Ultrastructural characteristics of human T cell clones with various cytolytic activities |

AbstractFifteen T cell clones with different (specific, antibody‐dependent or natural killer‐like) cytolytic activity were derived from mixed lymphocyte culture activated T cells and analyzed for their morphological characteristics and, in some instances, for their surface markers.All of the five cytolytic clones analyzed by electron microscopy possessed numerous electron‐dense granules and in some instances multivesicular bodies, with or without an electron‐dense matrix, that are the putative precursors of the granules. In addition, light microscopy examination of semithin sections of other ten cytolytic clones showed that the large majority of the cells in each clone had numerous toluidine blue‐stained cytoplasmic granules. It is of note that nine clones without detectable cytolytic activity analyzed by electron microscopy did not possess granules and presented the features of large agranular blasts. Ten cytolytic clones were analyzed for different surface markers including rosette formation with sheep erythrocytes (E rosettes), receptors for the Fc portion of IgG or IgM (FcγR and FcμR) and a group of antigens recognized by monoclonal antibodies including Ia, 4F2, OKT8 or OKT4. All the clones were Erosette+, Ia+and 4F2+. Expression of FcγR was restricted to the clones active in antibody‐dependent cell‐mediated cytotoxicity. Expression of OKT8 and OKT4 antigens was variable; in particular, five clones were OKT8+/OKT4−, whereas the other five expressed the OKT8−/OKT4+phenotype. It is noteworthy that the ultrastructural features of cytolytic T cell clones are similar to those of large granular lymphocytes, known to be the only lymphoid cells in normal peripheral blood which possess cytolytic activity. AbstractRabbit anti‐mouse brain (RaMBr) antiserum can induce Lyt‐1+, Lyt‐2−, T cells to proliferate and stimulates the same T cell subset to induce B cell proliferation. The aim of this report is to demonstrate that the mitogenic determinant recognized on the T cell surface by RaMBr antiserum is located on the Thy‐1 molecule expressing the products of the Thy‐1aand Thy‐1balleles. Evidence is drawn from serological and genetic experiments. The brain T cell cross‐reactive, mitogenic determinant is not expressed on Thy‐1−mutants of the BW5147 T cell lymphoma that fail to express the Thy‐1 molecule but do express other T cell surface proteins such as T‐200 and gp 69, 71. Monoclonal anti‐Thy‐1.1 and anti‐Thy‐1.2 antibodies block the binding to the appropriate T cells of the majority of the serum antibody from RaMBr antiserum. The absorption of mitogenic antibody was blocked in a similar fashion, thus demonstrating the close association of the determinant and the Thy‐1 antigen defined by monoclonal alloantibodies. The mitogenic and Thy‐1.1 determinants are probably located on the same molecule because of the data obtained with the BW5147 Thy‐1−mutants and the observation that Thy‐1aT cells, which express a lower level of surface Thy‐1 than Thy‐1bT cells, also express lower levels of the determinant recognized by RaMBr antiserum. Furthermore, in (AKR × DBA/2)F1mice (Thy‐1a/b) which express less Thy‐1.1 antigen than Thy‐1.2 at the surface, the mitogenic determinant was found to be prefentially associated with Thy‐1.2. The coordinated genetic control of the surface levels of the Thy‐1 determinant and the mitogenic determinant suggests that both determinants

AbstractIt has been previously reported that lipopolysaccharide‐stimulated murine astrocytes produce a factor which enhances the proliferative response of thymocytes to lectins. The present report demonstrates that a rat astrocytoma cell (C6cells)‐derived factor appears to be similar to macrophage‐derived interleukin 1 (IL1) in its biological activities and biochemical characteristics. Upon injection into mice, supernatants of C6cells induce the production of serum amyloid A. The C6cell‐derived factors enhance the response of thymocytes to phytohemagglutinin and the growth of fibroblasts whereas no effect on the growth of neuroblasts and of a strictly interleukin 2 (IL2)‐dependent T cell line was observed. On an AcA 54 column the C6‐derived factors acting on thymocytes and fibroblasts coeluted as a single peak of Mr>= 13500 to 18000; the semipurified factor was found to enhance the lymphocytes' production of IL2. AbstractA phagocytic cell of the thymic reticulum in culture (P‐TR‐C) has recently been isolated whose characteristics closely resemble those of the thymic interdigitating cell despite the modifications induced by culture conditions.In the present report it is demonstrated that this cell population is able to produce both interleukin 1 (IL1) and prostaglandin E2(PGE2). IL1 has no direct mitogenic effect on thymocytes but is comitogenic with concanavalin A.
