In order to establish reference values for total serum IgE in an adult non‐atopic population, 175 individuals. 17–85 years of age. were investigated. The usefulness of a sensitive method (PRIST®) for serum JgK determinations in discriminating atopy from mm‐atopic conditions in a Her go logical routine diagnosis was elucidated by investigating 445 patients with symptoms of asthma, rhinitis, urticaria and eczema. Comparisons were made with case histories.in vivatests and circulating IgE antibodies.The geometric mean for serum IgE in the reference material was 13.2 kU/1 with a 2 SD range of 1.53 to 114 kU/1. No significant difference between age groups or sexes was observed. In patients classified as non‐atopic and pronounced atopic, the geometric mean values for IgE were 40, 123 and 458 kU/1 respectively. The IgE level correlated with number of allergens positive in RAST and with skin test results.It is concluded that IgE determinations are of great help in discriminating atopic conditions from other diseases with similar symptoms. A serum IgE value above 100 kU/1 in a patient is strong evidence for the presence of an atopic disease while a value below 20 kU/l indicates that the symptoms are due to intrinsic or infectious

Following bronchial provocation tests with inhalent allergens, late asthmatic reactions (LAR) frequently follow immediate asthma reactions (IAR). An assay for estimation of quantity and quality of antigen‐specific IgG against two purified antigens from cow hair and dander is described. Quantitative immunoelectrophoretic methods were used for the analysis of the allergens in cow hair and dander and for comparison with related preparations. The immunoelectro‐phoretic precipitation pattern of an extract from cow hair and dander showed 17 precipitates. Crossed radioimmunoelectrophoresis showed four of these to be major allergens of the extract, and these allergens are common major allergens in four investigated cow breeds. The allergens are associated predominantly with hair and dander, but they were also demonstrated in cow saliva, urine, whey, amniotic fluid and beef, as well as in cow‐hide products. With an indirect immunofluorescencc technique 77 % of 96 patients with type I allergy and 40% of 20 patients with intrinsic bronchial asthma showed positive reactions for IgG ami‐IgG antibodies in serum. They were present partly in an aggregated state not directly detectable before treatment with dithiothreitol. The aggregates could be removed by precipitation with polyethylene glycol. The IgG ami‐IgG in hyposensitized patients were directed against both F(ab′)2and Fe fragments of rabbit IgG. Thirty of the type I allergic patients were examined once during hyposensitization as well. Before treatment 87% and IgG anti‐IgG (titres 9–72). After ≥ 13 months of treatment 100% were positive (titres 35–288), Fight patterns were also examined after hyposensitzation had been discontinued for at least 12 months. The titres of IgG ami‐IgG had then reverted lo the levels obtained before hyposensitization. Of 116 controls matched for sex and age, 79r had IgG ami‐IgG anybodies, is suggested that the production of IgG anti‐IgG may be stimulated by the presence of immune complexes and that purity, amount and/or combination of allergens administered during hyposensitization may influence the production of anti‐JgG antibodies. Bronchial anaphylactic reactions, estimated as increase in intratracheal pressure, were precipitated by intravenous injections of antigen into actively sensitized SD rats. The degree of bronchial reactivity was found 10 depend on both the challenge dose and the immunization dose of antigen; therefore the course of the capacity to respond was recorded as a function of these variables.The degree of the bronchial anaphylactic response could be reduced by pretreatment with disodium cromoglycate, terbutaline or a new anti‐allergic xanthine derivative, D 4026, in some groups of animals. 35 adult patients with atopic dermatitis were included in a double‐blind crossover trial comparing chromone‐2‐carboxylic acid, FPL 57787 18 mg × 4, with placebo. Each treatment period lasted for 6 weeks with clinical assessment every 3 weeks, 28 patients completed 3 weeks of each period. Significant differences at the 5 % level in favour of FPL 57787 were seen in the group starting on placebo. 17 patients completed the study.
