AbstractThe isolation of two chymotryptic peptides, each containing a tyrosyl residue considered to be derived from the hapten‐binding site of a rabbit anti‐p‐azophenyltrimethylammonium antibody preparation, was achieved by the paired‐iodination procedure. One peptide, from a heavy chain, corresponded to the sequence ‐Cys‐Ala‐Arg‐Asp‐Pro‐Tyr‐ which, on the basis of comparison with other sequences, represents positions 97–102 of the heavy chain. AbstractInsoluble immune complexes and immune aggregates were found to stimulate the activity of the hexose monophosphate shunt as measured by nitroblue tetrazolium reduction in human polymorphonuclear leukocytes. This phenomenon depends more on the size of the complex than on its ability to activate complement.

AbstractCortisone‐resistant thymocytes, spleen cells, thoracic duct lymphocytes, peritoneal exudate cells and peripheral blood lymphocytes of BALB/c mice were immunizedin vitroagainst syngeneic HPC‐108 plasma cell tumor cells. Cocultivation of spleen lymphocytes together with HPC‐108 cells generated the highest cytotoxic immune response in comparison to other lymphocyte sources. Cytotoxicity was tested in a 6 hour51Cr release assay using HPC‐108 cells as target cells.The use of AKR anti‐θ C3H serum indicated that thymus‐derived (T) lymphocytes are essential to the initiation phase of the immune response to plasma cell tumor cells. AbstractThe nature of mouse lymphoid cells induced to proliferatein vitroby a number of stimulating agents has been explored by 1) karyotypic analysis of the mitoses observed in stimulated spleen cell cultures (arrested on sequential days) of chimaeras bearing chromosomally marked T cells; 2) comparison between the response in DNA synthesis of total spleen cells and of spleen cells depleted of thymus‐dependent lymphocytes by treatment with anti‐MSLA (anti‐mouse‐specific lymphocyte antigen) + C, and 3) study of the response of cortisone‐resistant thymocytes (CRT) to some of these agents. Pokeweed mitogen stimulated both B and T cells equally and simultaneously. Rabbit anti‐mouse Ig induced moderate proliferation of B cells exclusively. The response to lipopolysaccharide was entirely B cell‐dependent, but evidence was found of some T cell proliferation following or accompanying the stimulation of B cells. A similar observation was made in the case of “primary” or “secondary”in vitroresponse to bacteriophage T4. Good stimulation with sheep red blood cell stromas or mitogen‐treated xenogenic spleen cells were observed only in cultures from primed animals, and consisted of a mixed B and T cell proliferation with a majority of B mitoses, even at the earlier stages of the response. That this “secondary” response was not totally T cell‐dependent was suggested by the observation of a stimulatory effect of these antigens on T‐depleted spleen cell cultures which were found in parallel experiments to be totally unresponsive to PHA. Finally, using chimeric mice treatedin vivowith vinblastine prior to establishment of the spleen cell cultures, evidence was obtained that part of the B cells stimulated to dividein vitroby several of these mitogens belong to a rapidly

AbstractThe effect of dextran sulfate on [3H]dThd incorporation in lymphoid cells was investigated. The polyanion activated DNA synthesis in spleen and bone marrow cells of normal mice. The highest rate of activation was detected in spleen cells of athymic (nude) mice; the ratio of [3H]dThd incorporation was higher in TxBM spleen cells of thymectomized, lethally irradiated and bone marrow protected mice than in spleen cells of normal donors. Thymus cells of normal mice could not be stimulated, but a slight response was obtained in cortisone‐resistant thymus cells. A synergetic effect was found in normal thymus cells using a combination of dextran sulfate and phytohemagglutinin (PHA). AbstractThe effect of eleven different polyanions, among them dextran sulfate, and of two different preparations of dextran was investigated on [3H]dThd incorporation in spleen cellsin vitro, and on PFC response to SRBCin vivo.In general, polyanions stimulated DNA synthesis in spleen cells and enhanced PFC response to SRBC.Dextran itself had no effect on DNA synthesis and on PFC response. The degree of activation of DNA synthesis by polyanionsin vitrowas found to correlate with the adjuvant activity of the substancesin vivoIt is postulated that activation of DNA synthesis in lymphoid cells is essential for the adjuvant activity of polyanions.

