of bilirubin and ammonia were evidence of the hepatotoxicity of the combined effect of the drugs. The renal lesions were accompanied by increased concentrations of urea‐nitrogen and creatinine in serum. We conclude that the induction of hepatic microsomal enzymes in goats by pre‐treatment with dieldrin or phenobarbitone increases the hepatotoxic effects of carbon tetrachloride and chloroform.ZusammenfassungModifikation der Lebertoxizität von Tetrachlorkohlenstoff und Chloroform bei Ziegen durch Vorbehandlung mit Dieldrin und PhenobarbitalDie orale Verabreichung von Tetrachlorkohlenstoff und Chloroform bei Ziegen, bei denen die mikrosomale Enzymaktivität der Leber durch Dieldrin oder Phenobarbital aktiviert worden war, wirkte akut hepatotoxisch und letal. Die histologischen und enzymhistochemischen Veränderungen in der Leber sowie der Anstieg der GOT und der Bilirubin‐ und Ammoniumkonzentrationen waren Ausdruck der Hepatotoxizität der kombinierten Wirkungen der Verbindungen. Die Nierenschädigung führte zu einer Zunahme des Harnstoff‐Stickstoffes und des Kreatinins im Serum. SummaryThirty‐six buffalo heifers were given a single intramuscular injection of 33.5 mg. of PGF‐2 α or 500 μg. of Cloprostenol (a synthetic analogue of PGF‐2 α) on day 9 or day 15 of the oestrous cycle to study the jugular plasma changes in oestradiol‐17 β. Plasma oestradiol‐17 β increased significantly (P<0.001) from 8.8 ± 0.7 to 20.1 ± 3.1 pg./ml. and from 6.8 ± 1.6 to 18.3 ± 1.3 pg./ml. (Mean + SEM) after 72 hours of the treatment with PGF‐2 α on days 9 and 15, respectively. Similar treatment with cloprostenol on days 9 or 15 raised the oestradiol‐17 β level from 8.3 ± 0.7 to 24.6 ± 2.5 pg./ml. and from 6.7 ± 1.0 to 28.0 ± 6.5 pg./ml. after 72 hours of each treatment. In most of the buffaloes a sharp peak of oestradiol‐17 β was recorded a day before the onset of induced oestrus with these drugs.ZusammenfassungWirkung von Prostaglandin‐F 2 α und Cloprostenol (ICI, 80, 996) auf Plasmaöstradiol‐17 β beim Büffel (Bubalus bubalis)36 Büffelfärsen erhielten am 9. oder 15. Das Plasmaoestradiol‐17 β stieg signifikant (P<0.001) an von 8,8 ± 0,7 auf 20,1 ± 3,1 pg/ml und von 6,8 ± 1,6 auf 18,3 ± 1,3 pg/ml (Mittelwert ± SEM) innerhalb von 72 Stunden nach der Verabreichung von PGF‐2 α am Tag 9 bzw. 15. Die Behandlung mit Cloprostenol an den Tagen 9 bzw. 15 führte zu einem Anstieg des Plasmaoestradiol‐17 β von 8,3 ± 0,7 auf 24,6 ± 2,5 pg/ml bzw. von 6,7 ± 1,0 auf 28,0 ± 6,5 pg/ml 72 Stunden nach der Injektion. L'oestradiol‐17β du plasma a augmenté significativement (P<0,001) de 8,8 ± 0,7 à 20,1 ± 3,1 pg/ml et de 6,8 ± 1,6 à 18,3 ± 1,3 pg/ml (valeur moyenne ± déviation standard) en l'espace de 72 heures après l'application de PGF‐2 α au 9e et respectivement au 15e jour. Le traitement au Cloprostenol au 9e et au 15e jour a provoqué une augmentation de l'oestradiol‐17β du plasma de 8,3 ± 0,7 à 24,6 ± 2,5 pg/ml et respectivement de 6,7 ± 1,0 à 28,0 ± 6,5 pg/ml 72 heures après l'injection.
