SummaryA substance, proteinaceous in nature, with an anti‐agglutinic action on clumped sperm has been extracted from human semen. It appears to exist in two forms: an active reduced form and an inactive oxidised form.The protein was extracted from oxidised seminal plasma by salting with saturated ammonium sulphate. The extract was purified by preferential adsorption on a calcium carbonate column, the eluate being collected as fractions. The antagglutinin gave an extinction curve, as determined by ultraviolet spectrophotometry, similar to that as reported for the bull.Extraction recoveries, from semen stored frozen at different temperatures, indicate that the substance is possibly enzyme labile.An effect of the antagglutinin on clumped human sperm is illustrated.Die Isolierung eines möglichen Antagglutinins aus dem menschlichen SpermaZusammenfassungAus dem menschlichen Sperma wurde eine Proteinsubstanz mit einer antagglutinierenden Wirkung auf verklumpte Spermatozoen extrahiert. Diese Substanz liegt offenbar in zwei Formen vor: eine aktive, reduzierte Form und eine inaktive, oxydierte Form.Das Protein wurde aus oxydiertem Spermaplasma durch Aussalzung mit gesättigten Ammoniumsulfat extrahiert. Der Extrakt wurde durch Vorzugsadsorption an Calciumkarbonatsäule gereinigt, das Eluat wurde in Fraktionen gesammelt. SummarySteroidal and gonadotropic secretion were studied in infertile men with varicocele. Hormonal values were compared with those from control groups. No significant difference was found beteen control and pathologic groups in relation to prolactin secretion. FSH and LH showed increased basal values in 16% and 8% these cases in the group studies. Exaggerated gonadotropic response to LH‐RH was observed in 33.3% of the patients. Basal testosterone and estradiol levels were found to be reduced in 18% and 30% of the cases, respectively. Testosterone response to LH‐RH was also depressed in men with varicocele. Significant correlations were still observed between FSH and LH, LH and testosterone and testosterone and estradiol in infertile men with varicocele. These findings suggest both a tubular and interstitial involvement in varicocele and support the hypothesis of a Leydig cell deficiency in this pathology.Endokrine Bewertung bei unfruchtbaren Männern mit VarikoceleZusammenfassungBei unfruchtbaren Männern mit einer Varikocele wurde die Steroid‐ und Gonadotropin‐Sekretion untersucht; die Hormonwerte wurden mit denen von Kontrollgruppen verglichen. Es konnte keine signifikante Differenz zwischen den Kontrollen und der Varikocelen‐Gruppe hinsichtlich der Prolaktin‐Sekretion festgestellt werden. Für FSH und LH ergaben sich erhöhte Basalwerte in 16% bzw. 8%. Eine übersteigerte Gonadotropin‐Reaktion auf LH‐RH konnte in 33.3% der Patienten beobachtet werden. Für Testosteron und Oestradiol wurden die Basalwerte in 18% bzw. 30% herabgesetzt festgestellt. Signifikante Beziehungen ergaben sich zwischen FSH und LH, LH und Testosteron sowie zwischen Testosteron und Oestradiol. Daraus geht nach Auffassung der Autoren eine tubuläre und interstitielle Beteiligung bei der Varikocele hervor; hierdurch wird die Hypothese einer Leydigzellinsuffizienz bei diesem Kran

SummaryTo elucidate the mechanism of clomiphene action in the treatment of male infertility, we studied the effect of three‐month clomiphene treatment (50 mg daily) on the radio‐immunoassayable concentrations of gonadotropins (FSH, LH), prolactin (PRL), estradiol (E2), and testosterone (T) in seminal plasma and peripheral serum in eleven normogonado‐tropic subfertile men. Clomiphene therapy increased (p<0.01) the levels of FSH and LH in seminal plasma, but to a lesser extent (p<0.01) than in peripheral serum. PRL did not change in seminal plasma or peripheral serum. The seminal plasma and peripheral serum T concentrations doubled following clomiphene intake without any demonstrable correlation with the sperm count. The concentrations of E2increased about ten times (p<0.005) in seminal plasma, a significantly greater (p<0.05) increase than that in the serum. There was a significant correlation (r = 0.804, p<0.01) between the sperm counts and seminal plasma E2levels following clomiphene treatment. These results suggest that Sertoli cell activation may be a key event in clomiphene action in the treatment of male infertility.Gonadotropine, Prolaktin, Testosteron und Östradiol im Seminalplasma: Therapie‐Effekt von Clomifen (Dyneri®)ZusammenfassungUm den Mechanismus der Clomiphen‐Wirkung in der Behandlung männlicher Subfertilität