Summary:Transferrin (TF) concentrations were assessed in semen from 10 fertile controls and 79 infertile patients by a radioimmunoassay method. A positive correlation between seminal TF concentration and sperm count was found in the whole sample of patients and within the control group. A statistically significant difference was found between azoospermic and severely oligospermic patients and all the others. No correlation was found with other seminal parameters or hormonal values.Zusammenfassung:Es wird eine Untersuchung über die Transferrinkonzentration (TF) im Sperma bei 10 fertilen Männern und bei 79 infertilen Männern vorgestellt; Methode: Radioimmunassay. Es ergab sich eine positive Korrelation zwischen der Sperma‐TF‐Konzentration und der Spermatozoendichte in der Gesamtgruppe an Patienten und in der Kontrollgruppe. Summary:Four hundred and forty couples were treated by artificial insemination using fresh donor semen. The main indication was azoospermia. The overall pregnancy rate was 84, 9% resulting in 287 take‐home babies. The rate of ectopic pregnancies, perinatal deaths and spontaneous abortions were comparable to the general population. The accumulating pregnancy rate within 5 months was 86.8. Women older than 35 years had similar pregnancy rates than younger patients. Socioeconomic class, length of infertility and ovulatory disturbances either before or during treatment affected significantly the pregnancy rates.Zusammenfassung:Bei 440 infertilen Paaren wurde künstlich eine heterologe Frischsamen‐Insemination vorgenommen. Die häufigste Indikation war therapieresistente Azoospermie. Die Schwangerschaftsrate war 84,9%, und 287 gesunde Kinder wurden entbunden. Die akkumulative Schwangerschaftsrate innerhalb 6 Monaten lag bei 86,8%. Bei Frauen, welche älter als 35 Jahren waren, wurde eine ähnliche Schwangerschaftsrate wie bei jüngeren Frauen beobachtet. Summary:Total, free and bound sialic acid concentrations were determined in sperm‐free luminal fluid removed by micropuncture from different regions of the rat and hamster epididymides. In the rat, total sialic acid concentrations (mean ± s.e.m.) in the proximal caput, the mid corpus and the proximal cauda were, respectively, 25.7 ± 1.3, 23.9 ± 1.7 and 28.8 ± 1.9 mM compared to 4.4 ± 0.1 mM in blood plasma. In the hamster, total sialic acid concentrations in the distal corpus, the proximal cauda and the distal cauda were, respectively, 32.9 ± 3.8, 26.4 ± 1.4 and 26.6 ± 3.2 mM compared to 4.7 ± 0.5 mM in blood plasma. Free sialic acid accounted for approximately 70–80% of total sialic acid present in the epididymal plasma of the rat. Similarly, 82% of sialic acid in the rat blood plasma was in free form. The levels of free and bound sialic acid were not changed in different regions of the rat epididymis. Unilateral ligation of the rat efferent ducts had no effect on total, free or bound sialic acid concentrations in all regions except in the proximal cauda in which a transient increase (P<0.01) in free sialic acid was observed on Day‐3 after EDL.Zusammenfassung:Im spermatozoenfreien Sekret des Ratten‐ und Hamsternebenhodens wurden Gesamt‐, freie und gebundene — Sialinsäure bestimmt, nachdem das Sekret durch Mikropunktion gewonnen war. Bei der Ratte ergaben sich unterschiedliche Konzentrationen für Gesamtsialinsäure (GS) in den verschiedenen Nebenhodenabschnitten (Kopf: 25.7 ± 1.3 mM; Mittelteil: 23.9 ± 1.7 mM; Schwanz: 28.8 ± 1.9 mM) gegenüber einer Konzentration von 4.4 ± 0.1 mM im Blutplasma. Beim Hamster ergab sich folgendes: Distaler Corpus: 32.9 ± 3.8 mM; proximale Cauda: 26.4 ± 1.4 mM; distale Cauda: 26.6 ± 3.2 mM gegenüber einer Konzentration von 4.7 ± 0.5 mM im Blutplasma. Freie Sialinsäure wurde für ungefähr 70–80% der Gesamtsialinsäure berechnet, die im Nebenhodenplasma der Ratte vorhanden war. In ähnlicher Weise lag bei der Ratte 82% der Sialinsäure im Blutplasma in freier Form vor. In den verschiedenen Abschnitten des Rattennebenhodens ergaben sich keine Veränderungen der Werte für freie und gebundene Sialinsäure. Eine einseitige Ligatur der Ductus efferentes zeigte bei der Ratte keinerlei Einfluß auf die Werte für Gesamt‐, sowie freie oder gebundene Sialinsäure mit Ausnahme der proximalen Cauda, bei der ein vorübergehender Anstieg (p<0.01) an freier Sialinsäure am d

Summary:A morphometric study of the volumetric parameters of human spermatozoa has been carried out, volume densities being obtained by point counting and converted to absolute volumes by employing the point‐sampled intercept method. Data for the Spermatozoa from the semen of 15 recently fertile men are presented. The significance of the results is discussed in relation to results obtained by other methods and in relation to the building of a diagnostic reference database.Zusammenfassung:Eine morphologische Studie der volumetrischen Parameter von menschlichen Spermatozoen wurde durchgeführt, wobei die Volumendichte durch Punktauszählung erhalten und in absolute Volumen vermittels der Punkt‐Stichproben Schnittpunkt‐Methode umgewandelt worden ist. Daten für die Spermatozoen von 15 bis vor kurzem fruchtbaren Männern werden vorgelegt. Die Bedeutung der Ergebnisse wird in Beziehung gesetzt zu Ergebnissen, die durch andere Methoden erzielt worden sind, und in Beziehung auf diagnostische Bezugsdatenbasis

