SummaryAnimal studies have shown that zinc and copper are both essential for growth and that maternal zinc deficiency may cause severe fetal skeletal abnormalities.Concentrations of zinc and copper were measured during the second trimester of pregnancy in 122 samples of amniotic fluid from women who were at risk of fetal malformation. Twelve of the fetuses had a neural tube defect and the amniotic fluid zinc concentration in these cases was significantly higher than in 110 normal fetuses (P<0·001). Amniotic fluid copper concentration gradually increased during pregnancy but there were no significant differences between samples from normal fetuses and those from fetuses with neural tube defects. There was a highly significant correlation between amniotic fluid zinc and alpha-fetoprotein concentrations, and this relationship was confirmed in cases with anomalous alpha-fetoprotein results. Serum levels of zinc and copper were not significantly different in 12 women who had an abnormal fetus when compared with 110 women with a normal fetus

SummarySecretory IgA is an immunoglobulin (molecular weight = 400 000) found primarily in secretions. Using a sensitive double-antibody radioimmunoassay, the level of the secretory piece of IgA in amniotic fluid was found to be about 2μg/ml at 15-22 weeks maturity. During the last 10 weeks of pregnancy, the concentration of the secretory piece increases continuously, reaching levels of 15 to 20μg/ml at term. Secretory IgA in body fluids probably has a defensive role against various bacteria. Measurement of the secretory piece in amniotic fluid could possibly be of value in the detection of gastrointestinal and bronchial disorders of the fetus

SummaryConcentrations of the platelet-specific proteinβ-thromboglobulin were measured in the urine of women in the third trimester of pregnancy. Urinary levels ofβ-thromboglobulin were greater than the 95 per cent confidence limit of the normal pregnant group in 7 out of 16 patients with moderate or severe pre-eclampsia, and this elevation could not be fully explained by an overall increase in protein excretion. One out of 15 patients with mild pre-eclampsia also had elevated urinaryβ-thromboglobulin levels. Levels ofβ-thromboglobulin were also elevated in 11 out of 88 pregnant women without hypertension or proteinuria. Eight of these 11 patients had some complication of pregnancy, namely infarcted placenta (3), intra-uterine growth retardation (2), placenta praevia (1), premature rupture of membranes (1) and ante-partum haemorrhage (1). SummaryA sensitive, specific and accurate radioimmunoassay for plasma androstenedione (androst-4-ene-3,17-dione) without chromatography is described. There was good correlation between analyses with and without use of a Sephadex LH20 column. The intra-assay coefficient of variation was 2·7 per cent for 15 samples and the inter-assay precision was 7·1 per cent for 45 samples. The specificity of the antibody was satisfactory when examined with 25 different steroids, testing their ability to compete with [3H]-androstenedione for binding sites. Sensitivity was 5 pg per sample and the blanks were negligible. Normal androstenedione levels (mean±s.d.) were 0·93±0·32 ng/ml (3·24±1·12 nM, n−150) in normal females and 0·83±0·26 ng/ml (2·89±0·91 nM,n= 30) in males
