AbstractNine cases of lymphoblastic lymphoma and eight cases of small, noncleaved undifferentiated non‐Burkitt's lymphoma were studied. All cases were stained with monoclonal antibodies, directed to the leukocyte common antigen (LCA), B‐ and T‐cell markers as well as Calla. Small noncleaved, undifferentiated non‐Burkitt's lymphomas were LCA positive and expressed one or more B‐cell markers. Three cases of lymphoblastic lymphoma were LCA positive and showed reactivity with antibodies OKT4, OKT6 and OKT8. The six remaining cases of lymphoblastic lymphoma were LCA negative, except for only one case that was weakly LCA positive using more sensitive detection methods. Two of the LCA negative cases expressed Calla, and four cases did not show any reactivity with the monoclonal antibodies used. In all LCA negative cases, small residual non‐neoplastic nodules representing remnants of lymphoid follicles, could be detected. Since five out of nine cases of lymphoblastic lymphoma were LCA negative, lack of staining for LCA does not preclude the diagnosis

AbstractThe ultrastructure of interdigitating dendritic cells in dermatopathic lymphadenopathy from patients with mycosis fungoides was investigated. The most remarkable findings were: (i) a marked hypertrophy of the smooth endoplasmic reticulum with a peculiar concentric arrangements of the cisternae around lipid droplets, (ii) an abundance of cytoplasmic lipid droplets and (iii) the presence of paranuclear chromatoid‐like bodies. AbstractRecent evidence suggests that cimetidine given pre‐operatively in primary hyperparathyroidism (1°HPT) might cause structural changes in parathyroid glands, while its suppressive effects on the disease are disputable.To determine these possible changes we studied 38 patients with 1°HPT who underwent parathyroidectomy. In 14 of these (group I) cimetidine was given pre‐operatively (1000 mg orally daily for 4 weeks). The remaining 24 patients (group II) did not take any drug. Parathyroid function was estimated by nephrogenous cAMP (NcAMP) and serum immunoreactive parathyroid hormone (iPTH) measurements. Histological examination of the parathyroids was made by conventional techniques.In group I at the end of cimetidine treatment, the only change observed was a small but significant (p<0·05) decrease of plasma calcium (−0·77 mg/dl). Histologically, the glands of group I—compared with those of group II—showed the following findings: (1) increased gland mass: mean increase 1050 mg (adenomas) and 700 mg (hyperplasias); (2) central oedema inallthe cases of group I only; (3) increased (about 50 per cent) cellular size and (4) intranuclear ‘inclusions’ in 10 out of 14 cases of group I only.It is concluded that treatment with cimetidine in 1°HPT is followed by histopathologic alterations leading to increased size of the dis

AbstractThe present study describes the distribution of Leu M1 (CD15) immunoreactivity in 13 normal epithelia and 14 epithelial neoplasms. The findings are compared with those obtained using other epithelial markers. Cautionary points and broad recommendations are made with regard to use of anti Leu M1 antibody.

AbstractDNA content and cell proliferation were studied retrospectively in 125 patients presenting from 1974–1981 with rectal adenocarcinoma. The presence of DNA aneuploidy or a high level of cell proliferation were associated with a poor prognosis and when combined were the best predictor of survival after Dukes's stage excluding all other pathological assessments investigated. High cell turnover was significantly associated with an infiltrative pattern of growth, an observation that might explain the poorer prognosis of this pattern in rectal adenocarcinoma

AbstractA monoclonal antibody 1‐6E10 against the protein product p62c‐mycof the c‐myc oncogene was used to assess, by immunohistology, a variety of non‐neoplastic and preneoplastic disorders of gastric and colonic mucosa. There were low levels of expression of the c‐myc oncogenic product in normal gastric and colonic tissue. In gastric mucosa, increased expression was observed with inflammatory, metaplastic and dysplastic histological appearances. In the normal colon low levels of expression were observed, but there was increased expression in inflammatory disorders including Crohn's disease and ulcerative colitis. There was also increased expression in colonic dysplasia associated with ulcerative colitis. The oncogene product was localized in the cytoplasm, nuclei and Golgi apparatus. C‐myc 1‐6E10 may therefore be used as a marker to identify the cellular proliferative response in gastric and colonic mucosa that is associated with inflammation as well as potentially neoplastic hyperprolife

AbstractCryostat sections from seventy‐eight female breast carcinomas were assayed for oestrogen receptors by isoelectric focusing. Adjacent cryostat sections stained by Miller's elastic/van Gieson's method were graded for elastosis. Elastosis was similarly graded on near‐equatorial paraffin sections from the same tumours. A positive correlation was obtained between elastosis in the near equatorial sections and oestrogen receptor positivity (p<0·0005), menstrual status (p<0·05) and parity (p<0·01) but no correlation was found between these factors and elastosis graded on cryostat sections from the more peripheral areas which had been selected for oestrogen receptor assay. These observations suggest that the central region of breast carcinomas, where connective tissue responses are fully developed, exhibits grades of elastosis with greater clinical significance. This may explain the conflicting published observations on the correlations between elastosis and oestrogen receptor status, which we believe are due to the lack of uniformity in tissue sampling. AbstractIn serial studies of immunopathologic changes in an animal model of thyroiditis 52 rabbits were immunized with either bovine, porcine or human thyroglobulin (Tg) in Freund's complete adjuvant, while another 47 animals served as noninjected or adjuvant injected controls. The immunized animals were divided into two groups, one receiving an initial series of only three Tg injections while the other received, in addition, challenging injections over an 8‐week period. The immunized animals were killed over a period of 6–34 months after the last Tg injection, and untreated controls were killed at comparable ages. In Tg immunized animals, lymphocytic thyroiditis was encountered in 25 per cent and thyroid amyloid in 17 per cent; glomerular amyloid was encountered in 44 per cent with diffuse lesions in 8 per cent, nodular lesions in 17 per cent and a mixture of the two in 19 per cent. That the thyroid and glomerular hyaline deposits contained amyloid was shown by various histochemical criteria, as well as by the presence of typical fibrils on electron microscopy. Immunohistochemical studies indicated that the amyloid was predominantly of the AA type. Rabbits receiving challenging Tg injections, in addition to the initial series, showed only thyroiditis and nodular glomerular lesions most of which were amyloid. Whilst the vast majority of rabbits with lymphocytic or amyloidotic responses showed both thyroid and renal lesions, a small percentage of animals showed only a lesion of one or the other of these two organs. AbstractThe present study was undertaken in rats to examine the significance of sinusoidal circulatory disturbance by microthrombosis in the pathogenesis of hepatic damage and dysfunction due to endotoxin. Administration of endo‐toxin induced fibrin deposits and infiltration of polymorphonuclear leukocytes in the sinusoids, focal random coagulative hepatocellular necrosis and elevation of serum transaminase activities. When heparin was given simultaneously with endotoxin, the formation of fibrin thrombus in the sinusoids was prevented, and the endotoxin‐induced morphological and functional changes in the liver were markedly inhibited. Infusion of thrombin into the portal vein induced a large amount of fibrin thrombi in the sinusoids, focal random necrotic foci resembling the lesions produced by endotoxin and elevation of levels of serum transaminases.
