SummaryAfter separation of three epididymal acid phosphatases their biochemical properties were differently studied. With appropriate substrate and inhibitor selection the distribution of the enzymes in different segments as well as the subcellular fractions of the rat epididymis was also demonstrated. The same biochemical differences were also utilized in the histochemical localization of the enzymes.It was found that Enzyme I had a pH‐optimum at 5.0, a molecular weight of 97 000 and Km‐constant of 0.901 mM. It was highly sensitive to tartrate and fluoride and it was localized in lysosomes as well as in the epididymal spermatozoa.Enzyme II had an optimum at pH 5.7, a molecular weight of 67 000 and Km‐constant of 0.806 mM. It was also inhibited by fluoride but more resistant to tartrate. Its subcellular site was also particulate, but it was also found in the epididymal fluid.Enzyme III had an optimum at pH 5.2, a molecular weight of 135 000 and Km‐constant of 0.685 mM. It was resistant to low concentrations of fluoride and tartrate but sensitive to heavy metal ions. The enzyme was soluble and it behaved incoherently in thermal inactivation. All enzymes revealed the highest activity in the thin middle segments of the epididymis.Histochemically naphthol substrates gave a diffuse reaction in the epididymal epithelial cells. With the lead salt methods glycerophosphates and p‐nitrophenylphosphate gave somewhat different results depending on their specificity as substrates for the epididymal enzymes. Both substrates gave a strong reaction supranuclearly in the Golgi area of the chief cells. This activity was inhibited by tartrate and was most probably due to Enzyme I. The epididymal corpus and cauda showed additionally a very strong apical activity in the chief cells with p‐nitrophenylphosphate. This activity was resistant to tartrate but sensitive to fluoride. It was concluded that this enzyme represents Enzyme II activity. Similar activity was also found in the dissolving “holocrine” cells of the corpus and the cauda. The activity of the soluble Enzyme III could not be revealed with the present methods and the spermatozoa in the tubular lumina remained unstained.ZusammenfassungNach Trennung der drei sauren Phosphatasen des Nebenhodens wurden ihre bioche‐mischen Eigenschaften untersucht.Enzym I hat ein pH‐Optimum bei 5.0, ein Mol. Nebenhoden‐Korpus und ‐Cauda zeigten zusätzlich eine starke apikale Aktivität in den Hauptzellen mit p‐Nitro‐phenylphosphat. Diese Aktivität war resistent gegenüber Tartrat aber sensibel gegen Fluorid. Die Autoren ziehen die Schlußfolgerung, daß dieses Enzym die Aktivität von Enzym II darstellt. SummaryThe metabolism of nine radioactively labeled steroids in the epithelium of the seminal vesicle of the mature guinea pig has been studied. The rapid assimilation and metabolism of these steroids demonstrate the very active biochemical nature of this tissue. Based on the use of several thin‐layer chromatography systems and comparison with the locations of known standards, the following have been disclosed. Testosterone was rapidly converted to dihydrotestosterone and androstanediol. The latter was the major metabolite of dihydrotestosterone and of androsterone. Androstenedione was readily converted to androstanedione, testosterone, and dihydrotestosterone, although it formed little androstanediol. Dehydroepiandrosterone was converted to small amounts of androstenediol and androstanediol. Pregnenolone was rapidly converted to an unidentified highly polar compound only. Progesterone was converted to 5α‐pregnane‐3β‐ol‐20‐one and its 3α‐isomer. Dehydroepiandrosterone and progesterone were also significantly converted to unidentified highly polar compounds.The major metabolites of 17‐hydroxyprogesterone co‐chromatographed with standard androstanediol, testosterone, and an unidentified metabolite possessing intermediate chromatographic mobility. In addition, 17‐hydroxyprogesterone was converted to small amounts of compounds possessing identical Rfvalues as standard