Solid-phase radioimmunoassay, polyacrylamide gel electrophoresis and thin-layer chromatography were used to compare, identify, and characterize the Lewis antigens from human salivas, using monoclonal antibodies directed to the Leaand Lebdeterminants. Sialylated Leaglycolipid was detected in saliva from individuals with Le(a+b+) and Le(a+b-) phenotypes. Immunoaffinity chromatography of the saliva from individuals with different phenotypes revealed a glycoprotein of molecular weight greater than 200 kD bearing the Lewis antigenic determinants.

Monoclonal antibody R11D10 to human cardiac myosin, which also cross-reacted with canine cardiac myosin, was used to demonstrate in vivo localization and visualization by gamma scintigraphy of experimental myocardial infarction. R11D10 Fab with a Ka of 5 x 108 M-1was labeled with technetium-99m (99mTc) by the dithionite reduction method of technetium pertechnetate, via a bifunctional chelating agent, diethylene triamine pentaacetic acid (DTPA). Uptake of99mTc R11D10 Fab in the infarct can be visualized as early as 2 h after intravenous administration. Comparison of R11D10 uptake to thallium-201, an analogue of potassium which is sequestered by normal myocardium, showed an inverse relation (r = -0.75, -0.87, -0.89), similar to that obtained with125I labeled polyclonal antimyosin Fab. Ratios of R11D10 Fab in the infarct to normal myocardium were as high as 30:1 where access of antibody to antigen was not blood flow limited. However, with severe blood-flow restriction, the ratios were lower at about 10:1. Despite the theoretical limitation of a single epitope per myosin molecule available for binding by R11D10 Fab, the immense excess of myosin in the infarcted myocardium allowed adequate concentration of radiolabeled R11D10 for visualization of the infarct by external gamma scintiscanning.

The major component of the core structure of avian sarcoma leukosis viruses is a 27 kD molecular weight polypeptide, p27. Spleen cells from mice immunized with the Schmidt-Ruppin strain of Rous sarcoma virus (RSV) were fused with mouse myeloma cells (SP2/ 0), and hybridoma cell lines producing monoclonal antibodies to p27 were isolated. The monoclonal antibodies were all of the IgG1subclass with kappa light chains. These antibodies immunoprecipitated p27 and its precursor proteins from extracts of RSV-transformed cells. Reciprocal competitive binding experiments defined five nonoverlapping antigenic determinants within p27. The monoclonal antibodies also immunoprecipitated the transforming protein, p110gag-myc, from avian myelocytomatosis virus transformed cells. Their usefulness in studies of virion maturation and viral oncogenesis is discussed.

From a series of hybridomas that produced monoclonal antibodies reactive with the surface of human embryonal carcinoma cells, two that specifically recognized determinants of the liver/bone/kidney isozyme of alkaline phosphatase were isolated. They did not cross-react with the intestinal or placental isozymes. Phylogenetic studies revealed that both antibodies cross-reacted strongly with liver alkaline phosphatase from higher primates, but exhibited marked differences in their respective cross-reactions with liver alkaline phosphatase from other mammalian species.

Six different monoclonal antibodies (MAbs) to human chorionic gonadotropin (hCG) were evaluated for radioimmunodetection in Golden Syrian Hamsters bearing a human choriocarcinoma in the cheek pouch (188-516 mIU of hCG/g). The affinity of three antibodies when determined were high and five of the antibodies were of IgG1 subclass. After i.v. injection of Staphylococcus Protein A (SPA) affinity purified radioiodinated antibodies, tissue distribution studies demonstrated that antibody 061 had higher target-to-nontarget ratios than the other five antibodies. The mean tumor:muscle ratio of131I-MAb 061 was 27.5 in four animals, where the mean tumor:muscle ratio of four animals injected with131I-labeled control monoclonal IgG1Kappa from MOPC-21 murine myeloma (MOPC-21) was only 2.2. Sharp images of the cheek pouch tumor were obtained at five days following a tracer dose of131I-061 MAb, but not with the same dose of a131I-MOPC-21.

Spleen cells of BALB/c mice immunized with human chorionic gonadotropin (hCG) were fused with NS-1 mouse myeloma cells. A hybrid cell line, clone PE4 secreting monoclonal antibody (MAb) to hCG was isolated by simultaneous screening with both a radioimmunoassay (RIA) and a hemagglutination inhibition assay (HAI). With the aid of the double-antibody radioimmunoassay, it was established that PE4-MAb recognizes the betasubunit of hCG. It shows an affinity constant of 0.4 x 1010L/M and cross-reactivity of 0.1% to other related human glycopeptide hormones. PE4-MAb agglutinates sheep red blood cells coated with hCG and can be used in an HAI assay for hCG. Dual screening procedures have, thus, led to a monoclonal antibody showing high sensitivity in two different assays, and hence useful for the qualitative detection and quantitative determination of hCG by both RIA and HAI methods.
