摘要:
A simple method is described to monitor rabbit and human corneal deturgescence while the tissue is suspended in tissue culture media. The composition of the media is such that the cornea slowly thickens at 4°but exhibits the classic“temperature reversal”thinning phenomenon when incubated at 23°-37°. Each cornea is cultured in a closed eye bank corneal viewing chamber during swelling and deswelling phases of an experiment, minimizing direct handling of the tissue. Optimal conditions for observing corneal deturgescence are described. This protocol has several advantages over corneal perfusion techniques and could be further developed for routine use in eye banks as a functional test of donor suitability for keratoplasty.

Diabetic rats were produced by intravenous injection of streptozotocin. Of these, eleven rats were treated with topical instillation of 0.5% aldose reductase inhibitor (ARI), while ten received vehicle alone. The corneal endothelium of these diabetic rats was examined by specular microscopy and compared to age-matched nondiabetic rats (ten rats). Computerized morphometric analysis of individual cells demonstrated that the endothelium of the untreated diabetic rats had marked polymegathism (increased coefficient of variation in cell area) and pleomorphism (decreased percentage of hexagonal cells), as previously observed in diabetic patients. Similar endothelial changes were also noted in the ARI-treated diabetic rats, but to a significantly lesser extent.These results suggest that topically applied ARI can be effective in reducing morphologic changes of the diabetic endotheliura, and that activation of the sorbitol pathway may be implicated in the etiology of such endothelial changes.

In a subcellular plasma membrane enriched fraction of bovine corneal epithelium, Ca2+stimulated Mg2+dependent ATPase activity was characterized. This membrane fraction was more than 5-fold and 4-fold enriched with 5′-nucleotidase and alkaline phosphatase activities, respectively, relative to the 100,000×g pellet. With 250μM ATP, maximum stimulation of a high affinity form of Ca2+stimulated Mg2+dependent ATPase activity was obtained with 1.7μM free Ca2+. This activation required no exogenously added Mg2+and was unaffected by either 0.1 mM ouabain, 3μM ruthenium red, 20 mM sodium azide or 0.2μg/ml oligomycin. Exogenous calmodulin (6μM) elicited a 53% increase in this activity which was completely inhibited by 300μM trifluoperazine (TFP). These effects of calmodulin and TFP are consistent with the notion of a plasma membrane origin for this activity and also suggest that this activity could be a basis for the regulation of intracellular Ca2+activity in the submicromolar range
