Data of the H218O/H216O ratios of must and wines from the most important German wine areas demonstrate that the statistical scattering of the isotope ratios is large, comparing subsequent years and including tendencies during the harvest period. Before a representative set of vineyards and grapes can be selected, the factors on which the variation depends must be known exactly. The statistical variation which we have measured between the single deliveries of grapes will be preserved during the whole procedure of wine making.

For studying the nitrogen metabolism in plants15N NMR spectroscopy can be used. Forin vivo15N NMR (natural abundance of15N: 0.37%) enrichment of the sample with the isotope15N is compulsory. The detection of time courses of15N assimilation from cells, which are enriched in culture is restricted in scope. Here, a method, the15N pulse labelling NMR spectroscopy, is demonstrated, which permits labelling of different nitrogen compounds in photoautotrophic microorganisms during the NMR spectroscopic measurement. Using an effective illumination system it is possible to maintain photosynthesis in plant samples of high biomass densities in the magnet necessary for ammonia assimilation. The technique thus enables to directly observe ammonia assimilation pathways by application of a15NO3−or15NH4−pulses.

Following intravenous injections of [15N]glycine (97 at.%15N, 400 mg in 1.5 ml isotonic saline) to Lewis rats several organs were excised. The time courses of the15N NMR spectral (40.55 MHz) were measured at 37°C after cold (4°C) or warm (37°C) storage. The application of the Overhauser POE technique yielded signal enhancement by a factor of -2.7 and -1.3 in sceletal and heart muscle, respectively, at 37°C, but no enhancement in liver and kidney.

Ziel der Arbeiten war die Untersuchung von Systemvarianten zur simultanen Bestimmung von Gesamtstickstoff und15N in organischem Probematerial für problemorientierte Anwendungen der15N-Tracertchnik auf Gebieten der Landwirtschaftsforschung, Biologie, Medizin und Umweltforschung unter Benutzung des emissionsspektroskopischen Analysators NOI-6 zur15N-Analyse. Von der Vielzahl der prinzipiell verfügbaren kommerziellen Stickstoffanalysatoren konnten folgende Geräte zur Kopplung mit dem NOI-6 eingesetzt werden:

Die emissionsspektroskopische15N-Analyse ist eine leistungsfähige Methode für15N-Tracerexperimente in Landwirtschaft, Biologie und Medizin. Vorteile des NOI-615N-Analysatorsystems sind die geringe Probemenge (10 μg), die kurze MeBzeit und der hohe Bedienungskomfort. In Koppulung mit einer Verbrennung nach Dumas ist die gleichzeitige Bestimmung der Gesamt-Stickstoffkonzentration und des prozentualen15N-Anteiles möglich.

Untersuchungen zur Resorption und Retention des essentiellen Spurenelements Molybdän (Mo) – wichtig als Molybdänkofaktor für die Funktion der Enzyme Xanthinoxidase, Sulfitoxidase und Aldehyddehydrogenase beim Menschen – liegen derzeit kaum vor. Im Rahmen von Stoffwechseluntersuchungen verwenden wir die Massenspektrometrie mit induktiv gekoppeltem Plasma als Ionenquelle (ICP-MS) zur Bestimmung der stabilen Isotope von Molybdän im Urin und Stuhl bei Erwachsenen. Das Meßverfahren ist bereits für Bioverfügbarkeitsstudien der Spurenelemente Fe, Cu, Zn und Se angewendet worden [1]; es ist jedoch noch nicht zu Molybdän-Tracer-Studien genutzt worden. In unserer Studie wird das Isotopenverhältnis98Mo/100Mo mit ICP-MS untersucht. Die ersten Ergebnisse zeigen, daß die ICP-MS-Bestimmung der stabilen Isotope98Mo und100Mo im Urin und Stuhl beim Menschen ohne störende Interferenzen gelingt. Die Methode ermöglicht die qualitative und quantitative Analyse von98Mo und100Mo im ng/ml und pg/ml-Bereich. Die Präzision der Messungen liegt bei 0,1–1,0%.
