摘要:
To study HSV specific antibody responses during latent HSV Infection and Induced HSV ocular shedding, rabbits were Infected blnocularly with McKrae strain HSV-1. The tlter and class of anti-HSV antibody response in serum, aqueous humor and eye washes were determined sequentially during latent HSV Infection and following Intentional reactivation. In all Instances the only antibody class detected was IgG. The highest anti-HSV titers were in the serum and aqueous humor, whereas a significantly lower level of anti-HSV IgG was found in eye washes. Anti-HSV IgG antibody tlters were consistently 100-fold higher in serum than in the aqueous humor, which suggests an absence of local ocular antibody synthesis. Neither anti-HSV IgM nor secretory IgA antibodies were detected in any samples.

Unidirectional sodium and bicarbonate ion fluxes were measured under short-circuit conditions across rabbit corneal endothelium. A net flux of bicarbonate was measured, direction tears to lens, however no net flux of sodium was measured. From these experiments it seems that the inequality between electrically measured short-circuit current and net trans-endothelial bicarbonate ion flux cannot be bridged by any supposed net sodium flux.

The effect of centrally administered corticotropin-releasing factor (CRF) on intraocular pressure (IOP) was investigated using the technique of ventriculocisternal perfusion. Various amounts of ovine CRF (0.01, 0.033, 0.1 and 0.33 yg/min) were perfused in conscious, male New Zealand albino rabbits for 1 hour. At a dose level of 0.1 or 0.33μg/min, CRF caused an acute IOP decrease of 2.5 mmHg, which lasted for 30 rnin. Perfusions with CRF at 0.033μg/min or higher caused delayed ocular hypotension, which oegan 20 hours after the perfusion and lasted for 2 days. Bolus intravenous injection of 3.3 yg CRF did not cause any IOP changes. These findings demonstrate that CRF can cause a centrally mediated IOP response. Superior cervical ganglionectomy eliminated the acute, not the delayed, CRF effect on IOP, indicating the involvement of ocular sympathetic nerves in the acute IOP effect.

The effects of feeding a diet containingtransfatty acids on the fatty acid composition of plasma membrane phospholipids, fluorescence polarization of diphenylhexatriene (DPH) and adenylate cyclase activity in the exorbital lacrimal glands of rats were studied. Three groups of male, weanling rats were fed semipurified diets containing 20% corn oil (CO), 20% partially hydrogenated soybean oil (PHSBO) (a source oftransfatty acids) and 19% PHSBO + 1% CO. Plasma membranes of the lacrimal glands from rats fed 20% PHSBO showed higher adenylate cyclase activity and lower fluidity as shown by a lower double bond index of the fatty acids of their phospholipids and higher fluorescence polarization of DPH. When 1% CO was included with the diet containing PHSBO, the adenylate cyclase activity and membrane fluidity tended to be normal. The results suggest that feeding of a diet containingtransfatty acids in the absence of sufficient linoleic acid (18:2) can result in a decrease in membrane fluidity and an increase in adenylate cyclase activity in the lacrimal glands.

摘要:
αA2-crystallin is one of the major water-soluble proteins of the mammalian lens. Using a cloned cDNA probe coding for mouseαA2-crystallin and Southern blot hybridization, DNA isolated from a panel of somatic cell hybrids prepared from mouse fibroblasts or mouse spleen cells fused with Chinese hamster fibroblasts was probed to determine the chromosomal localization of theαA2-crystallin structural gene. We have located this gene on mouse chromosome 17.

Timolol of 0.5% was applied in 6 eyes prior to cataract extraction, while 7 eyes were not treated. Aqueous humor was drawn after retrobulbar analgesia with 2% lidocaine and analyzed for epinephrine and norepinephrine using a radioenzymatic assay. Epinephrine levels averaged 15.8±16.2 and 19.3±27.5 pg/ml in the timolol treated group and controls respectively. The corresponding values for norepinephrine were 1.59±0.55 and 1.87±0.90 ng/ml. These differences were not statistically significant.Norepinephrine levels in this study were somewhat higher than in previous reports of catecholamine levels in human aqueous humor and considerably higher than plasma levels of norepinephrine. There was no significant correlation between aqueous humor norepinephrine levels and age in the two groups combined. The mean norepinephrine levels for men and women in the two groups combined were 1.73±0.89 and 1.76±0.56 ng/ml. The difference was not statistically significant. Timolol resulted in a decrease in the intraocular pressure.

A circadian rhythm of intraocular pressure in rabbits could provide a useful model for understanding the daily rhythm of intraocular pressure in humans and for studying mechanisms which regulate intraocular pressure. Our results confirm earlier work showing that New Zealand White rabbits housed in an environment with a lighting cycle of 12 hours light and 12 hours dark have a rhythm of intraocular pressure, and that this rhythm persists in constant dark. We show further that the cycle of light and dark is thezeitgeberfor entrainment of the rhythm of intraocular pressure, and therefore persistence of this rhythm in constant dark establishes it as a circadian rhythm. Cervical ganglionectomy demonstrated that intact sympathetic innervation to the eye is required for maintenance of the normal circadian rhythm of intraocular pressure in rabbits. Intraocular pressure in sympathectomized eyes is no different from control eyes during the light, but is significantly reduced during the dark.

Investigation of lens epithelial cells indicates that under normal conditions, essentially all of the detectable cellular glutathione is in a reduced state. However, exposure to levels of H2O2in the range found in the aqueous fluid of cataract patients causes rapid, very large changes in the glutathione redox ratios. Immediately following short-term exposure to 0.15–0.2 mM H2O2, reduced glutathione drops to 19% of its normal level and the remainder of the total glutathione is found in the oxidized form. Within the next few minutes, the redox ratio returns to normal. However, total glutathione levels remain approximately 20% below normal even one hour after exposure to H2O2. With exposure to a higher concentration of H2O2, a greater loss of glutathione is observed. The results suggest that the glutathione redox ratios change dramatically as a result of oxidative insult but quickly return to normal when the oxidative insult is removed. The formation of mixed glutathione-protein disulfide was also observed but only after long-term (1 hour) exposure to a high level (0.6 mM) of H2O2
