There are patterns in the distribution of the characteristics of vascular α-adrenoceptors. Sudden changes in their mechanism occur at particular sites along the main branches of the aorta; in some regional vascular beds, but not others, there is a progressive reduction in the response that can be elicited through the α-adrenoceptor; in smaller blood vessels, α- and β-adrenoceptors may be spatially separated in the vessel wall; in most blood vessels, the inner smooth muscle is more sensitive to α-agonists and more dependent on sequestered calcium than the outer. Presynaptic release-modulating receptors on sympathetic nerve fibres have been found not only in a great number of animal tissues but also in various human blood vessels. Although basically the same types of receptors are present in animals and man (e.g., inhibitory muscarinic receptors and α2-adrenoceptors; facilitatory β-adrenoceptors), there may be considerable species differences if a certain blood vessel or the effect of a certain agonist or antagonist is considered. Hormones may also influence noradrenergic transmission by acting presynaptically. Thus, ACTH increases the impulse-evoked noradrenaline (NA) release in the rabbit pulmonary artery by activating presynaptic ACTH receptors, and an adenylate cyclase may be involved in this effect. Results obtained with new pharmacological tools (forskolin and selective inhibitors of cAMP phosphodiesterase) support the suggestion that the sympathetic nerves of the rabbit pulmonary arteries are endowed with an adenylate cyclase activation of which increases evoked NA releas

摘要:
By use of monoamine reuptake inhibitors, it is possible to study the intra- and extraneuronal components of monoamine deamination in brain. In both man and rat, the ratios of activities of MAO-A/MAO-B are higher intra- than extraneuronally. As a result of this difference in cellular localisation in the brain, loss of neurones produces a change in the ratios of activities of MAO-A/MAO-B. It is also possible selectively to inhibit the activity of MAO-A within a given neurone population. The α-methyl substituted monoamine amiflamine has been shown to inihibit MAO-A within serotoninergic neurones at lower doses than are required for inhibition in other neurones. The usefulness of this compound in the elucidation of the functions of intra- and extraneuronal MAO are discussed
