Twenty‐five antigens were demonstrated in a crudeDermatophagoides farinae(DF) extract by means of crossed immunoelectrophoresis (CIE), using a rabbit anti‐DF serum. With the same antiserum, four antigens were demonstrated in a scabies mite preparation, indicating an immunological relationship between these scabies antigens and some of the DF antigens. In tandem CIE experiments the DF antigens were found to be partly identical to three of the scabies antigens. No IgE activity against any of these scabies antigens was evident in pooled sera from patients with house dust mite allergy by crossed radioimmunoelectrophoresis. Basophils disappear after challenge with specific antigen. A human basophil degranulation test has been carried out on a slide using an enriched cell suspension. On each slide, the same volume of cell suspension was deposited in wells with antigen diluted in buffer or with buffer only (control). The basophil count was made on an equal number of randomly distributed microscopic fields either by eye (optical) or by image analyser (automatic).In 35 subjects, the correlation coefficient between the numbers of non‐degranulated basophils counted by eye and by image‐analyser on control wells was found to be r = 0.91, P<10−8.In 12 patients (6 with hydatidosis and 6 widi schistosomiasis), the percentage of degranulation with three antigen dilutions was measured. The correlation between the results obtained by eye and by image‐analyser reached 0.76 (P<10−7).The authors now use the automated measurement of human basphil degranulation

The clinical effects of intranasal budesonide (16α, 17α (22 R,S) propylmethylenedioxypre‐gana‐1‐4‐diene‐11β 21 diol‐3, 20 dione)) ‐ a new non‐halogenated glucocorticoid ‐ were evaluated in a double‐blind comparison with placebo, in patients with pronounced hay fever.Aerosolized budesonide, using a daily dosage of either 400 μg or 200 μg, showed a highly significant reduction in the nasal symptoms as compared with placebo. No significant difference could be demonstrated between the effects of 400 μg and 200 μg per day, suggesting that the lower dosage is adequate.No significant effect on the eye symptoms and no fall in plasma cortisol values were observed with either dosage. The relationship between serum levels of allergen‐specific IgE (RAST) and skin prick test reactivity and allergic disorders was evaluated in 137 subjects randomly selected from an adolescent population. AH subjects were prick tested with six common allergens, interviewed and physically examined. In addition, serum was collected for RAST analysis with three to six allergens. At least one positive RAST result (score 1–4) was observed in 40% and at least two positive RASTs in 22% of the subjects. Boys experienced more RAST reactions and generally with higher scores than girls. For instance, 26% of boys but only 11% of girls were RAST positive to timothy grass pollen.The correlation between prick test and RAST results was better with pollens than with house dust and animal epithelia. When the test results were discordant, the skin test was usually positive and RAST negative. Many of the small skin reactions (weal diameter 3–4 mm) were accompanied by a negative RAST.Respiratory allergy was closely connected with both positive skin test and RAST reactivity, while atopic dermatitis was less related. In 17% of the adolescents positive skin tests and in 14% positive RASTs occurred in the absence of any allergic symptom. In a double‐blind controlled trial 41 hospitalized adults with severe, perennial asthma of unknown aetiology were allocated to either an antigen‐free elemental diet (Vivasorb®) or control diet, i.e. blended ordinary hospital food, for 2 weeks. At the time of entry into the trial all patients were in an active but stable phase of the disease. Medical treatment was given throughout the study as clinically indicated. Peak expiratory flow was measured hourly during the daytime and patients noted their symptoms daily on an assessment form. Validation of variables according to a scoring system indicated that the elemental diet resulted in an improvement of the patients' asthma (P<0.05). 708 unselected adolescents 15–17 years old were studied. They were physically examined, interviewed and skin prick tested with 12 common allergens. Questionnaires were distributed to the adolescents and their parents to investigate past and present symptoms and environmental influences. Emphasis was on the first year of life. In accordance with the clonal selection theory we intended to prevent the development of artificially induced birch pollen allergy in rabbits with the aid of the radiolabelled' pollen allergen (75–1000 μCi125I‐pollen/animal) intravenously administered prior to pollen sensitization. The birch pollen allergen, in accordance with Burnet's working hypothesis, reacts only with a genetically determining B cell subpopulation. The fixation of the radiolabelled birch pollen allergen to the receptors of the competent B cell clone causes the lesion of the latter. Compared with the control group, this group of rabbits showed an extensive suppression of anaphylactic reagin‐like PCA‐antibodies, and haemagglutinating antibodies in the blood as well as in nasal secretion. In addition, we tried to influence the already ongoing synthesis of the antibodies with the aid of a subsequent intravenously administered radiolabelled birch pollen allergen (750–1000 μCi125I‐pollen/animal). An intensive suppression of the synthesis of antibodies could also be proved in this case. The simultaneous immunization of the control rabbits with birch pollen and egg albumin resulted in the production of antibodies against both antigens, as expected.
