Summary.Serology, isoelectric focusing (IEF) of expressed antigens, and restriction fragment length polymorphisms (RFLP) were compared for the identification of BoLA class I haplotypes. Expressed antigens identified as bands by IEF correlated well with serological definition confirming and extending our earlier findings (Joostenet al.1988). Comparison of serology and isoelectric focusing bands with restriction fragments was more complicated; fragments were found which correlated both with broadly reacting and antigen specific sera. We also found correlation of fragments with two or more sera which showed no cross‐reactivity. Fragments unique to particular haplotypes were also observed.Serology remains the simplest method of typing BoLA class I antigens. Isoelectric focusing generally agrees with serological definition of antigens and detects antigens not yet defined by serology. It may also be useful in defining the products of other expressed BoLA class I loci. In order to identify RFLPs which could be used for typing, comparison with serology or IEF is essential. Summary.Two‐dimensional agarose gel (pH 8.6)‐horizontal polyacrylamide gel (pH 9.0) electrophoresis of horse serum samples, followed by general protein staining, revealed genetic polymorphism of an unidentified protein tentatively designated serum protein 3 (SP3). The SP3 fractions appeared distinctly when a 14% concentration of acrylamide was used in the separation gels. The 2‐D mobilities of SP3 fractions were quite similar to that of albumin. Family data were consistent with the hypothesis that the observed SP3 phenotypes were controlled by four co‐dominant, autosomal alleles (D,F,I,S). Evidence was provided that theFallele can be further divided into two alleles (F1andF2); the mobilities of F1and F2variants were very similar. Each of theSP3alleles gave rise to one fraction and each of the heterozygous types showed two fractions. More than 600 horses representing five different breeds (Swedish Trotter, North‐Swedish Trotter, Thoroughbred, Arab and Polish Tarpan) were typed for SP3, and allele frequency estimates were calculated. Summary.Cutaneous malignant melanoma of Sinclair swine (SSCM) is an inherited neoplasm present at birth in a majority of affected animals. We have characterized the swine leucocyte antigen (SLA) complex of the Sinclair herd at Texas A&M University using a one‐way mixed lymphocyte test and found that one particular haplotype, arbitrarily identified as haplotypeB, is associated with the expression of SSCM. Only a single dose of theBhaplotype is required for a dominant allele at a ‘tumour initiator’ locus to be fully penetrant. In addition, swine homozygous for haplotypeBdevelop more primary tumours between birth and weaning than those heterozygous for theBhaplotype. Summary.By using immunoblotting with antiserum specific to human plasma apolipoprotein A‐IV (apoA‐IV), a previously reported polymorphic plasma protein of dogsvizpostalbumin‐2 (Pa2) and one of horsesvizserum protein 2 (SP2), were identified as apoA‐IV of these species. Summary.Forty AI bulls were tested for BoLA class I antigens by means of eight specific polyclonal reagents. By means of immobilization and sperm penetration tests these antigens were not detected on sperm cells. Isoimmunization studies with the use of sperm as antigenic stimuli and insemination of frozen spermatozoa diluted in specific reagents did not prove the presence of BoLA class I antigens on bovine spermatozoa. The cytotoxic tests used in this investigation were not reliable.

Summary.In all, 363 alloantireagents were tested in Berne, Switzerland and in Oslo, Norway against lymphocytes from 1679 goats of different breeds. The same lymphocytotoxicity test was used at both laboratories. The test data were pooled, and correlation coefficients for pairs of sera were used to group the sera in clusters. Twelve clusters were accepted as defining lymphocyte antigen specificities believed to be coded by genes within the major histocompatibility complex (MHC). The specificities defined by these clusters were designated Eu 1‐Eu12. Two clusters defined specificities which were not coded from loci within the MHC. Summary.Restriction fragment length polymorphism analyses of swine leucocyte antigen (SLA) class I genes were performed on 70 Duroc and 38 Hampshire boars from the 1986‐87 national performance tests of each breed in the USA. Few boars were inbred. Southern blotting and hybridization procedures were performed on genomic DNA, isolated from white blood cells, usingPvuII endonuclease and a swine major histocompatibility complex (MHC) class I probe. Durocs had an average of 11 restriction fragments, with the most common being in 63% of the boars and the least common appearing in only one boar. Hampshire boars had an average of 12 restriction fragments, with the most common appearing in 73% of the boars and the least common appearing in only one boar. Least squares procedures and stepwise regression methods were used to examine the association between DNA restriction fragments and the selection index (INDEX), average daily gain (ADG), average backfat thickness (BF), loin muscle area (LEA), and age at 104kg (DAY104). In the Duroc breed one DNA restriction fragment was associated with decreased INDEX (P<0.05) and decreased ADG (P<0.05) whereas two other fragments were associated with increased BF (P<0.05). In the Hampshire breed two restriction fragments were associated with an increase in INDEX (P<0.05). Cluster analyses were used to group pigs of each breed on the basis of similar RFLP patterns. One cluster group in the Duroc breed was associated with lower average INDEX values (P<0.05), greater average DAY104 (P<0.05), and a larger mean LEA (P<0.05). In the Hampshire breed one cluster group was associated with lower INDEX (P<0.05). These results suggest there may be an association between swine MHC class I genes and performance traits in swine. Summary.A study was carried out to attempt to explain the basis of the association between theEslocus and starting proportion in Swedish Trotters which had been observed previously. The effect ofEsgenotype on starting proportion has diminished among horses born in the late 1970s. There are indications that the incidence of leg lesions varies betweenEsgenotypes. If this is true, it is possible that the decreasing effect of theEslocus on starting proportion is due to the environmental changes which have been made at race tracks in the early 1980s to reduce the strain on the legs of the trotters.
