AbstractEarly changes affecting the principal cellular components of pulmonary alveoli after inhaling239plutonium dioxide were followed by quantitative and qualitative electron microscopy in mice and rats. Different accumulated doses ofairradiation were achieved. The numbers of alveolar macrophages and interstitial mononuclear cells rose in mice especially after a higher dose of radiation, whilst in rats interstitial fibroblasts were increased. The evidence from mice suggested pronounced secretory activity of type II epithelial cells with subsequent uptake of phospholipid by alveolar macrophages, which developed large cytoplasmic inclusions, but rats were much less severely affected. Pneumonitis was not a feature and with the dosage of radiation employed endothelium escaped structural damage. Sensitivity between species differed, both according to cell type and to intensity of exposure, so demanding caution in the application of experimental findings to man.

AbstractA microvascular lesion characterized by endothelial and platelet damage leading to intravascular coagulation, thrombosis and vascular dehiscence with haemorrhage was found in capillaries and venules in the rectal mucosal lamina propria of adults with acute diarrhoea. The lesion morphologically resembled the local Shwartzman reaction. There was no correlation between the prevalence of the vascular lesion and the presence of recognized enteric pathogens, but there was a significant correlation with the clinical severity of the illness. It is suggested that this vascular lesion could be the result of the failure of the epithelioluminal barrier in the colon and it may also be one of the determinants of the clinical severity of illness in adults with acute diarrhoea.

AbstractHuman bone marrow was cultured and cells from individual monocyte colonies stained with a variety of polyclonal and monoclonal antibodies. Erythroid colonies and peripheral blood monocytes were used as controls. Both bone marrow cultured and peripheral blood monocytes stained with antibodies to lysozyme, non‐specific cross reacting antigen (NCA) and alpha‐1‐antitryspin and the cells stained variably for HLA‐DR. A small number of cells in each cultured bone marrow colony stained with antibody to human thymic antigen (HTA T6) and most of the cells stained with variable intensity using anti‐S‐100 protein. No cells reactive with these two antibodies were detected in peripheral blood monocyte preparations and erythroid colony cells were negative for all antigens studied. The presence of cells within individual bone marrow colonies with phenotypic properties of both phagocytic macrophages (lysozyme positive, NCA positive, alpha‐1‐antitrypsin positive) and Langerhans cells/interdigitating reticulum cells (HTA (T6) positive, S‐100 protein positive) suggests that these cells share a common stem cell and are both components of the mononuclear

AbstractSections of human atherosclerotic plaques were stained by the indirect immunoperoxidase technique using three rat anti‐human monoclonal antibodies: YAML 501·4 anti‐leukocyte‐common' (T200) antigen; YTH 8·18 (anti‐macrophage cytoplasm); and YPC 1/3·12 (anti‐smooth muscle cell). The cells of diffuse intimal thickening were almost all smooth muscle cells but there were rare subendothelial macrophages. Focal lesions, in contrast, contained numerous macrophages as well as smooth muscle cells. Macrophage ‘foam cells’ were particularly numerous in fatty streaks and in advanced fibro‐fatty plaques, but were less conspicuous in focal fibro‐elastic lesions. The results confirm that macrophages are an important component of atherosclerotic plaques and suggest that they may have a significant role in

AbstractThe importance of the glycocalyx in controlling responses to proteinases was studied by enzymic removal of sialic acids from the luminal surface of arterial endothelium. In the perfused rat aorta collagenase induced cell detachment in greater numbers from the desialylated than from the untreated endothelium. Neuraminidase treatment increased by three‐fold prostacyclin synthesis by the vessel wall in respons to thrombin. The supernatant from activated human neutrophils, containing elastolytic activity, when perfused together with neuraminidase enhanced and prolonged sialic acid release induced by neuraminidase alone. Thus the effects of different proteinases on various endothelial cell functions are potentiated by removal of surface sialic acid residue

AbstractAn immunocytochemical study was carried out on normal salivary gland tissue and ten salivary gland pleomorphic adenomas. Antibodies to myosin were used to stain myoepithelial cells. Duct cells were stained using an antibody to total keratin and a subpopulation of basal duct cells with an antibody to 45/46K keratins. Basement membranes were stained with anti‐type IV collagen. The results demonstrated that myoepithelial cells are relatively rare in the majority of pleomorphic adenomas and that many of the cells which have been classically described as myoepithelial in routine histological preparations do not clearly show this type of differentiation. However, the tumours presented a spectrum of differentiation patterns from those that were mainly ductal to the rare tumour which was largely myoepithelial. It is further suggested that the 45/46K keratin antibody is capable of identifying a subpopulation of cells which could possibly be important in the histogenesis of this tumou

AbstractA comparison has been made of the effects of different fixation protocols upon the ultrastructure of normal human breast. Pale (B) and dark (A) epithelial cells as described by previous authors are present in tissue fixed with hyperosmotic Karnovsky‐type fixatives, but not where the primary fixative is isosmotic glutaraldehyde. The observed subpopulations of pale and dark epithelial cells have been directly compared with sub‐groups of epithelial cells recognized by the monoclonal antibody LICR‐LON‐M8. AbstractBarrett's epithelium refers to the presence of ectopic mucosal types in the squamous‐lined oesophagus. Previous studies have documented argentaffin and argyrophil‐positive cells as well as gastrin‐like immunoreactivity in oesopha‐geal tissue extracts from patients with Barrett's mucosa. In the present study, 125 oesophageal biopsies obtained under direct vision at endoscopy from 22 patients with Barrett's oesophagus were systematically studied using fluorescence and peroxidase antiperoxidase single and double‐staining immunocytochemical methods employing highly specific antibodies to localize the following peptide‐containing cell types in Barrett's mucosa: gastrin, somatostatin, gastric inhibitory polypeptide, motilin, neurotensin and pancreatic glucagon. In addition, EC cells were localized using a cytochemical silver staining method.
