ZusammenfassungLH‐RH ist ein im Hypothalamus produziertes Proteohormon (Decapeptid), das die Freisetzung der in Hypophyse gebildeten Gonadotropine (LH, FSH) bewirkt. Es wurde der therapeutische Effekt und die Veränderungen der Hypothalamus‐Hypophysen‐Go‐nadenachse bei praepuberalen Knaben mit einem ein‐ und beidseitigen Kryptorchismus untersucht. Dabei erhielten 10 praepuberale Knaben im Alter von 5–10 Jahren mit einem ein‐ und beidseitigen Kryptorchismus über 14 Tage täglich 20 μg LH‐RH intramusculär verabreicht. Plasmatestosteron und LH wurden vor und nach der Behandlung im Serum bestimmt. Bei weiteren drei praepuberalen Knaben mit einem ein‐ und beidseitigen Kryptorchismus führten wir eine Belastung mit 200‐ 300 μg LH‐RH i.v. durch. Bei diesen Patienten wurde Plasmatestosteron als auch das LH im Serum vor Injektion, sowie 10, 20, 30, 60, 120 und 480 Minuten nach Injektion bestimmt. Nach 20, 30, 60 Minuten kam es nach i.v. Gabe von LH‐RH zu einem signifikanten Anstieg von LH im Serum, jedoch zu keinem Anstieg des Plasmatestosteronspiegels. Klinisch erzielten wir bei 8 von 10 Knaben einen Descensus der Hoden. In den 2 Fällen, wo unsere Therapie versagte, fanden wir bei der Leistenexploration eine große Inguinalhernie, an welchen der Hoden jeweils fixiert war. Diese Studie läßt vermuten, daß die Behandlung des Kryptorchismus mit LH‐RH erfolgreich ist.ResumenSe estudian los efectos terapéuticos de la administration a corto y largo plazo de LH‐RH sintético en criptorquídicos.13 niños entre 5 y 10 años, sin signos puberales y con criptorquidia uni o bilateral recibieron LH‐RH sintética.3 pacientes recibieron de 200–300 microgramos de LH‐RH por vía endovenosa. Se determinó LH y Testosterona antes de la administration y a los 10, 20, 30, 60, 120 y 480 minutos.10 pacientes recibieron por vía intramuscular 20 microgramos de LH‐RH, durante 14 dias.Los niveles de LH aumentaron significativamente a los 10, 20, 30 y 60 minutos, siendo la respusta máxima a los 20 minutos. La Testosterona plasmática no aumentó.No se objetivó aumento de LH ni de Testosterona después de 14 dias de administración intramuscular de LH‐RH.En 6 pacientes con criptorquidia bilateral y en 2 con criptorquidia unilateral se ob‐servó un descenso testicular cuatro semanas despues de iniciar el tratamiento.Relacionan los autores los datos de su trabajo con los de otros efectuados en niños prepuberales con testes descendidos y recalcan la falte de aumento de la testosterona plasmática en pacientes prepuberales,

ZusammenfassungVerff. berichten in Verbindung mit 2 eigenen Fällen über die sogenannten hypoplastischen Zonen und die Ringtubuli der Hoden und geben eine kurze Übersicht über die in der Literatur mitgeteilten Beobachtungen.SummaryThe authors report two cases with the so called hypoplastic zones and the so called ring‐like tubules in testicular biopsies. Hypoplastic zones were found in a patient with bilateral cryptorchidism, ring‐like tubules in a patient with hypogonadotropic hypogonadism. Both histologic changes are documented by photographic plates, their etiology and significance is discussed in connection with a short review of the literature.Hypoplastic zones may be defined as isolated lesions of the testis, consisting of tubules with prepuberal features. They were originally attributed to a delayed maturation of the seminiferous epithelium as a consequence of the abnormal position of the undescended testis. More recently, they were considered to indicate congenital, intrinsic defectiveness. This theory, however, has not remained unchallenged, since hypoplastic zones can be found with some frequency in descended and apparently normal testes as well.The appreciation of intratubular bodies in ring‐like tubules has been even more inconsistent. Other names for intratubular bodies include stones, concrements and calco‐spherules. Their origin has been traced to displaced spermatogonia, to a secretory product of Sertoli cells, or to oocytes. Even their intratubular location has been disputed. Two morphologic