Occurrence of autoantibodies against nuclear material was compared in groups of patients with rheumatoid arthritis (RAn= 22), systemic lupus erythematosus (SLE.n= 24), osteoarthrosis (OAn= 25), and chronic schistosomiasis mansoni (CSMn= 28). Anti‐ds DNA antibody was detected by an ammonium sulphate precipitation radioimmunoassay, antibodies against ex tractable nuclear antigen (ENA) were detected and differentiated in RNAse‐resistant and RNAse‐sensitive components (Sm and RNP antigens with an ELISA technique. IgG organ‐non‐specific and granulocyte‐specific antinuclear antibodies (ANA) were detected by immunofluorescence technique with quantitative titration of positive reactions and determination of complement‐fixing properties, The results in groups of patients with SLE, RA and OA were of confirmative nature and supported that the different methods detect different systems of autoantibodies and nuclear autoantigens. In CSM it was demonstrated that 23 of 28 cases had positive reactions to the RNAse‐resistant part of ENA (the Sm‐antigen), a significant difference from the three other groups of patients (P<0.001). The antibody was in all cases of IgM class, in seven cases also of IgA class. Antibodies against nuclear material in CSM arc probably a consequence of heavy disturbance of the immune system in this chronic infection with great permanent antigen load. Pharmacological modulation of the immediate and late phase reaction (LPR) to anti‐human IgE was further investigated in a double‐blind cross‐over study. Tranexamic acid (AMCA) 1 g t.i.d. 24 h prior to and following intradermal injection of anti‐IgE produced an approximate 40% inhibition of the LPR (P<0.01) without affecting the early response as compared with placebo in 10 volunteers. The concept of an obligate association between mast cell activation and development of a late phase reaction (LPR) to various agents in human skin was further elucidated. Skin sites were treated four times at 24 h intervals with non‐LPR‐inducing doses of the histamine liberator compound 48/80 in 10 volunteers. Poodle and Alsatian dog dandruff extracts were characterized by crossed immunoelectrophoresis (CIE) and crossed radioimmunoelectrophoresis (CRIE) using sera from 24 individuals clinically sensitive to dogs. By using a system with intermediate gel in immunoelectrophoresis, the content of dander‐specific and serum‐specific allergens was established. 29 antigens (18 dander‐specific and 11 serum‐specific) were identified in the mixed breed Poodle/Alsation dandruff extract. Of these, 24 antigens were radiostained in CRIE. 16 allergens were dander specific and the remaining eight were serum specific. Positive dog dander RAST (e5 and Poodle/Alsatian dandruff extract) results were observed in the tested dog hypersensitive subjects. Our results suggest that the mixture of Poodle/Alsatian dandruff extract may be a suitable preparation for the diagnosis and treatment of dog

A crossed radioimmunoelectrophoretic (CRIE) method for detection of specific IgG antibodies in patients' sera against horse hair and dander was developed. The unacceptable high non‐specific binding encountered when substituting125I‐labelled antihuman IgG for125I‐labelled antihuman IgE in an ordinary CRIE was eliminated by the combined use of125I‐labelled Protein A as detector, and F(ab')2‐fragments of the allergen‐specific rabbit antibodies. The low background binding thus obtained makes the method useful for detection of specific IgG in sera where the ratio between specific and non‐specific IgG is low. In order to determine the effect of PAF‐acether on the vascular bed in the human nose, 11 healthy volunteers were studied. Rhinomanometry was performed to determine the effect on the capacitance vessels, and the133Xe wash‐out method enabled determination of the mucosal blood flow, regulated mainly by the resistance vessels. These measurements were made before and after topical administration of PAF‐acether in three different doses; 0.26 μg 2.6 μg and 26 μg per nostril. The results indicate that PAF‐acether induces a dose‐dependent vasoconstriction of the capacitance vessels and a reduction in mucosal blood flow. That PAF‐acether should be a principal mediator in the allergic reaction in the human nose is not supported by
