SummaryScrotal sonography with a 7.5 MHz sector scanner was performed on 658 consecutive patients of our infertility clinic. The incidence of pathological findings was unexpectedly high. Forty per cent of the patients revealed pathological structures such as varicoceles (21%), hydroceles (7%), epididymal abnormalities (6%), spermatoceles (6%), intratesticular hyper‐ and hypoechoic changes (4.5%), intratesticular cysts (1%) and tumours or carcinomain situ(CIS) (0.6%).Sonographic evaluation and measurement of the caput epididymidis was compared with palpation. Sonography distinguished size ranges of “normal” and “thickened” epididymides as diagnosed by palpation. Cystic structures were proven in 56% of cases with “thickened” epididymides. The sonographically determined diameters of doppler‐negative blood vessels were significantly smaller than those of doppler‐positive vessels. Sonography revealed a higher occurrence of varicoceles than diagnosed by palpation (76% by palpation). Only 58% of sonographically identified hydroceles and only 67% of sonographically detected spermatoceles were detected by palpation. One testicular tumour and one case with CIS were only seen by sonography and not suspected on palpation.The results demonstrate that sonography represents a valuable tool in the routine diagnosis of andrological patients.ZusammenfassungBei 658 konsekutiven Patienten unserer Fertilitätssprechstunde wurde routinemäßig eine sonographische Untersuchung der Skrotalorgane mit einem 7, 5 MHz Sektor‐Schallkopf durchgeführt. Die Inzidenz pathologischer Befunde war überraschend hoch. Bei 40% der Patienten fanden sich pathologische Strukturen, wie Varikozelen (21%), Hydrozelen (7%), Anomalien des Nebenhodens (6%), Spermatozelen (6%), intratestikuläre hyper‐ und hypoechogene Veränderungen (4,5%), intratestikuläre Zysten (1%) und Tumoren oder Carcinoma‐in situ[CIS] (0,6%).Die sonographische Untersuchung und Vermessung des Nebenhodenkopfes wurde mit dem Palpationsbefund verglichen. Bei der Palpation wurde zwischen “normal großem” und “verdicktem” Nebenhoden unterschieden. Zystische Strukturen wurden in 56% der “verdickt” palpierten Nebenhoden nachgewiesen. Der sonographisch ermittelte Durchmesser von Doppler‐negativen Gefáßen war signifikant kleiner als der von Dopplerpositiven Gefäßen.Im Vergleich zur klinischen Untersuchung fand sich sonographisch eine höhere Inzidenz von Varikozelen (76% palpiert), von Hydrozelen (58% palpiert) und von Spermatozelen (67% palpiert). SummaryMorphological comparison of sperm in raw ejaculates and swim‐ups showed that the swim‐up process does not simply increase the proportion of ‘normal’ sperm. ZusammenfassungEs wurden im Rahmen dieser Studie 21 Werktätige, die gegenüber Pflanzenschutzmitteln (organische Quecksilberverbindungen) und Schädlingsbekämpfungsmitteln (halogenierte Kohlenwasserstoffe) exponiert waren, hinsichtlich ihrer Fertilität untersucht. Wenn die Konzentrationen der organischen Quecksilberverbindungen in der Luft am Arbeitsplatz ansteigen, sind auch die Spiegel dieses Schwermetalls im Urin und im Ejakulat erhöht und die Fertilität ist reduziert. Es konnte festgestellt werden, daß eine Korrelation zwischen der Konzentration der Schädlingsbekämpfungsmittel am Arbeitsplatz und der männlichen Fertilität besteht, besonders in den Fällen mit einer Überschreitung der zulässigen maximalen Arbeitsplatzkonzentration.SummaryTwenty‐one workmen exposed to herbicide as organic mercury compounds and to pesticide as halogenic hydrocarbon has been investigated concerning their fertility. If the concentrations of organic mercury compounds increase in the air of the place of employment then the levels of this heavy metal are also higher in the urine and in the ejaculate, and the fertility is reduced. SummaryEffects of gossypol on human spermatozoal enzymes were investigated. This compound was found to be a potent inhibitor of the NAD‐linked enzymes, glucelaldehy de‐3‐phosphate dehydrogenase (GA3PDH), lactate dehydrogenase (LDH), malate dehydrogenase (MDH), and isocitrate dehydrogenase (ICDH). MDH was inhibited by gossypol when the reaction was carried out in malate‐oxalacetate (direct) or oxalacetate‐malate (reverse) directions. The I50of gossypol for the direct reaction was 2.9μM, whereas that for the reverse reaction was 1.2 μM. Reciprocal plots due to Lineweaver‐Burk showed that MDH is inhibited in a noncompetitive manner with respect to both reactions. LDH was also inhibited by this compound when pyruvate or α‐ketobutyrate was used as a substrate. Gossypol was a noncompetitive inhibitor for LDH. The I50of gossypol for LDH were 9.8 μM and 11.3 μM, when using pyruvate and α‐ketobutyrate, respectively as substrates. The I50of gossypol for GA3PDH and ICDH were 110 μM and 2.7 μM, respectively.ZusammenfassungEs wurde der Einfluß von Gossypol auf die Enzyme der menschlichen Spermatozoen untersucht. Diese Substanz ist ein potenter Hemmer der NAD‐gebundenen Enzyme Gluzelaldehyd‐3‐phosphatdehydrogenase (GA3PDH) Lactathydrogenase (LDH), Malatdehydrogenase (MDH), und Isozitrathydrogenase (ICDH). MDH wurde durch Gossypol gehemmt, wenn die Reaktion in Malat‐Oxalazetat (direkt) oder in Oxalazetat‐Malat (umgekehrt) durchgeführt wurde. Der I50von Gossypol für die direkte Reaktion war 2.9μM, wohingegen der Wert für die umgekehrte Reaktion 1.2μM betrug. Reziproke Plots (Lineweaver‐Burk) zeigten, daß MDH in einer nichtkompetitiven Weise in Bezug auf beide Reaktionen gehemmt wird. SummaryWe have studied the ultrastructure of testes biopsies and immotile spermatozoa from a supposed secondary infertile male. A wide range of tail defects has been described in testes and semen. Among these, tail stump spermatozoa occurs rarely. Although the origin of this defect is unknown, testes sections revealed that damage occurs during spermiogenesis at the latest stages during flagellum elongation in spermatids.ZusammenfassungWir haben die Ultrastruktur der Hodenbiopsien und der unbeweglichen Spermatozoen von einem Mann studiert, der vermutlich eine sekundäre Unfruchtbarkeit präsentiert. Mehrere Arten von Anomalien des Schwanzes in dem Hoden und in der Samenflüssigkeit werden beschrieben. Dennoch sind Spermatozoen mit einem Stummelschwanz seiten vorhanden. SummaryMonoclonal antibodies specific for acrosomal proteins were prepared and characterized. Two of these monoclonal antibodies, ACR.3 and ACR.11, reacted with the same molecular specificity of acrosomal antigens (18–20 kDa), antibody ACR.4 reacted with the 25–27 kDa antigens, and antibody ACR.2 reacted with several forms of boar acrosin (55, 53, 45, and 38 kDa). All monoclonal antibodies labelled the acrosomes of undamaged spermatozoa in immunofluorescence test. This test should be convenient as an immunlogical test of the sperm quality.ZusammenfassungDie für akrosomale Proteine spezifischen monoklonalen Antikörper wurden vorbereitet und charakterisiert.