types of bodies may be distinguished: homogeneous and laminated bodies. The latter are usually associated with typical changes of the corresponding tubules, featuring two distinct layers of cells which lie against the basement membrane and closely around the central body, respectively. While several authors have taken intratubular bodies as evidence of testicular dysgenesis, others feel that they may have no serious implications. Our demonstration of intratubular bodies in a patient with hypogonadotropic hypogonadism and thus secondary testicular failure seems to support the latter opinion.ResumenLos autores aportan dos biopsias testiculares pertenecientes a un paciente criptor‐quidico y a un hipogonadismo hipogonadotrófico en las que encuentran «zonas hipoplásicas» y «túbulos como anillos».«Zonas hipoplásicas» se pueden definir como lesiones aisladas del testiculo, consistentes en túbulos con imágen prepuberal.Originariamente, se atribuyeron a alteración de la maduración del epitelio germinal secundaria a la posición anormal del testiculo no descendido.Recientemente, se considera como defecto intrinseco, congénita. Estas «zonas hipoplásicas», no obstante, se han encontrado también en testiculos descendidos y aparentemente normales.La apreciación de cuerpos intratubulares, ha sido mäs inconstante. SummarySemen is sampled in a special receptacle allowing accurate determination of its volume and protecting the spermatozoa against light and oxygen damage.The results of visual sperm counting are improved by the use of a microchamber consisting of a punched plastic foil of constant thickness, sandwiched between two silicone‐covered slides in a metal frame, and a standardised pipetting method.A X 10 phase contrast objective is used combined with a X 32 or X 25 compensating eye‐piece fitted with a stop (approx. 2 mm Ø) inserted on the diaphragm. A microscope with a hot stage at 37° C is used.In a scanning procedure the immotile spermatozoa in the live preparation are counted first, then the whole population is killed by gentle heating (60° C) and the same microchamber is counted again. This gives sperm density, total sperm number in the ejaculate and numbers and percentages of motile spermatozoa.By repeating the procedure after incubation of the semen at 37° C, the survival rates (half‐life periods) are determined as well.ZusammenfassungDer Samen wird in einem speziellen Behälter aufgefangen, damit genaue Volumenbe‐stimmung möglich ist und die Spermatozoen gegen Licht‐ und Sauerstoffschädigung geschützt werden.Die visuelle Zählung wird in einer Mikrokammer ausgeführt, welche zusammenge‐setzt ist aus einer durchlöcherten Kunststoff‐Folie von konstanter Dicke zwischen zwei Silikone‐überzogenen Objektgläsem in einem Metallbehälter. Durch wiederholte Zählungen nach Inkubation des Samens bei 37° C kann auch der Alterungsparameter der Spermatozoen (Halbwertzeit) ermittelt werden.RésuméPour recueiller l'éjaculat une réceptacle speciale est applique, quelle permette la détermination précise du volume et quelle protecte les spermatozoïdes contre la influence nocive de la lumière et de l'oxygène.Pour compter les spermatozoïdes on utilise une chambre à fluide se composant d'une pellicule plastique avec une épaisseur constante, perforée au milieu, parmi deux porte‐objets quelles sont couvert avec une couche de silicone et portées dans une monture métallique.La méthode de pipetter est normalisée.On utilise un objectif à contraste de phase xlO et un oculair compensatoire x32 ou x25, dans lequel est aménage un disque perforé au milieu (approx. 2 mm Ø) sur le diaphragme. Le microscope est équipé d'une platine chauffante réglée à 37° C.Premier, les spermatozoides immobiles sont compté en barayant la préparation vivante, après cela tout les spermatozoïdes sont tué sur une surface de chauffe (∼ 60° C) et la mëme préparation a compté à nouveau. Se emplea un microscopio con portaobjetos caliente a 37° C. La forma de proceder es primero contar los espermatozoides inmóviles en la preparación en fresco; luego, toda la población se mata con calor (60° C), y en la misma microcámara se cuenta de nuevo. Esto da la densidad espermática, el número total de espermatozoides en el eyaculado y el número y porcentaje de espermatozoides móviles.Asimismo, se determina repitiendo el mismo procedimiento después de incubar el sémen a 37° C, el porcent

