Cultures of human lens epithelial cells have been established using explants of the lens capsule from different aged donors. Outgrowth from the explants occurs more quickly using material from younger as opposed to older donors. The total number of cells as well as the cumulative surface area covered by cell growth is more extensive with the explants from the lens capsules of younger donors as compared with the amount of cell growth from older donor lens capsules. Thus, there is an inverse correlation between donor age and the replicative capacity of human lens epithelial cells in vitro. The lens epithelial cell cultures from human donors of all ages have a finite replicative potential.

The acyclovir-induced herpes simplex virus Type 1 (HSV-1) strain, R9C2, a double mutant in thymidine kinase (TK) and DNA polymerase (DNA pol), and its parental strain SC16 were compared for their effects on ocular pathology and systemic immunity after unilateral inoculation into the anterior chamber (AC) of BALB/c mouse eyes. Although AC-injected R9C2produced no retinal necrosis (0/18 eyes), this mutant induced active suppression (33–87%) of anti–HSV delayed type hypersensitivity similar to that induced by another HSV strain, KOS. AC-injected parental strain, SC16, caused fatal disease within 7–10 days, and induced bilateral retinal necrosis and suppression of DTH in 100% of the mice. Preimmunization with R9C2protected mice in a dose-dependent fashion from the pathologic and lethal effects of AC-injected parental virus. These data suggest that the immunogenicity of the TK and DNA pol double mutant remains intact despite the decreased ocular and systemic pathogenicity observed after intracameral inoculation.

Sixteen adult New Zealand white rabbits with previously confirmed herpes simplex virus type 1 (HSV-1) infections were stimulated by iontophoresis of 6-hydroxydopamine into the cornea and were followed-up by topical epinephrine treatment to confirm latency. A total of 224 ocular cultures were obtained, of which 73 were positive for HSV during the seven day cycle. Twenty-seven of the 32 eyes (84%) had at least one positive culture.Animals were randomly divided into two treatment groups. Upon repeat stimulation (Cycle 2), concurrent with oral and topical bromovinyl-deoxyuridine (BVDU) therapy, only 3/104 ocular cultures were HSV positive, while 24/112 ocular cultures from placebo–treated animals were positive. Anti-HSV serum titers were comparable before and after BVDU therapy and the HSV isolates from BVDU treated animals did not develop drug resistance (i.e., ED–50 values were approximately 0.1–0.2 ug/ml both before and after therapy). It was concluded that BVDU had a demonstrable therapeutic effect on the recovery of HSV-1 from ocular cultures during intentional reactivation, but the latent ganglionic infection was not eliminated.

Previous light microscopic autoradiographic results from our laboratory have shown a quantitative decrease in ATPase pump site density in dysfunctional corneal endothelium. In order to develop antibody techniques to correlate these findings with electron microscopic localization of pump sites, three types of corneas with dysfunctional endothelium (Fuchs' endothelial dystrophy, aphakic and pseudophakic bullous keratopathy) and two types of corneas with functional endothelium (age matched eye bank and keratoconus) were obtained at the time of transplant surgery. Corneas were fixed, frozen and cryostat sections were incubated in a rabbit kidney ATPase primary antibody followed by incubation in an HRP-labeled secondary antibody. Functional endothelia showed dense antibody labeling along the lateral cell membranes and there was a substantial reduction in labeling along the lateral membranes of dysfunctional endothelia. Positive tissue controls of rabbit kidney showed HRP reaction product in the convoluted tubules. Control tissue, incubated in either non-immune serum or primary antibody pre-absorbed with ATPase, confirmed the specificity by having diminished or no reaction product. These results provide immunohistochemical confirmation of our autoradiographic data showing a quantitative reduction in pump site density on dysfunctional corneal endothelium.
