AbstractRabbits were inoculated with affinity‐purified human antibodies to the common house dust miteDermatophagoides pteronyssinus(DPT) to produce anti‐idiotypic (Id) antibodies. The Id carried by human IgE antibodies were studied in a radio‐immunoassay in which specific anti‐DPT IgE were first insolubilized on rabbit F(ab′)2fragments against human Fcϵ.In a single individual specific anti‐DPT IgE antibodies shared bystander idiotopes (i.e.idiotopes located outside the antigen‐binding site) with IgG antibodies of the same specificity, since the adsorption on insolubilized anti‐DPT IgG almost completely eliminated the binding of anti‐Id antibodies to IgE. This differed from our previous observations (J.M. R. Saint‐Remy et al.,Eur. J. Immunol.1986.16: 575) wherein distinct paratope‐associated idiotopes were identified on IgG and IgE anti‐DPT antibodies.The frequency of cross‐reactions between idiotopes of anti‐DPT IgE antibodies observed in ten unrelated individuals was less than 10% for bystander idiotopes. This finding confirmed the private character of the above idiotopes that we found earlier on anti‐DPT IgG antibodies. However, one anti‐Id antibody preparation was found to react with bystander idiotopes of anti‐DPT IgE of most individuals, thereby suggesting the existence of a recurrent idiotope. AbstractIgG isolated from the plasma of seven individuals hypersensitive to the common house dust miteDermatophagoides pteronyssinus(DPT) was exhaustively adsorbed onto insolubilized DPT. The unbound fraction was found by radioimmunoassay to contain antibodies recognizing the variable region of both anti‐DPT IgG and IgE antibodies. This recognition was idiotype (Id)‐specific as it persisted after passage over insolubilized polyclonal IgG of unrelated specificity.Most of these anti‐Id IgG carried the internal image of the initial antigen in that they competitively inhibited the binding of anti‐DPT antibodies to DPT. Immunoadsorption of anti‐Id IgG onto insolubilized anti‐DPT IgG antibodies from the same individual completely eliminated their reaction with anti‐DPT IgG but not with anti‐DPT IgE, suggesting that idiotopes included in the antigen‐binding site of specific IgG and IgE antibodies were not identical. Anti‐Id IgG recognizing idiotopes located outside the antigen‐binding site (bystander idiotopes) were also completely removed by passage over insolubilized anti‐DPT IgG; in this case the reaction of the anti‐Id IgG with both anti‐DPT IgG and anti‐DPT IgE was inhibited, indicating that, for a given individual, bystander idiotopes were shared between anti‐DPT antibodies p

AbstractThe modulation of surface molecules on B‐chronic lymphocytic leukemia (B‐CLL) cells was studiedin vitroby incubation with CD19, CD20, CD21, CD5 and anti‐IgM antibodies. The cytoplasmic changes were evaluated by analyzing the organization of cytoskeletal F‐actin and the adhesive properties of stimulated B‐CLL cells. The following observations were made: (a) CD5 antigen is capped on the surface of B‐CLL cells (but not on normal CD5+B and T lymphocytes), (b) On B‐CLL cells, CD5 and CD21, the receptors for the C3d fraction of complement, co‐cap and co‐modulate while surface IgM as well as CD19 and CD20 do not cap. (c) B‐CLL cells, after capping of CD5 or CD21 surface molecules, fail to organize F‐actin into podosomes. (d) The pre‐incubation of B‐CLL cells with either CD5 or CD21 antibodies prevents their binding to purified C3d protein used to coat coverslips. These data indicate that an intimate spatial relationship exists between CD5 and CD21 molecules on the B‐CLL surface. AbstractMouse cell lines of different lineages have been established which constitutively secrete large quantities of recombinant mouse interleukins (mIL2, mIL3, mIL4 or mIL5). An existing bovine papilloma virus‐based expression vector, pBV‐lMTHA, was modified to allow transformed X63Ag8‐653 myeloma cells, NIH 3T3 fibroblasts and C127 mammary tumor cells to stably carry multiple copies of the vector, to express the inserted cDNA encoding a single interleukin constitutively, and to secrete the interleukin in high quantities.Cell lines transformed with mIL2 cDNA stably carried 30–100 copies of the plasmid per cell and constitutively secreted biologically active mIL2 in quantities similar to those produced by murine EL4 thymoma cells or rat spleen cells stimulated with mitogens. Deletion of the 3′ untranslated region containing AT‐rich sequences from the mIL2 cDNA resulted in a 100‐fold increase in the constitutive production and secretion of mIL2 by the transformants. Addition of a heavy metal further increased the production 2 to 6‐fold. Cells transformed with 3′‐deleted mIL3 cDNA constitutively secreted 300–1000 times higher activities of mIL3 than the myelomonocytic leukemia line WEHI 3. mIL 4 produced by the similar transformants induced [3H]thymidine uptake of a T cell line, a mast cell line and B leukemia cells, and enhanced the production of IgG1by B cells. IL 4 liters were 150 times higher than those produced by the concanavalin A‐stimulated T cell line 2.19. mIL 5 was secreted by similar transformants at 10‐fold higher titers than those produced by concanavalin A‐stimulated 2.19 T cells, as judged by the proliferation and maturation of B cell leukemia BCL1. The expression vectors should be useful in establishing eukaryotic cell lines producing proteins from full length cDNA clones at higher rates. AbstractMedia conditioned by human mononuclear cells contain a factor which renders freshly purified T lymphocytes motile and which is distinct