androstenedione and dihydrotestosterone. The identification of the products of 17‐hydroxyprogesterone metabolism and their physiologic significance must await critical evaluation.Because of its homogeneity, isolated epithelium of guinea pig seminal vesicle shows promise as a tissue preparation for use in future studies that might elucidate the role(s) of individual androgens in secretory tissues of the male accessory sex organs. Our demonstration of extensive steroid interconversions in this tissue is a logical prerequisite to such studies.ZusammenfassungIn der vorliegenden Studie wird über Untersuchungen mit neun radioaktiv markier‐ten Steroiden und ihren Stoffwechsel im Epithel der Samenblase des reifen Meerschwein‐chens berichtet. Die schnelle Assimilation der Stoffwechsel dieser Steroide zeigt, wie aktiv dieses Gewebe ist.Testosteron wurde sehr schnell zu DHT und Androstendiol konvertiert. Letzteres war der größere Metabolit von DHT und von Androsteron. Androstendion wurde gerne zu Androstendion, Testosteron und DHT konvertiert, obwohl es wenig Androstandiol bildete. DHEA wurde konvertiert zu kleinen Mengen von Androstendiol und Androstandiol. Pregnenolon wurde schnell konvertiert zu einer nicht identifizierten hochpo‐laren Substanz. Progesteron wurde konvertiert zu 5α‐pregnan‐3β‐ol‐20‐on und seinem 3α‐Isomer. DHEA und Progesteron wurden signifikant konvertiert zu nicht identifizierten hochpolaren Substanzen.17‐Hydroxyprogesteron wurde konvertiert zu niedrigen Mengen von Substanzen, die identische Rf‐Werite mit den Standard für Androstendion und DHT besaßen. Die Identifizierung dieser Substanzen des 17‐Hydroxyprogesteronstoffwechsels und ihrer physiologischen Bedeutung muß einer kritischen Auswertung überlassen bleiben. SummarySamples of human semen frozen in liquid nitrogen (‐196°C) with no glycerol, 5 and 10 % glycerol were compared with samples that were untreated, treated with 10 % glycerol but not frozen, and spermatozoa frozen at ‐20°C.SEM and TEM of the samples indicates that 10% glycerol caused fewer surface changes of the spermatozoa than other treatments. Motility counts after the various freezing treatments were also highest when 10% glycerol was used as the cryoprotectant. Nonetheless, cryopreservation is detrimental to spermatozoa and often causes considerable damage to the acrosome with a leakage of the acrosomal contents.ZusammenfassungEs wurden Proben von menschlichem Sperma miteinander verglichen, die in flüssigem Stickstoff (‐196°C) mit 5 und 10% Glycerin und ohne Glycerin eingefroren waren und solchen Spermaproben, die unbehandelt blieben bzw. mit 10% Glycerin versetzt waren ohne eingefroren zu werden und Proben, die auf ‐20°C eingefroren wurden. Die Ergebnisse zeigen deutlich, daß ein Zusatz von 10% Glycerin in geringerem Umfange morphologische Veränderungen an der Oberflächenstruktur der Spermatozoen verur‐sacht als es andere Behandlungsmaßnahmen tun. SummaryIn two patients exhibiting eunuchoid features in association with normal sized testes and complete spermatogenesis concommittant with only occasional Leydig cells between the tubuli, (proven by testicular biopsy) an attempt was made to elucidate the factors leading to this condition. Both patients responded with significant rise in both plasma FSH and LH after administration of synthetic GnRH indicating pituitary responsiveness. However, no rise in either FSH or LH could be observed after administration of clomiphene citrate during three weeks of treatment indicating hypothalamic unresponsiveness to chemical stimuli. Although plasma testosterone levels rose significantly after administration of Human Chorionic Gonadotropin, estradiol remained unchanged during three weeks of HCG administration. A hypothesis is discussed which defines this syndrome in these two cases as primary Leydig cell failure, expressed in the inability of these to transform testosterone into estrogens, thus depriving pituitary and hypothalamus from a proper steroidal milieu necessary to adequate functioning.ZusammenfassungBei