zu erklären, haben wir den Effekt von dreimonatlicher Clomiphen‐Therapie (50 mg täglich) auf die radioimmunologischen Konzentrationen von den Gonadotropinen (FSH, LH), Prolaktin (PRL), Östradiol (E2) und Testosteron (T) im Seminalplasma und in peripherem Serum bei elf normogonadotropen subfertilen Männern untersucht. Die Clomiphen‐Behandlung erhöhte (p<0.01) das FSH‐ und LH‐Niveau im Seminalplasma, aber in geringerem Maße (p<0.01) als in peripherem Serum. Das PRL‐Niveau blieb unverändert sowohl im Seminalplasma als auch im peripheren Serum. Die Konzentration von T im Seminalplasma und im Serum verdoppelte sich während der Clomiphen‐Gabe ohne irgendeine demonstrierbare Korrelation mit der Spermatozoenzahl. Die Konzentration von E2stieg um etwa zehnmal (p<0.005) im Seminalplasma, ein signifikant höherer (p<0.05) Anstieg als im Serum. Eine signifikante Korrelation (r = 0.804, p<0.01) zwischen Spermatozoenzahl und E2im Seminalplasma während der Clomiphen‐Therapie wurde gefunden. SummaryIn 17 cases of male sterility with so‐called “mixed atrophy” in testicular biopsies, the germinal epithelium was investigated by quantitative as well as by ultrastructural means. In 13 cases, the number of spermatids only was reduced, in 2 cases earlier stages of germinal cells were diminished also. Normal quantitative data were found in biopsies of 2 patients.Among ultrastructural alterations various disturbances of the acrosome were dominant. Additional ultrastructural findings in spermatids included binucleate forms and lack of mitochondrial sheaths in the middle‐piece. The largely uniform changes in the tunica propria (fibrosis of the innermost layers, extensive changes of the basement membrane) were considered to be at least one of the cofactors responsible for the alterations of the germinal epithelium.Quantitative und ultrastrukturelle Untersuchungen des Keimepithels bei sogenannter „gemischter” oder „bunter” testikulärer Atrophie in HodenbiopsienZusammenfassungIn Hodenbiopsien von 17 Patienten wurde im Rahmen der Sterilitätsabklärung das histologische Bild einer sogenannten „bunten” oder „gemischten” Atrophie gefunden. Das Keimepithel wurde darauf speziell quantitativ und ultrastrukturell untersucht. In 13 Fällen war nur die Zahl der Spermatiden vermindert, in zwei Fällen betraf diese Reduktion auch frühere Stufen der Spermatogenese. Bei zwei Patienten war das Keimepithel quantitativ normal. Ultrastrukturell wurden vor allem Mißbildungen der Akrosomenstrukturen der Spermatiden gefunden, ferner doppelkernige Spermatiden und gelegentlich ein Fehlen der Mitochondrienscheide der Mittelstücke. Die weitgehend gleichartigen Alterationen der Tunica propria wie Fibrose der innersten Bindegewebsschicht und ausgedehnte Veränderungen der Basalmembran lassen vermuten, daß derartige Veränderungen der Tunica propria zumindest als einer der für die Keimepithelveränderungen verantwortlich

SummaryA histological study of 23 testicular biopsies or surgical specimens from adult cryptorchid testes revealed the presence of multi‐vacuolated Leydig cells in 12 cases. These cells showed abundant lipid inclusions and scarce secondary lysosomes as well as lipofuchsin pigment granules. Some of these cells contained crystals of Reinke but others had only microcristal‐line inclusions. SummaryA microbiological method, using the carnitine‐requiring yeast,Torulopsis bovinaATCC 26014, was developed to identify samples of human semen which contained low levels (<250 μM) of total carnitine. Of 399 semen samples from a male infertility clinic which were tested, 30 (7.5%) were low in carnitine. Of these, 14 were azoospermic and 16 were severely oligozoospermic. Some azoospermic samples (19 = 58%) and severely oligozoospermic samples (51 = 79%) did not give evidence of low carnitine concentrations. These results indicate that decreased total carnitine concentration in semen occurs in certain classes of azoospermic and severely oligozoospermic patients.Der Carnitingehalt des menschlichen Spermas bei Bestimmung mittels einer semiquantitativen auxanographischen Methode zur Ermittlung niedriger Carnitin‐KonzentrationenZusammenfassungEine mikrobiologische Methode wurde zur Identifizierung von Spermaproben mit einem Carnitingehalt unter 250 μM unter Verwendung der Carnitin‐verbrauchenden HefeTorulopsis bovinaATCC 26014 entwickelt. ZusammenfassungDie in den Neutralfetten und Phospholipiden gebundenen Fettsäuren des Humanspermas bei der Astheno‐, Azoo‐, Oligo‐ und Teratozoospermie wurden gaschromatographisch analysiert.