SummaryThe effects of L.H.‐R.H. loading (0.1 mg, intramusculary) on plasma levels of F.S.H., L.H., and testosterone were evaluated by applying radioimmunoassay on 15 male sterility patients and 5 normal males. The results showed that the response of testosterone to L.H.‐R.H. loading was insignificant compared to the generally sharp response of both F.S.H. and L.H., implying that in all likelihood problems in the testicles rather than the pituitary are responsible for male sterility.The data presented in this report emphasize that in endocrinological examinations, use of the L.H.‐R.H. test is evidently more helpful than relying on the conventional H.C.G. test, or the recently popularized rapid H.C.G. test, particularly in view of its availability for simultaneous and efficient evaluation of both pituitary and gonadal functions.ZusammenfassungBei 5 gesunden Männern und 15 andrologischen Patienten (Oligozoo‐ und Azoosper‐mie) wurden die Plasmawerte für FSH, LH und Testosteron mittels Radioimmunassay nach Belastung mit LH‐RH (0,1 mg i.m.) untersucht. In den Ergebnissen kann gezeigt werden, daß die Antwort von Testosteron auf die LH‐RH‐Belastung zu keinen signifi‐kanten Resultaten führt verglichen mit der allgemein schnellen Reaktion bei FSH und LH, wobei in die Überlegungen einbezogen wird, daß Probleme an den Hod en aller Wahrscheinlichkeit nach sehr viel eher als solche im Bereich der Hypophyse für die Sterilität des Mannes verantwortlich sind.Die vorgelegten Ergebnisse zeigen, daß bei endokrinologischen Untersuchungen der LH‐RH‐Test evident nützlicher ist als der konventionelle HCG‐Test oder der kürzlich populär gewordene schnelle HCG‐Test; das gilt vor allem im Hinblick auf seine Anwend‐barkeit für simultane und wirksame Auswertung der hypophysären und gonadalen Funktionen.Für LH und FSH ergibt sich 20 Min. nach Gabe von 0,1 mg LH‐RH das Maximum für Normalpersonen und Oligozoospermiepatienten, während Azoospermiepatienten das FSH‐Maximum erst nach 60 Min. aufweisen. Die Testosteronplasmawerte verhalten sich demgegenüber uneinheitlich.ResumenEn un intento de arrojar alguna luz sobre las causas de esterilidad masculina, los autores efectuaron tests de sobrecarga de LH‐RH en 15 varones estériles y 5 normales, evaluando los cambios en los niveles plasmáticos de gonadotrofinas y testosterona. Diez de los pacientes estériles presentaban oligoastenozoospermia severa (menos de 12 millones). Cuatro azoospermia y uno astenozoospermia.Se determinó por RIA los niveles basales y a los 10, 20, 30, 60 y 90 minutos de la administration intramuscular de 0,1 mg de LH‐RH sintética.En los varones normales se objetivó a los 60 minutos un pico de FSH (2'4 veces más alto que el valor basal). El pico de LH apareció a los 30 minutos con un incremento de 5'8 veces sobre el nivel basal. El pico de testosterona (1'7 veces más alto que el nivel normal) se alcanzó a los 60 minutos.En todos los pacientes estériles aumentó la FSH y LH y testosterona con la administración de LH‐RH, si bien los aumentos fueron para la FSH y para la LH y testosterona mayores que en los normales.El test de LH‐RH tiene dos ventajas sobre el test de HCG: requiere horas y no dias como este último y se puede estudiar simultáneamente la función secretora adenohipo‐fisaria y de las células de Leydig.En este trabajo se muestra que los cambios en los niveles de FSH, LH y testosterona, como respuesta a la administración, fueron marcadamente diferentes en los pacientes oligozoospérmicos y en los azoospérmicos. SummaryThe uptake of glucose and galactose‐H3by hamster seminiferous tubules was studied 4 and 8 hours after intraperitoneal injection of the tracer by means of radioautographic methods for both