AbstractMouse spleen cells have been fractionated into three subpopulations by density gradient centrifugation and the effect of interferon on the ability of each fraction to generate an antibody responsein vitrohas been examined. The results show that only the antibody‐forming cell precursors (AFCP) of the high density, late responding fraction are suppressed by interferon. The presence of interferon had no effect on either the number or the mean size of clones of antibody‐forming cells in cultures of the low and medium density fractions. Interferon‐mediated suppression of the response of high density precursors was due to a decrease in the number but not the size of responding clones. These results suggest that interferon suppresses antibody responses by affecting the degree to which high density AFCP can becoming activated rather than the degree to which precursors prolif

AbstractNonadherent and nonphagocytic lymphoid cells from human peripheral blood became strongly cytotoxic against51Cr‐labeled chicken red blood cells and cells from an established human myeloma cell line when subjected to repeated cycles of washing in phosphate buffered saline or treated with trypsin or lecithinase. Prior to augmentation the effector cells pass nylon wool columns that remove practically all surface IgG‐positive cells, but after augmentation they are retained in such columns. Augmentation docs not make them phagocytic or adherent to plastic surfaces. Incubation at 37°C of augmented cells prior to addition on the target cells restores the original nonaggressive state. AbstractThe surface membrane of rat thymocytes and lymph node cells was analyzed by comparing the capability of different rat cell types and of solubilized and chromatographically separated rat thymocyte membrane fractions to absorb cytotoxic antibodies on rat thymocytes and lymph node cells from a heterologous anti‐rat thymocyte serum. The experiments described cause distinct antigenic specificities of the thymocyte membrane to be brought into relationship to structures of defined physico‐chemical properties, and thus are an approach of a thymocyte cell surface mapping in antigenic as well as in molecular terms. AbstractA specific state of resistance to local graft‐vs.‐host (GVH) reactions can be induced in F1hybrid rats, derived from Ag‐B incompatible matings, as a consequence of inoculation with low numbers of parental strain lymphocytes. The magnitude of GVH resistance is markedly and directly dependent upon the number of parental strain lymphocytes used in the pretreatment regimen. AbstractHuman peripheral blood lymphocytes were separated into subpopulations enriched or depleted with respect to B lymphocytes (Ig‐bearing cells), T lymphocytes, (cells forming rosettes with sheep erythrocytes: E‐RFC) and Fc receptor‐bearing lymphocytes (EA‐RFC).From the distributions and recoveries of the various cell types it could be concluded that there was very little overlap between Ig‐bearing lymphocytes and EA‐RFC. The latter cells partly belonged to “null” (non‐T, non‐B) cells; it was however demonstrated that 30 % of the EA‐RFC were T cells (E‐RFC).The lytic capacity in antibody‐dependent lymphocytotoxicity (ADL) was shown to correspond with the proportions of EA‐RFC in the various fractions. Non‐T cells showed enhanced ADL activity when compared to the unseparated cells. Purified T cell populations also displayed ADL activity. AbstractThe nature of Ig receptors carried by lipopolysaccharide (LPS)‐stimulated mouse spleen B cells was analyzed by surface iodination, direct antiserum precipitation and polyacrylamide gel electrophoresis. LPS activation led to a rapid decrease of surface IgD to 20 % and 80 % of the original level on days three and five of culture, respectively. AbstractThe anti‐Ig column technique of Wigzell et al.(Scand. J. Immunol.1972.1: 75) was adapted to the separation of mouse cells with different H‐2 antigens. AbstractThe development and growth of transplanted syngeneic Lewis lung carcinoma and their relation to aging was investigated. The Lewis lung carcinoma (3LL) is a metastasizing tumor which developed spontaneously in a C57BL/6J mouse. We found that the frequency and rate of tumor growth were increased when the tumors were implanted in aged as compared to young mice. In order to determine whether this finding involves the participation of the immunological system of the host, spleen cells of mice at various ages were mixed with tumor cells at a ratio of 100:1 and injected into one footpad of syngeneic recipient mice. Spleen cells from older mice were found to strikingly enhance tumor growth in the recipient. Only living T cells were responsible for this enhancement, sincein vitroirradiation of donor spleen cells or use of spleen cells from thymectomized donors abolished this enhancement. The quantitative distribution of various cell populations in the spleen did not vary greatly with age, these results suggesting that a change occurs in the intrinsic properties of lymphoid cells during aging. It is suggested that the tumor‐enhancing properties of aged lymphoid cells are due to autoimmune activity of the cells. AbstractThe cellular nature and specificity of suppressor cells and the mechanisms of tolerance to 2,4‐dinitrophenyl‐l‐fluorobenzene (DNFB) in mice was investigated. Mice tolerized with hapten‐modified self membrane,i.e.dinitrophenylated spleen cells (DNP‐SC), generated suppressor cells as shown by their ability to transfer unresponsiveness to normal animals. Such suppressor cells were specific for DNFB as they did not interfere with sensitization of normal animals with 2,4,6‐trinitro‐1‐chlorobenzene (picryl chloride). These suppressor cells were of the thymus‐derived lymphocyte (T cell) lineage as (a) their activity was abolished by treatment with anti‐Θ serum plus complement, and (b) these cells could not be raised in T cell‐deprived mice. Kinetic studies of the development of tolerance showed discrepancies between the rate of induction of unresponsiveness in the donor (“phenotypic” tolerance) and the ability to transfer tolerance. One day after receiving 5 × 10 7DNP‐SC mice were phenotypically tolerant but could not serve as the donors of suppressor cells. 7 days after tolerization with DNP‐SC mice were still fully tolerant and also contained suppressor cells, which were no longer demonstrable 14 days after tolerization when mice were still phenotypically tolerant. The ability to transfer tolerance was eliminated by pretreatment with cyclophosphamide (Cy). We postulate that two independent mechanisms of tolerance occur after tolerization with DNP‐SC ‐ the rapid induction of clone inhibition and the slower, transient induction of suppressor T cells.
