Studies on trypsin-digested globulin degradation products given to rats intra-peritoneally or intraventricularly, revealed psychodepressive effects on the central nervous system. Peptides with a molecular weight of approximately 1,300 were the most active.

By use of two novel techniques for detecting extremely small amounts of PGs, studies were conducted elucidating the mechanism of urate crystal-induced inflammation. Rats deficient in EFA and thus deficient in PGs received injections of monosodium urate crystals into the footpad. The EFA-deficient animals developed less swelling than did normal animals. However, when a 1-ng dose of PGE1 PGE2, PGA2 or PGF2α was added along with the urate injection the swelling was enhanced to approximately normal levels. In the second technique the swelling induced by injection of two different urate crystals was compared. Urate crystals heated to 200 °C produced less swelling in normal rats than did similar urate crystals heated only to 50 °C. However when a 1-ng dose of PGE1 PGE2 or PGF2α was added to the heated urate crystals injection the swelling increased to approximately the swelling in normal controls receiving urate heated to 50 °C. 摘要:
These studies describe the effects of ethanol on the adrenal medulla of rats. Ethanol intake for 3 weeks in the form of a liquid diet increased catecholamine content by 14 %; the increase was mainly due to a rise in epinephrine content. Concomitantly, there was a 16-percent increase in PNMT activity. On withdrawal of ethanol from the diet, catecholamine synthesis from 3H-tyrosine was greatly stimulated; this effect and the behavioral symptoms of withdrawal could be partially prevented by giving an injection of ethanol. Gradual ethanol withdrawal from the diet did not lead to stimulation of the adrenal medulla.

摘要:
The anticatatonic action of various antidepressants in moderate doses and their combination with sub-effective doses of diphenhydramine was correlated with brain histamine levels. The catatonia was induced by perphenazine with a concomitant and remarkable increase in the brain histamine level. Diphenhydramine partially protected the animals from the increase in brain histamine and mitigated the catatonia. The antidepressants, i.e. monoamine oxidase inhibitors and tricyclic antidepressants, partially protected animals from both the increase in brain histamine and also the catatonia. The tricyclic antidepressants were, however, found to be more potent in both respects. The combination of sub-effective doses of monoamine oxidase inhibitors and diphenhydramine was effective in terminating experimental catatonia and the elevation of brain histamine while tricyclic antidepressants failed. A relationship between the duration of protection against catatonia and brain histamine level due to antidepressants alone and their combination with diphenhydramine was observed.

摘要:
Tranylcypromine (TCP), a monoamine-oxidase inhibitor with amphetaminelike property, has at first a depressant effect and 5 h later a stimulating effect on spontaneous motor activity and learned conditioned behaviour. This latter effect can be demonstrated by means of a modified conditioned avoidance response schedule and a specific time-schedule interval. While the depressant effect of tranylcypromine may be due to the initial increase of brain serotonin caused by this drug, its delayed stimulating effect is, very likely, related to norepinephrine brain increase occurring a few hours after TCP administration.

Injection of thiopental sodium, 25 or 40 mg/kg, i.p., into normal rats produced a prompt sleep which lasted 8.3 ± 2.9 and 16.8 ± 2.7 min, respectively. Injection of morphine sulphate, 20 mg/kg, i.p., itself produced only mild sedation but prolonged the thiopental induced sleep to 50 and 67.3 min, respectively. At 1 min after onset of sleep the brain thiopental concentration in morphine pretreated animals was significantly lower than in control animals. Furthermore the latter also awoke with higher brain thiopental concentrations; the morphine pretreated animals slept for a longer time despite lower brain thiopental concentration. The t½ of brain thiopental was longer in the morphine pretreated animals, 25 min, than in the control animals, 9 min. Distribution of thiopental in several tissues at various time periods following injection of thiopental revealed some time-related quantitative differences between the two groups but a similar qualitative pattern.