Summary:In this study we investigated whether elevated levels of the inflammatory mediator granulocyte elastase in seminal plasma were associated with increased numbers of CD4+ T helper/inducer lymphocytes and monocytes/macrophages in semen, the principal host cells of the human immunodeficiency virus (HIV). Semen samples were obtained from 105 men attending an infertility clinic. CD4+ lymphocytes, monocytes/macrophages and cells expressing the common leukocyte antigen (CD45) were identified by monoclonal antibodies (MAb's) in a biotin‐streptavidin immunoperoxidase technique. Granulocyte elastase levels in seminal plasma were determined by an enzyme‐linked immunosorbent assay. In 17 men, granulocyte elastase levels were higher than 1000 ng/ml seminal plasma, indicating male genital tract inflammation. Compared to men with low/normal granulocyte elastase levels in semen (<250 ng/ml), these men showed significantly higher mean numbers of total leukocytes, CD4+ lymphocytes and monocytes/macrophages in semen (P<0.001); median cell numbers for the group with high/inflammatory granulocyte elastase levels were increased 38‐fold for total leukocytes (19,800,000 versus 520,625 per ejaculate), 19‐fold for monocytes/macrophages (2,594,000 versus 134,565), and 6‐fold for CD4+ lymphocytes (82,900 versus 14,100). Because of the increased numbers of potential HIV‐host cells in inflammatory semen, male genital tract inflammation may be an important cofactor in the sexual transmission of the human immunodeficiency virus.Zusammenfassung:Wir untersuchten, ob ein Zusammenhang besteht zwischen erhöhten Seminalplasmaspiegeln des Entzündungsparameters Granulozytenelastase und erhöhten Zahlen von CD4+‐Lymphozyten und Monozyten/Makrophagen im Sperma; letztere sind potentielle Wirtszellen des Human Immunodeficiency Virus (HIV). In 105 Ejakulaten von Infertilitätspatienten wurden anhand von monoklonalen Antikörpern in einer Immunperoxidasetechnik folgende Zellen quantifiziert: CD4‐positive‐T‐Helferlymphozyten, Monozyten/Makrophagen und alle Zellen mit dem “Common Leukocyte Antigen” (CD45), also die Gesamtheit aller weißen Blutzellen im Sperma. Das Vorliegen einer Entzündung im Bereich des männlichen Genitaltraktes wurde durch enzymimmunologische Messung der Granulozytenelastase im Seminalplasma bestimmt. Bei 17 Männern lagen Spiegel über 1000 ng/ml vor, ein starker Hinweis auf einen entzündlichen Vorgang. Im Vergleich zu Männern mit niedrigen Granulozytenelastasespiegeln (1000 ng/ml) signifikant erhöhte Zahlen an Gesamtleukozyten, CD4+‐Lymphozyten und Monozyten/Makrophagen im Sperma (P<0,001); die Medianwerte in der Entzündungsgruppe waren 38fach erhöht für Gesamtleukozyten, 19fach erhöht für Monozyten/Makrophagen und 6fach erhöht für CD4+‐Lymphozyten. Unsere Ergebnisse zeigen, daß beim Vorliegen entzündlicher Veränderungen im männlichen Genitaltrakt die Zahlen potentieller HIV‐Wirtszellen im Sperma stark erhöht sind. Summary:Low dose of gossypol (5 μM) completely inhibited uptake of 2‐deoxy‐D‐glucose by rat spermatids. With gossypol 2‐deoxy‐D‐glucose transport into spermatids increased curvelinearly for 10 minutes after the start of incubation, but reached a plateau afterwards, whereas without gossypol it continuously increased. The levels of fructose‐1,6‐bis‐phosphate (FBP) and ATP were inversely correlated when the cells were exposed to glucose. However, in the presence of gossypol the levels of ATP and FBP decreased simultaneously. In addition, gossypol at 10 μM stimulated oxygen consumption when lactate was present. These results suggest that gossypol probably inhibits glucose transport into spermatids by lowering the ATP level by uncoupling respiratory chain‐linked phosphorylation.Zusammenfassung:Geringe Dosen von Gossypol (5 μM) hemmen vollständig die Aufnahme von 2‐deoxy‐D‐Glykose durch Rattenspermatiden. Mit Gossypol steigt der 2‐deoxy‐D‐Glykose‐Transport in die Spermatiden für 10 Minuten nach Inkubationsbeginn krummlinig; danach erreicht er ein Plateau, während ohne Gossypol ein kontinuierlicher Anstieg erfolgt. Die Werte für Fruktose‐1,6‐bis‐Phosphat (FBP) und ATP waren umgekehrt korreliert, wenn die Zellen Glukose ausgesetzt waren. In der Gegenwart von Gossypol fielen die Werte für ATP und FBP gleichzeitig ab. Zusätzlich stimulierte Gossypol bei 10 μM den Sauerstoffverbrauch, wenn Laktat vorhanden war. Diese Ergebnisse legen es nahe, daß Gossypol wahrscheinlich den Glukosetransport in die Spermatiden hemmt mittels Herabsetzung des ATP‐Werte
