摘要:
Retention measurements of alkyl benzenes were carried out on the synthesized silica-based octyl bonded phases. The partition-displacement model was employed to analyze chromatographic properties of these phases. Namely, the methylene selectivity data were used to calculate the equilibrium sorption constant, the surface phase composition, and the sorption excess of water for each octyl phase. The differences in the surface phase composition of the phases studied were correlated with the structural and surface properties of the modified silica gels.

摘要:
A high performance capillary electrophoresis (HPCE) assay for the quantitation of (-) and (+) pentazocine in human serum was developed. Resolution of the pentazocine enantiomers was achieved using 15mM methyl-β-cyclodextrin (Me-β-CD) in 100mM phosphate buffer pH 2.5. A 72cm uncoated fused-silica capillary at a constant voltage of 20kV was used for the analysis and 0.03mM hexadecyltrimethylammonium bromide (HTAB) was added to the buffer to decrease the adsorption of endogeneous substances onto the silica wall. The analytes of interest were extracted from serum using a solid phase extraction(SPE) procedure. The phenyl SPE cartridge gave good recoveries in excess of 90% for both (-) and (+) pentazocine, without any interferences.

摘要:
The application of capillary zone electrophoresis (CZE) to the separation and determination of creatinine in human urine with photodiode array detection was described. Optimum conditions for the separation were established. The effect of pH of the background buffer on the existence state of creatinine was investigated. Photodiode array detection permitted the rapid identification of creatinine in the samples analysed. By selecting 234 nm as monitoring wavelength, the interference from urea in urine was eliminated. By selecting a suitable pH, the interferences from other substances in urine were also eliminated. The linear range, detection limit, precision and recovery of the method were investigated.

High performance liquid chromatographic methods have been developed for the assays of idarubicin-etoposide and idarubicinondansetron mixtures in 0.9% sodium chloride injection USP. The separation and quantitation of the idarubicin-etoposide mixture was achieved on an octylsilane column at ambient temperature using a mobile phase of 73:27 v/v 0.02 M aqueous monobasic sodium phosphate - acetonitrile at a flow rate of 1.0 mL/min with UV detection at 254 nm. The separation was achieved within 13 min. The method showed linearity for idarubicin hydrochloride and etoposide in the 2–32 and 10 – 160 μg/L ranges, respectively. Accuracy and precision were in the 0.1 – 2.9% and 0.1 – 2.1% ranges, respectively, for both analytes.

摘要:
The liquid chromatographic separation of catecholamine acidic metabolites using tetrabutylammonium bromide as ion interaction agent is investigated. The effect of several experimental variables, such as mobile phase pH, organic modifier concentration, ion interaction agent concentration, and temperature is analyzed. The mobile phase composition that produces the best chromatographic resolution for the analysis of the compounds of interest is determined. The hydrodynamic voltammogram of the analytes showed that good selectivity can be achieved with electrochemical detection, allowing for the analysis of different compounds at different applied potentials. The Signal-to-Noise ratio is studied as a function of the applied potential.

摘要:
A thin layer chromatography method was developed for quantification of glucose in human blood serum using preadsorbent silica gel plates, acetonitrile-water (85:15) mobile phase, methanolic sulfuric acid detection reagent, and scanning densitometry. Results are reported for the measurement of glucose by the TLC method before and after consumption of a sucrose solution. The method was validated by determining the precision for replicate analyses and accuracy by analyzing a spiked sample and also comparing results with those given by a commercial kit intended for use by diabetics for home monitoring of glucose levels. The TLC method is simple to perform and can serve for the analysis of other sample types and as an interesting experiment illustrating quantitative TLC in college analytical chemistry or biochemistry laboratory courses.

Analytical methodology utilizing solid-phase extraction and reversed phase narrowbore high-performance liquid chromatography is described for the specific determination of the decapeptide RWJ-47428-099 in the plasma of monkeys and rats. Assay validation included the demonstration of specificity, recoverability of added standard, stability, accuracy, and precision in both matrices. Linearity was demonstrated over the concentration range of 0.031 - 3.0 μg/mL in monkeys and 0.047 - 9.20 μg/mL in rats. This methodology was subsequently implemented in support of preclinical drug metabolism investigations.
