Eight rats were trained to barpress for water reinforcement under a variable interval 60-sec operant schedule. Nine acute oral administrations of (–)Δ9-trans-tetrahydrocannabinol (Δ9-THC), in amounts ranging from 0.25 to 64 mg/kg, produced dose-related effects on responding; overall response rate increased at lower doses, while higher doses produced a suppression of responding and a corresponding loss of reinforcements. Twelve dogs were anesthetized with halothane, methoxyflurane, pentobarbital and electricity. Groups of four animals received a premedication of saline, morphine or scopolamine. In the saline and scopolamine pre-medicated animals anesthetized with the conventional chemical agents there was a progressive decrease in rectal temperature throughout the period of anesthesia. The respiratory rate also decreased. In the same animals anesthetized with electricity, the rectal temperature and rate of respiration increased progressively for 90 min and then declined slowly. The temperature and respiratory changes seen after morphine premedication during electrical anesthesia resembled those seen with conventional, chemical anesthetic agents. It is suggested that morphine premedication would be desirable in procedures using electricity as the principal anesthetic agent.

The influence of sodium salicylate, dipyrone and indomethacin on the electroencephalographic effect of morphine was quantitatively studied in rabbits bearing chronically implanted electrodes. Salicylate alone produced a stimulant EEG effect, while dipyrone and indomethacin did not significantly influence the basic EEG values. Salicylate and indomethacin significantly reduced the EEG depression of morphine, this effect being more marked against the highest dose of morphine used, while the influence of dipyrone was only mild and nonsignificant. These results are compared with previous work on the influence of anti-inflammatory drugs on morphine-induced analgesia, which appears to be unrelated with the electroencephalographic effects and possibly mediated by different independent mechanisms.

Totally isolated canine stomachs, perfused with homologous blood, were used for the study of the effect of prostaglandin-E2 (PGE2) on gastric secretion and circulation. Ex vivo stomachs were stimulated by a constant infusion of histamine into the gastric artery. When histamine-induced secretion was stabilized, PGE2 was introduced intra-arterially for 1 h. Doses of PGE2 tested were 500, 250 and 125 µg/h. After 1 h PGE2 infusion was interrupted, but histamine stimulation continued. PGE2 significantly inhibited the output of gastric juice, HCl and pepsin and reduced the concentration of HCl. The effect of PGE2 was still very marked after it was discontinued. PGE2 had a potent vasodilator action on gastric circulation: it reduced the gastric peripheral resistance and the gastric arterial perfusion pressure. This hemodynamic action of PGE2 was present only during infusion of the inhibitor
