In order to establish normal values in an adult population for serum IgE concentration, sera were obtained from 446 ambulatory Canadian Caucasian subjects with negative allergy histories. A standard isotopic procedure, the Phadebas paper radioimmunosorbent test (PRIST), was compared with the new enzymatic counterpart, the Phadezyme PRIST. By the isotopic method, serum IgE concentrations in women and men were comparable from one age group to another with no age‐related trend in the seven age groups examined (15–20, 21–30, 31–40, 41–50, 51–60, 61–70, above 70). The median and 95th percentile units (U)/ml respectively were 17.5 and 145 for 224 women and 25.5 and 275 for 222 men. Mean values ± 1 SD for women were 43 ± 102 and for men, 58 ± 137. Levels were significantly higher in men as a group. Sera with IgE concentrations above 100 U and a sampling of additional sera were tested for specific IgE antibodies to 13 common allergens by the radioallergosorbent test (RAST). After exclusion of RAST‐positive sera, the mean U/ml values ± 1 SD were 22 ± 29 for 204 women and 37 ± 54 for 196 men. Geometric mean U/ml values for these sera were 14.6 for women and 22.3 for men and the median and 95th percentile U/ml respectively for the women were 15 and 66, for the men, 24 and 135. These 95th percentile values are considered the upper limits of normal in this population. The RAST identification of antibodies to allergens to which sensitization was demonstrated provided a potential explanation for serum IgE concentrations above 100 U/ml in less than 30% of the sera in this population with negative allergy histories. The isotopic method and the counterpart enzymatic method (Phadezyme PRIST) were highly comparable; the correlation coefficient (r) for all the sera was + 0.93 (P<0.001). The occurrence of proteins cross‐reacting with allergens in hen's egg white was studied in turkey, duck, goose and seagull egg whites, in hen egg yolk, and in hen and chicken sera and flesh. The study was based upon quantitative immunoelectrophoretic techniques. The different egg whites were all found to contain proteins cross‐reacting with most of the allergens in hen's egg white, but the degree of cross‐reactivity varied considerably among the various egg whites. All egg whites contained proteins able to bind human IgE‐antibody in the sera of patients with allergy to hen's egg white. Several proteins cross‐reacting with allergens in hen's egg white were also detected in egg yolk and in hen and chicken sera and flesh. Clinical implications of the results are

The present experiments were undertaken to investigate the role of phospholipase A2(PLA2) activation in histamine release from human basophils. A PLA2inhibitor, P‐bromophenacyl bromide (BPB), inhibited IgE‐mediated anti‐IgE‐induced histamine release from human basophils with a concentration of drug required to produce 50% inhibition (IC50) of 1.5 × 10−6m when leukocytes were preincubated with this agent for 15 min. Histamine release induced by calcium ionophore A23187 and formyl‐l‐methionyl‐l‐leucyl‐l‐phenylalanine was also blocked by BPB with IC50of 4.1 × 10−6m, and 3.5 × 10−6m, respectively. A PLA2activator, 12–0‐tetradecanoylphorbol‐13‐acetate (TPA) caused basophil histamine release with a dose‐dependent fashion. BPB inhibited TPA‐induced histamine release (IC50: 2.5 × 10−6m). However, another PLA2activator, melittin, and PLA2did not release histamine through non‐cytotoxic mechanisms. Crossed immunoelectrophoresis reference patterns were established for the antigenic constituents of rat fur, urine and saliva. Using quantitative immunoelectrophoretic techniques, extensive cross‐reactivity was observed between these extracts, and with epithelia, shaved pelt, serum, faeces and dust. No major qualitative sex differences were observed, though levels of certain components differed. Constitutents of many source materials were found to show a reaction of partial identity with previously isolated rat urinary allergens; and relative skin test reactivity of a range of these extracts was associated with the amount of urinary allergen present. Circulating immune complexes (IC) were determined in sera from 41 patients with Hashimoto's thyroiditis by a polyclonal rheumatoid factor (pRF) assay based on the inhibition of the agglutination of IgG‐coated latex particles. Elevated levels of IC were found in 63% (26/41) of the sera. There was a significant correlation (Rho = 0.91,P<0.001) between results obtained before and after treatment of sera with dithiothreitol (DTT). By precipitation with 2.5% polyethylene glycol (PEG) before pRF inhibition assay, the activity of IC was found in only 7% (3/41) of the sera. Size chromatography studies of the sera showed the inhibitory activity predominantly in the intermediary region. When found in the IgM‐region the activity was not reduced by DTT. By use of a polyethylene glycol complement consumption test (PEG‐CC) the occurrence of IC was 10% (4/41). It was not possible to find any correlation between the detectable IC and the presence of microsomal, thyroglobulin, or thyroid‐stimulating antibodies. Based on our studies the sizes of IC seemed to be heterogeneously distributed and the majority were not precipitated by PEG (2.5%, final concentration). The antibodies involved in the formation of complexes seemed to be of IgG or IgA classes. HLA‐D typing of the patients showed a non‐significant association between HLA‐Dw5 and low levels of IC while the presence of HLA‐Dw4 was significantly associated with a high level

The effect of leukotriene D4(LTD4) on lung function was investigated in a controlled study on four normal subjects. A pronounced decrease in airflow was found after inhalation of less than 0.5 nmol, and was most pronounced in the variables generally accepted as indicators of the function of the small airways. No subjective symptoms were experienced.