from interleukin 1 and interleukin 2. The motility induced by this factor is morphologically indistinguishable from spontaneous lymphocyte motility. The factor is most effective when allowed to attach to the substratum. The effect of the factor on lymphocyte motility is detectable within 30 min but increases over a period of several hours. The relatively rapid action taken together with the ability to interact with both lymphocytes and the substratum point to the possibility that the factor rendering lymphocytes motile is a ligand which mediates cell‐substrate adhesion. AbstractThe effects of transforming growth factor‐beta (TGF‐β) on the proliferative response of human B cells to the low molecular weight B cell growth factor (BCGF) have been investigated in this study. It was found that TGF‐β”, at picomolar concentrations, strongly inhibited the BCGF‐induced proliferation of anti‐μ. chain orStaphylococcus aureusCowan I‐activated human B cells and also of a BCGF‐dependent cell line derived from a human lymphocytic nodular lymphoma. This inhibitory effect was detected in normal and serum‐free culture conditions. The suppression was greatly reduced when TGF‐β was added to the culture one day after BCGF and could be reverted by removing TGF‐β from the culture medium. Since TGF‐β has been detected in supernatants from activated T cells, this factor may represent an important regulatory molecule in the feedback

AbstractSurface‐labeled Epstein‐Barr virus (EBV)‐transformed lymphoblastoid RPMI 8866 cells release in their supernatant a radiolabeled 25‐kDa polypeptide which reacts with the Fee RL/CD23‐specific monoclonal antibody (mAb) 25 and which binds to IgE but not IgG (IgEBF/sCD23). IgE BF/sCD23 had an isoelectric point of 4.5–5.0. The reactivity of mAb 25 with IgE BF/sCD23 allowed us to set up a radioimmunoassay for detection of IgE BF/sCD23 in cell culture supernatants. Supernatants from Fee RL/ CD23+cell lines were found to contain IgEBF/sCD23. Addition of human recombinant interleukin 4 (IL4) to normal human B cells cultures induced the production of IgEBF/sCD23. Activation of B cells with anti‐IgM antibody coupled to beads enhanced the IL4‐induced production of IgEBF/sCD23 when compared to nonactivated B cells. This correlates with the finding that anti‐IgM antibody‐activated B cells cultured with IL4 express more FceRL/CD23 than B cells cultured with IL4 alone. The biochemical characteristics of radiolabeled IgE BF/sCD23 immunoprecipitated by mAb 25 from the supernatants of normal B cells cultured with IL 4 were identical to those of the IgEBF/sCD23 isolated from EBV‐transformed cell line supernatants. Addition of interferon‐γ to B cells cultured with IL 4 strongly decreased the level of IgE BF/sCD23 in culture supernatants correlating with the observed decrease of FceRL/CD23 on B cell surface. AbstractSera from 120 Kenyan schoolchildren who were infected withS. mansoniwere individually examined, using an enzyme‐linked immunoabsorbent assay (ELISA), for the presence of IgG and IgM antibodies reactive with antigens derived from adult worms, the outer membrane of the schistosomulum or from the parasite egg. In addition, antibodies against more purified egg antigens, an egg stage‐specific glycoprotein preparation and a polysaccharide egg antigen known to share epitopes with the schistosomular surface were measured in ELISA, as were antibodies reactive with trichloroacetic acid‐soluble and periodate‐insensitive antigens derived from the outer membrane of schistosomulum and antigens shed when schistosomula were culturedin vitro.IgG subclass responses to the unfractionated egg antigen were also measured. The results from each of these assays were compared with the results of each other assay and with the number of parasite eggs excreted by each child, using Spearmans rank correlations.These comparisons revealed a number of statistically significant positive correlations. IgG4anti‐egg antibodies correlated better with intensity of infection than did other IgG subclasses. Total IgG responses against polysaccharide antigens did not correlate with intensity of infection as well as IgG responses against other antigens; epitopes shared between the schistosomulum surface and the adult worm were different to those shared with the parasite egg; and, there was antigen‐directed restriction of IgG subclass responses to some egg and adult worm antigens which carried these shared epitopes. AbstractPreviously we have shown that interleukin 2 (IL 2) is an essential mediator in T cell‐dependent B cell differentiation induced by pokeweed mitogen. Here we show that activation with monoclonal anti‐CD3 antibodies of peripheral blood T cells led to the induction of helper activity for IgM secretion by human B cells from a prolymphatic leukemia. AbstractcDNA clones which seem to represent the alleles of HLA‐A and ‐B expressed by the thymoma MOLT‐4 have been isolated and used as locus‐specific probes to measure the corresponding mRNA levels in MOLT‐4 and other human thymocyte lines, and the effect of interferon (IFN)−α and −γ on these levels. It is shown in MOLT‐4, and in its derived line YHHH, that HLA‐B mRNA levels are undetectable before treatment but respond to IFN‐α and −γ more markedly than those of HLA‐A. This differential induction is best shown with YHHH, which is hypersensitive to IFN‐α, where the HLA‐B mRNA levels increase to a level threefold those of HLA‐A. Other thymocyte lines tested also showed preferential induction by IFN‐α of HLA‐B, although the basal levels of HLA‐A and ‐B tended to be similar.