zwei Patienten, bei denen die Diagnose “Fertile Eunuchen” auf Grund des typischen eunuchoiden Erscheinungsbildes in Verbindung mit normalgroßen Hoden und kompletter Spermatogenese gestellt worden war, wurden die pathophysiologischen Hintergründe untersucht. Beide Patienten reagierten mit einem signifikanten Anstieg der FSH und LH‐Werte nachdem synthetisches GnRH verabreicht worden war. Eine hypophysäre Dysfunktion konnte somit ausgeschlossen werden. Nachdem während drei Wochen täglicher Clomiphenzitratgabe kein Anstieg der FSH‐ und LH‐Werte zu verzeichnen war, schlossen wir auf eine Nichtansprechbarkeit des Hypothalamus auf chemische Stimuli. Die Funktionsfähigkeit der Leydig‐Zellen wurde mit Hilfe des HCG‐Tests untersucht. Es zeigte sich, daß Testosteron zwar normal ansprach, daß aber auch nach fortgesetzter HCG‐Gabe kein Anstieg der Oestradiol‐Werte zu verzeichnen war. Wir stellen eine Hypothese zur Diskussion, in der das Syndrom des “Fertilen Eunuchen”, zumindest in den von uns beschriebenen Fällen, als eine primäre Leydigzell‐Schädigung verstanden wird, die sich darin ausdrückt, daß die Leydigzellen nicht imstande sind, Testosteron zu Oestrogenen zu aromatisieren, oder Oestrogene zu sezernieren. Hierdurch würde das notwendige steroidale Milieu, welches für die normale Funktion von Hypothalamus und Hypophyse unerläßlich ist, gestört. Eine isoliert

SummaryTridacnin M, a galactosyl‐specific reagent prepared from the bivalve clam Tridacna maxima (Röding) was used for the demonstration of 2 different glycosubstances with terminal galactosido units in human semen. The results obtained by immunodiffusion tests indicate that the seminal plasma contains a water‐soluble glycoprotein with two carbohydrate chains, one of them having a terminal ß‐galactosyl group whereas the other one seems to have a terminal N‐acetyl‐neuraminic acid group and a subterminal ß‐galactosyl group. This glycoprotein is a secretion product of the seminal vesicles and, therefore, of diagnostic significance: it is absent in cases of bilateral occlusion of the ampullae. Another glycosubstance with terminal galactosido‐residues could be demonstrated on the surface of sperm cells by agglutination reactions. This glycosubstance is an integral part of the spermatozoan membrane because it cannot be removed by repeated washings.ZusammenfassungUnter Verwendung von Tridacnin M, einer galactosyl‐spezifischen Substanz aus der Muschel Tridacna maxima (Röding) konnten im menschlichen Samen mit Immunodiffusions‐ und Agglutinationsreaktionen 2 verschiedene Glykosubstanzen mit end‐ständigen Galaktosylgruppen nachgewiesen werden. Unsere Ergebnisse weisen darauf hin, daß das Spermaplasma ein wasserlösliches Glycoprotein mit 2 Kohlenhydratketten enthält, darunter eine mit endständiger ß‐Galactosylgruppe, während die andere eine terminale N‐acetyl‐Neuraminsäuregruppe und eine subterminale ß‐Galactosylgruppe besitzt. Dieses Glycoprotein ist ein Sekretionsprodukt der Samenbläschen und somit von diagnostischer Bedeutung: bei doppelseitigem Verschluß der ductus ejaculatorii fehlt es im Ejakulat. Eine zweite Glykosubstanz mit endständigen Galactosylgruppen konnte an der Oberfläche der Spermatozoen nachgewiesen werden. SummaryDiagnosis of oligospermiogenesis was made in 76 infertile subjects submitted to testicular biopsy. Histological examinations demonstrated a reduction in mature spermatids (Sc. Sd.‐Clermont classification) in the tubular sections. Comparison of histological data with findings from seminal fluid analyses in these subjects revealed: a) a pathological seminal fluid pattern in all cases of oligospermiogenesis, b) these patients may show either an azoospermic or oligozoospermic seminal fluid pattern; statistical analyses confirmed an almost equal distribution of the two conditions.ZusammenfassungBei 76 andrologischen Patienten wurden die Diagnose einer Oligospermatogenesis mittels der Hodenbiopsie gestellt. Die histologische Untersuchung ergab eine Reduzierung der reifen Spermatiden in den Tubuli seminiferi.
