Abstract—The amount of chloroform‐methanol‐soluble protein obtained from rat brain tissue homogenates which have been subjected to washing by repeated contrifugation or dialysis is a several‐fold greater than that obtained from untreated homogenates. The increase consequence of the removal of loosely bound electrolytes during the process of centrifugation or dialysis: little or no increase is observed upon (a) the addition of the supernatant, the ashed supernatant, or the diffusate to the washed homogenate; (b) the addition of inorganic salts to the washed homogenate; or (c) washing of the homogenate with a salt solution.The previously observed effect of sucrose in increasing the amount of chloroform–methanol‐soluble protein obtained from subcellular fractions is apparently superimposed upon the effect of the remov

Abstract—The occurrencc of water soluble lipoprotein in the 105,000gsupernatant of rat brain homogenate has been demonstrated.The lipoproteins of brain differed from those of the serum with respect to lipid composition, density and reaction to serum lipoprotein antibodies.Acrylamide gel electrophoresis resolved the brain supernatant into four zones which stained with lipid dye. It was possible to demonstrate that these zones also contained protein by double staining of the gel for lipid and protein. Two of the zones differed in their mobility from serum lipoprotein.Preparative acrylamide gel electrophoresis isolated protein fractions in supematant which contained both lipid and protein and permitted preliminary identification of the lipoprotein bands amongst all the protein bands resolved by acrylamide gel electrophoresi

Abstract—Optic lobes of the brains ofOctopus dofleiniwerehomogenized hasaline–sucrose medium and subjected to differential and density gradient centrifugation. The fractions and subfractions thus obtained were analysed for acetylcholine (ACh) (bioassay) and protein content and were subjected to electronmicroscopy. Bound ACh was associated with particulate fractions, and a large portion of it could be recovered in subfractions that contained predominantly nerve endings.Expressed in terms of amount of ACh per mg protein, the ACh content of the nerve ending fractions was nearly 100 times greater than that of corresponding fractions previously obtained by others from mammalian brain. Calculations show that this was the minimal amount of ACh to be expected if the isolated nerve endings were predominantly cholinergic.Octopusbrain tissue is in therefore very promising for future studies on ACh metabolism and compartmentation cholinergic nerve endi

Abstract—Studies were made of the effects of fluphenazine, chlorpromazine and triflupromazine on tissue concentration, liver synthesis of ascorbic acid and its distribution in different areas of the brain. All the three drugs were found to increase liver concentration and synthesis of the vitamin at 24 hr after administration of a single oral dose of the vitamin, but only fluphenazine was found to increase its concentration in the adrenals and brain; the increase in the latter case was found to vary in different regions of the brain, the olfactory lobes, hypothalamus and residual brain showing maximum increases, andthe basal ganglia, visual cortex and remaining dorsal cortex showing minimum increases. Abstract—Two uridine nucleotides, UDPG and UDPGA, have been measured in brain by a procedure whereby they are extracted from tissue with subsequent utilization, either directly with (UDPGA) or after dehydrogenation (UDPG), in a glucuronidation reaction with harmol or harmalol as cosubstrate. The concentrations of UDPG and UDPGA in brains of a few species of animals examined are, respectively, 11.5–23.3 μmoles/100 g and 0.7–2.5 μmoles/100 g tissue. Abstract—The ability of subfractions of disrupted synaptosomes to bind tetanus toxin has been studied. Synaptic vesicles had little toxin‐fixing capacity, but a fraction believed on morphological appearance and enzymic composition to be rich in external synaptosome membranes had a toxin‐fixing capacity about 10 times that of the synaptic vesicles and over twice that of the initial brain homogenate. Abstract—Dogs were given repeated doses of pentylenetetrazol to maintain a condition of status epilepticus for periods of 30–40 min. Analyses of cerebral tissue frozenin situshowed significant increases in alanine, arginine, γ‐aminobutyric acid, glycine, histidine, leucine, lysine, phenylalanine, serine, tyrosine, and valine. Decreases occurred in the glutamic and aspartic acid levels. Other measured amino compounds were unchanged. Ammonia was increased, but not more than occurs in seizures of brief duration. A large decrease was noted in the ganglioside fraction, and a decrease also in a fraction containing the lecithins and sphingomyelins. The cephalin, cerebroside–sulphatide, and cholesterol fractions were not affected.Similar repetitive seizures induced by bemegride and continuing for shorter periods (5–9 min) brought about less extensive changes. Glutamic acid was increased, in contrast to the pentylenetetrazol effect, and the apparent decrease in aspartic acid was not statistically significant.