insoluble and water soluble substances.The uptake of glucose into macromolecules occurs mainly in Sertoli cells associated to advanced spermatids. Radioautographical reaction due to diffusible substances is scattered but more in Sertoli cells associated to spermatids in the phases of acrosome or maturation. Thus a certain metabolic relationship seems to exist between Sertoli cells and the associated spermatids. Glucose, however, is a rather inspecific tracer to follow the metabolism of complex carbohydrates in the testis. The radioautograms suggest a slow synthesis of glycogen or a rapid mobilization of that synthesized during the availability of the tracer, with possible reutilization of the soluble materials.Galactose, which is known in other tissues to contribute chiefly to the synthesis of complex polysaccharides, gives a better localization over Sertoli cells both with techniques for soluble as well as for insoluble molecules. Labelling was seen more over Sertoli cells associated to spermatids in the first two steps of spermiogenesis, but as for glucose, no clear labelling was found over the acrosomes. Mostly in freeze dried specimens, however, labelling was found in the cytoplasm of early spermatids.It is suggested that radioautography of polysaccharides at the electron microscope level may help understanding the metabolic relationships between Sertoli and germ cells.ZusammenfassungDie Aufnahme von Glucose und Galactose‐H3in die Tubuli seminiferi des Hamsters wurde 4 und 8 Stunden nach der intraperitonealen Injektion der markierten Substanz mit autoradiographischen Methoden für wasserlösliche und ‐unlösliche Substanzen untersucht.Der Einbau der Glukose in Makromoleküle findet hauptsächlich in Sertolizellen statt, die in enger Beziehung zu fortgeschrittenen Spermatiden stehen. Die durch die lös‐lichen Bestandteile verursachte autoradiographische Markierung zeigt eine diffuse Ver‐teilung, tritt jedoch besonders im Zytoplasma von Sertolizellen in der Nachbarschaft von der Akrosom‐ oder der Reifungsphase der Spermatiden in Erscheinung. Es scheint also eine gewisse Stoffwechselbeziehung zwischen den Sertolizellen und den benachbarten Spermatiden zu bestehen. Die Glukose ist jedoch ein ziemlich unspezifischer Tracer für das Studium des Kohlenhydratstoffwechsels im Hoden. Die Autoradiogramme las‐sen vermuten, daß die Synthese des Glykogens sehr langsam vonstatten geht oder eine schnelle Mobilisierung des synthetisierten Materials während der Verfügbarkeit des Tracers erfolgt mit einer möglichen Weiterverwendung der so entstandenen Substanz.Die Galaktose, von der bekannt ist, daß sie in anderen Geweben einen wesentlichen Anteil an der Synthese komplexerer Polysaccharide hat, zeigt eine bessere Lokalisierung in den Sertolizellen mit den Techniken sowohl für lösliche als für unlösliche Moleküle. Die Markierung tritt vor allem in Sertolizellen in der Nachbarschaft von Spermatiden der ersten Phasen der Spermatidendifferenzierung auf. Wie für die Glukose, ist auch hier keine eindeutige Markierung des Akrosoms festzustellen. Vor allem in gefriergetrockneten Präparaten findet man außerdem eine Markierung des Zytoplasmas früher Spermatiden.Es ist zu erwarten, daß die Autoradiographie von Polysacchariden im elektronenmi‐kroskopischen Bereich zum Verständnis von Stoffwechselbezìehungen zwischen Sertoli‐und Keimzellen beiträgt.RésuméOn a étudié l'incorporation du glucose et du galactose‐H3dans les tubules séminifères chez le Hamster, 4 et 8 heures après l'injection intrapéritonéale de l'isotope, en utilisant des techniques de radioautographie pour substances solubles et insolubles.L'incorporation du glucose dans des macromolecules a lieu dans les cellules de Sertoli associées à des spermatides müres. La réaction radioautographique due a des composés solubles est diffuse; on la distingue quand méme plus nettement dans le cytoplasme des cellules de Sertoli associées aux spermatides dans les phases de l'acrosome ou de maturation. En effet, il.semble qu'il y ait un certain rapport métabolique