AbstractThis paper describes the pathology of pulmonary histiocytosis developing in a patient treated with amiodarone, and reports comparable changes in the lungs of rats and mice to which this drug had been administered. Ultrastructural studies indicate that the experimental lung changes are similar to those produced by iprindole and chlorphentermine. The patient also displayed focal necroses and an eosinophilic bronchiolitis: these may represent further effects of amiodarone on the lung.

AbstractThe origin of resident peritoneal macrophages was studied in radiation mouse chimaeras with and without reconstitution of the peritoneum with viable isogeneic peritoneal cells. The selection of host and donor strains were such that the isoenzymes of glucose‐6‐phosphate isomerase could be used to distinguish host from donor bone marrow derived cells. AbstractHalf of the offspring from nicotine exposed and control mother mice were cross‐fostered at birth to form four groups of those exposed to nicotine only during the gestational period (NC), only during the lactational period (CN), throughout both periods (NN), or exposed to none (CC). Nodular mucosal bulges (neuroepithelial bodies, NEB) were found in all groups; their number decreased with the increase in the age in the control group (CC). The bulges were large and irregular in all nicotine exposed groups, especially in the NN group. Although they also decreased in number with age, NEB were significantly retained and abnormal in the NN group compared with the NC and CC groups on day 30 (p<0.01). In addition, the bronchioles were tortuous in the nicotine exposed groups probably due to the expansion of mucosal depressions, pits, and fenestrations. In mice, maternal exposures to a small dosage of nicotine throughout both gestational and lactating periods appear to induce more baby lung changes than when the exposure to nicotine is limited to either period alone. AbstractThe binding of peroxidase‐labelledwheat germ agglutinin(WGA) to a series of 125 human breast carcinomas has been studied. Routine fixation and processing has been shown to have no deleterious effect on reactivity. In contrast to the homogeneous binding of WGA to normal and hyperplastic human breast previously reported, the binding of WGA to breast carcinomas exhibits heterogeneity. This is of two forms: (1) in relation to cell reactivity, which is significantly correlated to tumour differentiation, with fewer cells reacting in those carcinomas showing loss of histological differentiation; (2) with regard to the interaction between WGA and specific saccharides of cellular glycan‐components, binding to sialic acid appearing to be of significance to varying degrees in some carcinomas but not in others nor, as previously reported, in normal breast. The alteration in WGA–saccharide interaction is an indicator of the heterogeneity of the carbohydrate chains of glycoproteins within neoplastic cells of the same tumour and between carcinomas. AbstractTissue sections from 80 human breast carcinomas have been examined for evidence of binding of the two fucose‐specific lectins,Lotus tetragonolobusandUlex europeus I, with enzymes utilized as an indicator system. Both singlestaining and double‐staining methods (Lotus tetragonolobus‐peroxidaseandUlex europeus I‐alkaline phosphatase) have been employed. In contrast to normal breast in which there is consistent reactivity of all epithelium withLotus tetragonologusa variable loss of binding has been found within carcinomas. Surprisingly, this bears no relationship to tumour differentiation; nor does it correlate with local lymph node metastasis.Ulex europeus Ihas a variable reactivity with carcinomas, as it does with normal breast epithelium, and this likewise has no correlation with differentiation. However, a relationship has been found between specific patterns of binding and axillary lymph node status. AbstractX‐ray energy spectroscopy (XES) of all particulate matter found by light microscopy in histological sections of 222 oral lesions revealed a much wider range of elements than expected from dental amalgam. Subsequent XES of a variety of endodontic materials, impression materials and toothpastes indicated possible sources for virtually all elements identified in the histopathological lesions. AbstractTissues from five cases of angiofollicular lymph node hyperplasia have been studied. All had the histological structure of the hyaline‐vascular type of lesion; large numbers of very compact lymphoid follicles were distributed evenly throughout a highly vascular tissue. The follicles were characterized by their small size, a vascular poorly cellular and frequently hyalinized centre, and a ‘tight’ concentric mantle of small lymphocytes arranged in layers producing an ‘onion‐skin’ appearance. The interfollicular tissue was characterized by the large numbers of small vessels mainly hyalinized capillaries and a few high endothelial venules and the presence of variable numbers of lymphocytes, plasma cells, immunocytes and immunoblasts. The immunoperoxidase method demonstrated polytypic cytoplasmic immunoglobulin in the small numbers of centroblasts and plasma cells within the follicle centres and in the plasma cells and immunocytes in the interfollicular tissue. Large numbers of suppressor T cells were present in the interfollicular areas and only scattered helper T cells were seen within the lymphocyte mantles. A strong reaction for factor VIII‐related antigen was seen in the endothelium of the interfollicular high endothelial venules but only a weak reaction in the vessels in the follicle centres. A concentric distribution pattern of the dendritic reticulum cells was seen with the metalophil impregnation method of Marshall and with the enzyme histochemical methods for acid α‐naphthyl acetate esterase and 5′‐nucleotidase. This pattern differs from the zonal distribution of these cells seen in reactive lymphoid follicles. The nature and possible pathogenesis of AFLNH are discussed and contrasted with
