Investigation of a mild hemorrhagic tendency in a French family revealed the father to be heterozygous for hypoprothrombinemia while the mother was heterozygous for dysprothrombinemia. All possible genetic combinations could be demonstrated. Among the children the double heterozygosity encountered in 3 of them allowed us to discover an abnormal prothrombin, prothrombin Metz, which generates an abnormal thrombin less sensitive to inactivation by antithrombin III than normal thrombin.

A first level of pharmacological interference with platelet function is located at the site of the agonist-receptor interaction (receptors for collagen, adenosine diphosphate, serotonin, fibrinogen). A second level of interaction is to prevent the mobilization of intra-cellular calcium ions which can be obtained by inhibition of phosphodiesterase, activation of cAMP (e.g. with prostacyclin analogues), inhibition of thromboxane A2 formation or blocking of its receptors. Compounds preventing the platelet contractile process or platelet secretion could theoretically also prevent some platelet functions.

A patient with Glanzmann’s thrombasthenia has been analysed by high resolution two-dimensional polyacrylamide gel electrophoresis and crossed immunoelectrophoresis. Conflicting reports have been published concerning the spontaneous release of prostacyclin (PGI2) from endothelial cells and vascular tissue in vitro. In this study the effect of using various incubation medias on the release of PGI2 and 6-keto-PGF1α from primary cultures of confluent endothelial cell monolayers (ECM), grown from human umbilical cord veins, was examined. The incubations were carried out for 5 min at 22 °C on a rocker platform. The release of these compounds differed substantially with the various incubation media. Hypotonic and thrombin-containing media produced the greatest release. The 51Cr release from prelabeled ECM was also assayed. A high correlation existed between PGI2, 6-keto-PGF1α and 51Cr occurring in the various media. In an attempt to investigate the effect of the proposed interaction between corticosteroids and adrenaline in stressful situations on fibrinolysis and coagulation, blood plasminogen activator and clotting factor VΠI were measured before and after a prolonged adrenaline infusion (3.5 μg/min for 30 min) in 4 asthmatic patients on long-term corticosteroid (prednisolone) therapy and their responses were compared with healthy controls.