entre les cellules de Sertoli et les spermatides y associées. Cependant, le glucose est un précurseur peu spécifique pour l'étude du métabolisme des polysaccharides dans le testicule. Les radioautogrammes suggérent une synthèse de glycogéne très lente ou bien une mobilisation rapide aussi longtemps que l'isotope est disponible; l'utilisation des matériels solubles aussi produits paraît aussi possible.Le galactose, qui dans d'autres systémes tissulaires contribue à la synthèse de polysaccharides complexes, se révèle dans une localisation plus nette sur les cellules de Sertoli par les techniques adaptées aux matériels insolubles. Le marquage est plus im‐i‐portante dans les cellules de Sertoli associées aux spermatides dans les deux premières phases de la spermiogenèse. Mais, comme dans le cas du glucose, on n'a pas trouvé de marquage net dans les acrosomes. Cependant, surtout dans les préparactions réalisées par congélation et dessication, on met en évidence le marquage dans le cytoplasme des spermatides jeunes.La radioautographie à haute résolution peut être utile pour l'analyse des rapports métaboliques entre les cellules de Sertoli et les cellules germinales.ResumenLa incorporación de glucosa y galactosa‐H3en tubulos seminiferos de hamster se estudió a las 4 y 8 horas luego de injección intraperitoneal de la marca, por medio de radioautografia para sustancias solubles e insolubles.La incorporación de glucosa en macromoléculas tiene lugar en las células de Sertoli asociadas a espermátidas tardías. La reacción radioautográfica debida a compuestos solubles se presenta difusa pero especialmente en el citoplasma de células de Sertoli asociadas a espermátidas en las fases de acrosoma o de maduración. Por lo tanto, parece haber una cierta relación metabólica entre las células de Sertoli y las espermátidas asociadas a ellas. La glucosa es, sin embargo, un trazador poco especifico para el estudio del metabolismo de hidratos de carbono en el testiculo. Los radioautogramas sugieren que la sintesis de glicógeno es lenta o que una rápida movilización ocurre durante el período de disponibilidad de la marca, siendo posible la utilización de los materiales solubles asi originados.La galactosa, que en otros sistemas tisulares contribuye principalmente a la síntesis de polisacáridos complejos, da una mejor localización sobre las células de Sertoli tanto con las técnicas para material soluble como insoluble. SummaryEarly effects of two benzimidazole‐derivatives (U 32.104 and U 32.422 E), both having male antifertility activity, were studied in rats using transillumination of freshly isolated seminiferous tubules and phase contrast microscopy of freshly isolated cells as rapid screening methods. The findings were confirmed with conventional light microscopy of paraffin embedded PAS‐hematoxylin stained sections. Transillumination and phase contrast microscopy were useful techniques permitting rapid orientation of early specific damages. One to 4 hours after administration oflarge doses of both drugs spermatids at stages VII b–c and IX–XIV–I lost their contact with the Sertoli cells. Smaller doses caused cell death in meiotic division and in stage I b in young spermatids. The exfoliated cells were later found in the initial segment of the epididymis. It is postulated that these drugs in high doses primarily affect Sertoli cells and therefore induce premature release of spermatids.ZusammenfassungDie frühen spezifischen Wirkungen von zwei Benzimidazol‐Derivaten U 32.104 und U 32.422 E, beide mit männlicher Antifertilitätswirkung, wurden an Ratten studiert. Als erste Stufe in der Analyse diente ein einfacher Durchleuchtungstest der frisch isolierten lebendigen Samenkanälchen. Die beschädigten Segmente erschienen trüb und eine schnelle Orientierung über die groben Veränderungen war möglich. Danach folgte eine phasenkontrastmikroskopische Analyse der Zellen und eine gewöhnliche Lichtmikroskopie der Paraffin‐eingebetteten und PAS‐Haematoxylin gefärbten Schnitte.Eine hone Dosis von beiden Arzneien