AbstractBrief exposure of mouse spleen cell suspensions to levan (LE)in vitroinduced a state of stable, specific unresponsiveness in these cells, as demonstrated by LE challenge 1 – 15 days after transfer of LE‐treated cells into lethally irradiated recipients. These mice showed unimpaired responsiveness to type 3 pneumococcal polysaccharide (SIII) and sheep erythrocytes as compared with recipients of untreated cells.Suppression was attained by incubation in amounts of LE varying from 25 μg up to 10 mg/spleen equivalent and the increment in effect with rising concentrations of LE was relatively small. Tolerance inductionin vitrowas not dependent on the extremely high molecular weight of LE (mol. wt. 2 x 107) because it was equally demonstrable when hydrolyzed LE with a 1000‐fold reduction in average mol. wt. was used. Induction of unresponsiveness to LE could be prevented by exposure of spleen cells to anti‐mouse Fab serum prior to incubation with LE.In contrast, attempts to produce similar suppression using SIII were completely unsuccessful. It is concluded that the difference in behavior of these two thymus‐independent antigens may be a reflection of weaker overall binding of SIII to IgM receptors on B cells than is the case with LE. Consistent with this view is the demonstration that highly depolymerized LE retained the capacity to inhibit the detection of LE‐specific plaque‐forming cells, whereas SIII of similar mol. wt. had lost

AbstractMouse myeloma cells have been stained in suspension at 0 °C or at room temperature with fluorescein‐labeled rabbit anti‐mouse immunoglobulin sera (F‐RAMIG) or with unlabeled RAMIG followed by fluorescein‐labeled goat anti‐rabbit immunoglobulin sera (F‐GARIG). Tissue culture adapted murine myeloma cells 5563 (IgG2aκ) have been used most extensively in these studies. 95–100 % of the cells show surface fluorescence at both temperatures after staining with F‐RAMIG or with RAMIG followed by F‐GARIG. At 0 °C the staining is diffuse over all the surface, at room temperature the staining is much brighter and patchy. The nature of the H and L‐chains at the surface of the myeloma cell is shown to parallel the type of secretory product. Thus, 5563 cells stain brilliantly with anti‐IgG and anti‐κ chain sera, but not at all with anti‐μ or anti‐α chain sera, which respectively stain TEPC 183 (IgMκ) and MOPC 47A (IgAκ) cells brilliantly. Also, MOPC 315 cells (IgAλ) do not stain with antisera against 5563 myeloma protein, but do stain with antisera against MOPC 47A and MOPC 104E myeloma proteins.Immediately after staining at 0 °C or room temperature, the fluorescence is located over the whole surface. However, if the stained cells are incubated at 37 °C in tissue culture medium, the stain gradually localizes to one pole of the cell such that by 5 h, about 50 % of the cells show staining localized to one hemisphere. The presence of the RAMIG‐antigen complex seemed to have little or no effect on the rate of secretion of myeloma protein during a 5 h period. AbstractThe capacity of mice to produce antibody‐forming cells (AFC) to sheep erythrocytes and to low doses of two haptenated proteins increases logarithmically with age in the postnatal period. This increase is prevented by thymectomy. These findings, together with those on the ability of suckling mice to make high numbers of AFC to high doses of at least one of the antigens used, indicate that T cells are the limiting cell type in neonatal humoral immune responses. An analysis of variance of the number of AFC in individual spleens as a measure for T cells showed that the data are in agreement with random generation and subsequent proliferation of these cells during ontogenesis.
