AbstractOn the basis of structure‐functional analysis of the development ofAcremonium chrysogenum, e.g. under conditions either stimulating antibiotic synthesis or not conductive to production, a scheme was proposed representing the various ways in which morphological differentiation occurs in the culture in dependence on the directions of its metabolism.Three types of culture differentiation were determined. Type 1 differentiation is characterized by the transition of the vegetative stage into the reproductive one with the formation of conidia. Type 2 differentiation is characterized by the formation of typical arthrospores also being the reproductive form. Type 3 differentiation is characterized by the multistage transformation of the mycelium organization into the yeast‐like one which is metabolically more active and is a producer of antibiotics and enzymes.In addition to the defined regularities in the development and differentiation ofAcremonium chrysogenumstructural peculiarities were observed which could be helpful to the search for regulators or specific enzymes taking part in the culture developm

摘要:
AbstractFour pure methylotrophic bacterial strains isolated from Kuwaiti soil were designated KISRI‐5, KISRI‐6.1, KISRI‐512 and KISRI‐5112, and deposited in NCIMB under NCIB numbers 12135, 12136, 12137 and 12138, respectively. Characterization tests showed that they grew well on methanol and slowly on glucose, at temperatures up to 44 °C and pH 7.0. However, they could not grow on numerous other organic carbon sources. The four strains were very close taxonomically, with minor differences in colony characteristics, lipids pattern, mole G + C% and resistance to certain antibiotics. All strains utilized monomethylamine, but not methane or trimethylamine; all contained ubiquinones with eight isoprene units (Q8), and had C16as their major fatty acid component. Their characteristics indicate that they are different strains of restricted type M facultative methylotrophs, and could be considered new members of the genusMethy

AbstractThe activities of fructose‐1,6‐bisphosphatase, malate dehydrogenase and PEP carboxykinase were tested during discontinuous growth of then‐alkane‐assimilating yeastCandida maltosaon glucose or ethanol.As expected, the highest activities of the enzymes were measured in the early log phase of growth on ethanol and the lowest in the early log phase of growth on glucose. However, the differences in the activities are much smaller than inSaccharomyces cerevisiaeand other yeasts under similar conditions. 摘要:
AbstractThe nucleic acid content inCandida utiliswas autolytically reduced only after previous lyophilization of yeast cells or pretreatment of the cells with ethyl acetate. These pretreatments provided a sufficient permeability of the cell membrane system and thus enabled a reduction of the nucleic acid content. While deep freezing of the cells (−35 °C) had no positive effect on the degradation of nucleic acids, lyophilization of yeast cells led to a 75% decrease of the nucleic acid content. Furthermore, a 55% reduction of the nucleic acid content was proceeded without a concomitant loss of proteins. Similar results were obtained by using ethyl aceta

AbstractCurrent models for the mechanism of SOS mutagenesis inE. colipropose the involvement of a new or modified DNA polymerase III holoenzyme in error‐prone replicative bypass of bulky DNA lesions assuming an inhibited or excluded 3′ → 5′ proofreading exonuclease function of DNA polymerase. By promotor fusion togalK, gene fusion tolacZand SI analysis thein vivoregulation ofdnaQcoding the proofreading subunit of DNA polymerase III holoenzyme was analyzed under conditions of induced or constitutive SOS expression. The results presented here clearly show that, at least on the level of gene expression no regulatory event seems to contribute to the assumed decrease of proofreading activity during SOS mediated error‐prone bypassing of bulky lesions.