verursachte eine Loslösung der späten Sper‐matiden in den Stadien VII b–c und IX–XIV–I von den Sertolizellen schon 1–4 Stunden nach der Injektion. Kleinere Dosen verursachten einen Zelltod bei den meio‐tischen Zellteilungen und in den jungen Spermatiden in der Stufe I b. Die losgelösten Zellen wurden später in dem Initialsegment der Epididymis gefunden.Es wurde festgestellt, daß die Arzneien einen Primäreffekt auf die Sertolizellen haben und dadurch ein vorzeitiges LosloUsen der späten Spermatiden verursachen.ResumenLa mayoria de compuestos conocidos que inducen infertilidad masculina por supresión de la espermatogénesis son – inutilizables en el hombre, debido a la toxicidad general o al potencial mutágeno de los mismos.Los autores han desarrollado una técnica para análisis – morfológicos en fresco de túbulos seminiferos aislados, de rata usando un test simple de transiluminación.A 42 ratas macho Sprague‐Dawley se les administró intraperitonealmente una solución acuosa de U‐32.422 E, ó U‐32.104 oralmente.Un gran número de derivados benzimidazoles, tienen actividad antitumoral, actuando, al parecer como venenos del huso. Estas substantias causan un significativo aumen‐to en el % de mutaciones.Ambas drogas mostraron el mismo efecto:a) rápida exfoliación de espermátides si la dosis eraelevada.b) a dosis bajas se observó muerte celular en división – meiótica, siendo el primer signo de muerte celular, – observado a las 9 horas de la adminsitración del fármaco.La exfoliación de los espermátides que sigue al tratamiento con estas drogas es debida, según los autores a alteraciones de la función sertoliana y no a alteracio

ZusammenfassungDas Verteilungsmuster der DNase‐Aktivitäten im Seminalplasma von 9 Normalpersonen und 18 Patienten mit einem pathologischen Spermabefund wurde mit dem mikro‐disc‐elektrophoretischen Verfahren untersucht. Dabei ergaben die Aktivitätsbestimmungen der DNase bei Patienten mit einer Oligoasthenoteratozoospermie als auch bei alien Probanden, die einen pathologischen Spermabefund aufwiesen, in der 1. und 2. Bande deutliche Aktivitätsminderungen im Vergleich zum Seminalplasma von Normalpersonen. Nach den vorliegenden Untersuchungsbefunden stellt sich die Frage, ob den DNasen im Seminalplasma die Aufgabe zukommt, geschädigte und überalterte Spermatozoen, deren Membran permeabler geworden ist, abzubauen oder sich andererseits eine Zunah‐me pathologischer Spermienformen erst als Folge einer zu geringen DNasen‐Aktivität einstellt.SummaryThe distribution pattern of deoxyribonuclease (DNase) activities in the seminal plasma of 9 healthy subjects and 18 patients with pathological spermatic findings has been examined by micro‐disc‐electrophoresis. Thereby the determination of DNase activities as well of patients with a oligozooasthenoteratospermia as well of other testpersons with pathological spermatic findings resulted in significant decrease of activity of the first and second band in comparison with seminal plasma of normal persons. By these findings the question arises whether the DNases in seminal plasma may have a function of digestion i.e. elimination of weak and aged spermatozoa with relative permeability or otherwise the increase of pathological spermatozoa is a consequence of low DNase activities.ResumenSe ha examinado, por microdiscos electroforéticos, el patrón de distribución de la actividad desoxirribonucleasa en el plasma seminal de 19 hombres sanos y de 18 pacientes con alteraciones espermáticas La determination de la actividad DNAasa tanto en pacientes con oligoastenoteratospermia como en otras condiciones patológicas espermáticas, resultó en una disminución significativa de la actividad de la primera y segunda banda, en comparación con el plasma seminal de personas normales.De estos hallazgos surge la cuestión de si las DNAasas pueden tener, en el plasma seminal, una función de digestión, es decir, eliminación de espermatozoides débiles y viejos con permeabilidad relativa; o si, por el contrario, el aumento de espermatozoides patológicos es consecu
