AbstractCo‐cultivation of peripheral blood mononuclear cells, from a 4‐year‐old boy with infectious mononucleosis, with a human T lymphotrophic virus type I (HTLV‐I)‐producing T‐cell line resulted in the establishment of an HTLV‐I infected Epstein‐Barr virus (EBV)‐producing B‐cell line. Ultrastructural examination revealed the presence of ribosome‐lamella complexes (RLCs), in a small proportion of long‐term cultured cells. Evidence is presented of continuity between RLC and the rough endoplasmic reticulum, and in some instances there was also an intimate relationship with tubuloreticular structures. The findings support the view that RLC is derived from the rough endoplasmic reticulum and also provide further evidence of the non‐specific nature of RLC

AbstractThec‐erbB‐2oncogene encodes a 190 kD transmembrane growth factor receptor which is closely related to the EGF receptor and has been found to be amplified and overexpressed in a number of human adenocarcinomas, particularly of the breast. We have analysed, by immunocytochemistry using the 21N antibody, expression ofc‐erbB‐2in a retrospective series of pancreatic adenocarcinoma, chronic pancreatitis, and examples of histologically normal pancreas. In three cases (21 per cent) of chronic pancreatitis, there were focal areas of cytoplasmic immunoreactivity in regenerating epithelium. In 15 cases (17 per cent) of pancreatic adenocarcinoma, cytoplasmic immunoreactivity was seen, while in two cases (2 per cent) strong membrane staining of tumour cells was seen which could be blocked by peptide controls.c‐erbB‐2immunoreactivity was also demonstrated using a second antibody, 20N, which recognizes another peptide sequence of thec‐erbB‐2protein. There was no relationship between immunoreactivity and histological subtype or grade, but there was absolute concordance between staining in primary and metastatic deposits. Since the rat homologue (neu) of thec‐erbB‐2oncogene may be activated by a specific point mutation in its transmembrane region, we have analysed 23 cases from this series for mutations by polymerase chain reaction amplification and sequence‐specific oligonucleotide hybridization. We were unable to identify activity mutations in this series. These data suggest that there is abnormal expression ofc‐erbB‐2oncogene in nearly 20 per cent of cases although mutational activation of this gene is not seen in human

AbstractType I and type III intestinal metaplasia in gastric mucosa have been examined using morphometric methods. Tissue (volume per cent gland, lumen, epithelium, goblet cell vacuoles) and nuclear parameters (area, with related standard deviation, and form factors) were used as indicators of gland crowding, nuclear‐cytoplasmic ratio, nuclear atypia, and pleomorphism. In type III intestinal metaplasia, there is significantly (i) greater nuclear pleomorphism, (ii) a higher nuclear‐cytoplasmic ratio, and (iii) smaller and less numerous goblet cell vacuoles in both the upper and the lower parts of the crypts. These two parameters have significantly higher values in the lower than in the upper part of individual crypts. AbstractMast cells are closely associated with nerves in the mucosa of the appendix vermiformis, and obliteration of the appendiceal lumen by fibrous tissue is accompanied by neurogenous hyperplasia. However, changes in the density of mast cells in this process have not been reported. Accordingly, fibrosis was graded in haematoxylin and eosin sections from 46 samples of human appendix. This was compared with mast cell number in toluidine blue‐stained slides and nerve density in PGP9.5‐immunoreactive sections. In the mucosa, the mast cell number in the samples with minimal fibrosis was three times greater than in those classified as normal (P<0.0001), and this declined in the more fibrotic samples. The mucosal nerve scores paralleled the mucosal mast cell changes, and stereological analysis revealed a correlation of mast cell number and nerve density within the lamina propria of the same specimens (r=0.49–0.90). In the submucosa, mast cell numbers and nerve scores were not significantly different in the different histological grades and obliterated samples resembled normal submucosa, except that a dense axial block of nerve staining was often present. AbstractIn two members of an affected family with a hereditary syndrome of proctalgia fugax and constipation, a hypertro‐phied internal anal sphincter was found with histological features suggesting a myopathy of this muscle. In these two patients, and in an unrelated patient with a similar clinical syndrome, smooth muscle fibres of the internal anal sphincter showed numerous vacuoles, many of which contained ovoid inclusion bodies. The structural features and histochemical reactions of the inclusion bodies were consistent with a polyglucosan composition. Histological examination of the internal anal sphincter may reveal smooth muscle abnormalities in functional bowel disorder

AbstractThe combination of trioxsalen bath and long‐wave UV‐A is used in dermatology to treat selected patients with psoriasis. Such treatment induces side effects whose clinical aspects have already been described. We report the histological aspects of these lesions. The most striking features occur in the epidermis, where we found vacuolar alterations, disorganization of the stratum malpighi, necrotic cells, and Bowenoid changes. Langerhans cells are reduced in number, and melanocytes may become large and atypical. In addition, factor XIIIa‐positive dendrocytes of the superficial dermis increase in size, and some of them migrate into the epidermis, which is a rare location for these

AbstractChronic serum sickness glomerulonephritis was induced in rats using125I‐labelled cationic bovine serum albumin as antigen. During the recovery period the animals were given protamine or polyethyleneimine (PEI), both of which are cationic, or heparin, which is anionic. A control group received saline. The cationic molecules were not shown to influence the rate of removal of antigen from the glomeruli, but heparin increased removal. In subsequent experiments this effect of heparin was confirmed, and a similar effect was demonstrated using low molecular weight heparin. These results indicate that large doses of heparin can enhance the removal of antigen from well‐established glomerular deposits. Fibrin is not detectable in the glomeruli in this model, so this effect of heparin may be independent of its anticoagulant propert

AbstractThis paper reports the occurrence of a rare, yet distinct pituitary adenoma which was surgically removed from a 42‐year‐old male with both clinical and biochemical evidence of acromegaly and mild hyperprolactinaemia. The monomorphic adenoma consisted of mature cells which were ultrastructurally indistinguishable from those of a prolactinoma. Electron immunocytochemistry, including a series of double‐labelling techniques using selected colloidal gold particles as markers, indicated the presence of a pituitary adenoma in which the cells were capable of simultaneously producing growth hormone and prolactin and packaging them within the same secretory granule.
