AbstractA procedure is described in which a wide bore capillary column is used as an alternative to the more traditional packed column for the quantitative analysis of amino acids as theirN‐heptafluorobutyryl isobutyl ester (HBB) derivatives. The column, installed in a gas chromatograph previously configured for use with a packed column, is shown to give good reproducibility by repeated determination of amino acid response factors (RSD values for all amino acids are below 3%). AbstractThe performances of some numerical methods to improve the signal to noise ratio are compared and applied to enhance noisy signals obtained in gas chromatography with capillary columns and a flame Ionization detector. Several methods have been considered: cutoffs In the Fourier transform of the recorded signal; real time numerical filtering; theoretical model curve fitting; and the correlation of a chromatogram recorded from a pseudorandomly injected sample with the pseudorandom injection function. Numerical real time filtering is shown to be the most convenient method when the main periodic component of the noise has been determined by Fourier analysis.

AbstractIncreased O2content of the carrier gas due to connections which were not diffusion tight and long cooling time of the cryotrap due to insufficient insulation were the main drawbacks of a commercial thermodesorption unit intended for coupling to a GC/ mass selective detector. These shortcomings were overcome by redesigning the cryotrap part and testing different seals for the desorption tube. Furthermore, the modified unit can now be easily mounted on various gas chromatographys without any risk of cold spots in the transfer line. A detailed working description of the modifications is given.

AbstractProcedures are described for the TLC and reverse phase HPLC analysis of the antihormonal steroid RU 38486 and its metabolites. Conditions are developed for the HPLC of the pure drug substance and a tablet formulation, and also for following its pharmacokinetics and metabolism.
