Brucella abortusstrain 19 vaccine became widely available in New Zealand in 1950. In 1966, vaccination was made compulsory and age restrictions were imposed. Surveys of the cattle population were carried out between 1966 and 1970 to establish the prevalence of brucellosis and to compare the performance of various diagnostic tests under New Zealand conditions. In 1969 and 1970 a voluntary test-and-slaughter scheme was introduced basedon the brucellosis card test as a screening test, the complement fixation test as the definitive test, and the milk ring test to screen accredited dairy herds. This scheme was extended to a compulsory national Brucellosis Eradication Scheme in September 1971, with the aim of bringing all eligible cattle under test by 1977.

The New Zealand Brucellosis Eradication Scheme was introduced in 1971, following five years of compulsory calfhood vaccination withBrucella abortusstrain 19. Each of the 25 veterinary administrative districts was divided into zones and the eradication effort was concentrated on one zone at a time, ensuring best use of manpower and resources and orderly expansion of the scheme to involve all eligible cattle by 1977. Every effort was made to inform each farmer of the requirements of the scheme and to scheduling the testing programme to fit the farmer's management system.

Automation of the complement fixation test for diagnosingBrucella abortusinfection in cattle has allowed this difficult, time-consuming and labour-intensive hut most specific test to he used for large scale serodiagnosis in New Zealand's Brucellosis Eradication Scheme. The automated test has the advantages of eliminating much human error, of improving accuracy and of reducing the costs of labour and reagents.

摘要:
The Auto-Analyzer is used to perform complement fixation testing for the Brucellosis Eradication Scheme. In this system, the reactions of the complement fixation test are carried out using the continuous flow analysis principle. A colorimeter measures the transmittance of red cells left in suspension at the end of the reactions and this is traced on to the Auto-Analyzer recorder chart.

All automated complement fixation test results and testing procedures carried out at the Central Brucellosis Laboratory for the Brucellosis Eradication Scheme undergo quality control checks. Auto-Analyzer performance is monitored by reagent concentrations, peak heights of the standard positive sera and percent transmission for 0% and 100% lysis of sheep red blood cells. Deviations in the size and form of sample peaks indicate problems with the serum sample or the testing system.

The mandatory reporting and investigation of bovine abortions provides a system for the surveillance of the progress of the Brucellosis Eradication Scheme and a warning of resurgence of brucellosis in tested herds. Before the scheme was introduced in 1971, about a quarter of all abortions were caused byBrucella abortus. By 1976, the percentage of abortions due toBrucellain herds under test for brucellosis had been reduced to 3.4%. MostBrucellaabortions occurred during the seventh and eighth months of gestation. Cows in the 4- to 6-year-old age group were at greater risk of aborting fromBrucellainfection than heifers. Approximately 94% of all aborting cows were in herds which experienced three or fewer abortions. Less than 4% of herds experienced four or more abortions annually. In most cases of abortion due to brucellosis, more than one diagnostic test yielded positive results. However, 13.3% of cases were recognised by positive bacteriological results alone.

The scale, complexity and short time-scale of the Brucellosis Eradication Scheme made the development of a comprehensive information system imperative. The Brucellosis Information System is derived from three data bases, a computerised record of test results obtained in the Central Brucellosis Laboratory and manually maintained records of problem-herd investigations and within-herd records of routine testing. Reports derived from manual records are presented in a standardised format. The computer record consists of codes representing the type of test performed, the date of test, the number of animals tested and the number of reactors.
