A comparison was made betweenAspergillus fumigatusand A. umbrosus antigens regarding their usefulness in serological tests of farmer's lung. Mycelial and culture fluid antigens of both microbes were used to test sera of 30 Finnish farmer's lung patients for antibodies in the ELISA and in the gel precipitation (GP) test. Antibodies to A.umbrosuswere more common than to A.fumigatus; this was especially evident in ELISA. ELISA gave more positive results with all lour antigens than GP. Mycelial antigens showed more positive reactions in ELISA than the corresponding culture fluid antigens. The situation was reversed in GP. Selection of antigens in the diagnostic panel of farmer's lung requires careful consideration. Our results suggest that otherAspergilli(in the Finnish populationA. umbrosus) might be preferable toA. fumigatusin the antigen panel.

Flunisolide (F). a potent fluorinated corticosteroid or its propylene/polyethylene glycol vehicle (V) were intranasally administered to 50 patients with ragweed hayfever and 75 patients with perennial rhinitis. Each patient was randomly assigned in a double‐blind manner an indistinguishable metered pump spray device containing either F or V. Each hayfever patient administered two pulls in each nostril b.i.d. Each perennial rhinitis patient administered two puffs in each nostril t.i.d. Each puff of E contained 25 μg V. Hayfever and perennial rhinitis patients inhaling F demonstrated significantly lower symptom scores compared to their V controls. Skin test positive patients with perennial rhinitis inhaling F demonstrated significantly larger reductions in symptom scores and significandy greater subjective control of symptoms compared to their V controls than their skin test negative counterparts inhaling F and V.Morning plasma cortisol measured prior to and at the end of 1 month and 3 months of F revealed no significant diminution in cortisol level compared to patients inhaling V. Workers in the coffeee industry were investigated. Study I comprised 50 selected cases of whom 25 had work‐related symptoms and 25 had not. Prick tests and RAST investigations with different factory dust extracts were performed. Study II was a cross‐sectional study comprising 129 workers who were prick‐tested with one factory dust extract and with castor bean (CB). More than 40% described occupationally related asthma, rhintis, and in about half of these cases sensitization with one or two allergens found. One allergen comes from coffee beans and is found in the factory dust, mainly where the raw coffee is handled. In order to determine the allergenic activity of five purified horse allergens, 22 children allergic to horses according to history, skin test, and leukocyte histamine release were evaluated. Washed leukocytes from all patients were tested for allergen‐induced histamine release utilizing four epidermal horse allergens (Ags 6, 9,11, and 15) and horse serum albumin. Crossed radioimmunoelectrophoresis was carried out with a standardized horse dander extract and serum from each patient.The results showed considerable variation in the individual allergoprints. Ag 11 had the highest mean allergenic activity. Sensitivity to horse serum albumin was demonstrated three times. The effect of exclusive breast‐feeding throughout the first birch pollen season of life was examined in 59 children compared to 67 children on cow milk formula and to 27 children weaned to cow milk‐based formula duting theit first birch season. The infants were about 3 months of age in their first birch season, and allergy to birch pollen was evaluated at 5 years of age by history and prick and provocation tests. Breast‐feeding throughout the first birch season did not prevent birch pollen alletgy; a similar birch alletgy prevalence of about 10% was found in children initially fed on human or cow milk. Instead. weaning to cow milk‐based formula during the first birch season seemed to protect from subseauent development of birch pollen allergy. Analogous results were obtained regarding allergy to grass pollen. Two new β2‐adrenoceptor agonists, D‐2M3 and QH 35, have been studied in asthmatic patients. Animal experiments indicated less effect on skeletal mucsle than on bronchial muscle.D‐2343 (four i.V. cumulatively increasing doses, 0.21‐6.3 μg kg‐1min‐1) and i.v. terbutaline (four cumulative doses 0.07‐2.1 μg kg‐1min‐1) were single‐blindly compared in a randomized crossover study in eight asthmatics. It was shown that D‐2343 is an effective β2‐adrenoceptor agonist with 5‐6 limes lower potency than terbutaline. The tremor‐inducing effect of D‐2343 was the same as that of terbutaline, when the same degree of bronchodilation was obtained.The effect of QH 25 (0.25. 0.75 and 2.25 mg cumulatively) administered orally, was single‐blindly compared with the effect of oral salbutamol (2, 6 and 18 mg) in a randomized crossover study in eight asthmatics. The possible mode of action of the lately demonstrated steroid effect on the immediate type allergic reaction was investigated. The influence of a topical steroid, budesonide, on the effects that released mediators have on the nasal mucosa was also studied. A nasal histamine challenge study was performed in a double‐blind. cross‐over fashion, a 1‐weck. Pretreatment with budesonide or placebo preceding the challenge. We have examined the use of3H‐histamine for evaluatingin vitrobasophil responses. Incubation of3H‐histainine for 4 h with basophil‐enriched mononuclear fractions containing autologous plasma, resulted in the best incorporation of the label. Even so, less than 3 % of the added label was actually recovered and other leukocytes also bound or incorporated3H‐histamine. Labeled mononuclear preparations failed to release3H‐histamine when challenged with zymosan‐activated serum or anti‐IgE, but endogenous histamine released under the same experimental conditions could be determined fluorometrically. The calcium ionopbore, A23187, however, did cause a dose‐dependent release of3H‐histamine. The possibility of the penetration of pollen grains into the tracheobronchial tree is still a controversial question. To investigate this phenomenon further two different approaches were conducted: 1) Acetolysis (acidic degradation of organic constituents) of mucosal samples from different levels of the pig respiratory tract showed that specific (plane tree) inhaled pollens were not found beyond the main bronchi. 2) In contrast, a noticeable noticeable number of pollens were present in acetolyzed residues of lung surgical samples: this was shown to be due to contamination. In the first approach this problem was avoided: however, the analysis of surgical samples could be a valuable technique if suitable controls were included.
