摘要:
The bovine coagulation factors II, VII, IX and X were separated from other plasma proteins, and each factor was obtained in a largely pure state by various chromatographic procedures in the presence of diisopropylfluorophosphate (DFP). Factor X was separated from factors II, VII, and IX by DEAE Sephadex chromatography, followed by G-100 filtration and hydroxylapatite chromatography. Separation of factors II and IX from factor VII was achieved by chromatography on DEAE cellulose, followed by G-100 gel filtration and hydroxylapatite chromatography. Factors II and IX were separated by preparative polyacrylamide electrophoresis. After DEAE cellulose chromatography, factor VII was purified by gradient elution from hydroxylapatite.

Thrombotest dilution curves and specific inhibitor experiments were carried out in the abnormal factor X (factor X Friuli) plasma and in coumarin plasma. Thrombotest dilution curves obtained with Friuli plasma suggested the presence of a PIVKA-like inhibitor which was not confirmed by the specific experiments. The inhibitory capacity found in Friuli plasma was-0.09 arbitrary units, whereas that found in coumarin-treated patients with a factor X level ranging from 1 to 14% varied between 0.34 and 0.21 arbitrary units.

An activity that converts bovine fibrinogen to fibrin has been isolated from the crude venom of Trimeresurus Okinavensis by means of gel chromatography and ion exchange chromatography on arginine-Sepharose 6B. A high degree of homogeneity is suggested by gel chromatography, gel electrophoresis, isoelectric focusing and end-group analysis. The purified fraction is an acidic glycoprotein with a molecular weight of about 35,000. It catalyzes the hydrolysis of certain arginine esters, but has very low proteolytic activity towards casein. The fraction has a strong catalytic activity for converting fibrinogen into a fibrin-like compound. A certain factor-X-activating ability persists. It does not activate human factor-XIII zymogen in plasma.

Following moderate denaturation of purified fibrinogen by heat or alkali, the thrombin time was more seriously affected than was clottability. Clottability was unaffected by time moderate heat denaturation, denoting that retained clottability after various preparative procedures does not necessarily rule out conformational changes in the fibrinogen molecule. During the action of thrombin upon denatured fibrinogen, enhanced generation of N-terminal glycine was demonstrated, reflecting increased susceptibility to thrombin. This is in accordance with the increased susceptibility of denatured proteins to proteolytic enzymes generally found. Since visible gelation was delayed in denatured fibrinogen, this means that the native conformation of fibrinogen is not essential for the proteolytic action of thrombin, but is essential for the subsequent polymerisation of the fibrin formed.

Hemorrhagíc tendency and granulocytopenia due to prolonged contact with trichloroethylene and toluene are described in a boy whose hobby was model air-plane assembly. The hemostatic defects found were hypoproconvertinemia, moderate hypofibrinogenemia, and increased capillary fragility. The clinical and laboratory abnormalities disappeared following interruption of the exposure. The abnormalities in the coagulation mechanism are ascribed to a toxic effect of the trichloroethylene on the liver, while the granulocytopenia is considered to reflect a bone marrow-depressing action of benzene contaminating the toluene. The increased capillary fragility is presumed to be due to a toxic effect of the chemicals on the blood vessels

摘要:
The fibrinolytic activity of a solution is frequently determined by the fibrin plate method, lytic activities of drops being related to the lysed areas in a fibrin layer. A simple device was designed to measure the diameters of fibrinolysis zones. Using the principle of light scattering by the fibrin layer, we could clearly distinguish lysis zones from non-lysed substrate, even from very transparent fibrin. Application of this principle offers the great advantage that zone diameters can be read off directly, rapidly, and accurately on millimetre graph paper underneath the fibrin plate.
