The aim of this study was to determine the relative rate of c‐erbB‐2 oncoprotein immuno‐detection on matched fine needle aspiration (FNA) smears and surgical specimens of breast cancer, and to correlate the c‐erbB‐2 expression with the assessment of the DNA ploidy status. the expression of c‐erbB‐2 oncoprotein was evaluated using an immunoalkaline phosphatase technique in 49 breast aspirates (four benign and 45 malignant lesions) and 21 matched surgical specimens. the DNA ploidy status was assessed by densitometric techniques on Feulgen‐stained smears. Fifty‐eight per cent of the smears obtained from 45 malignant lesions and 43% of the 21 corresponding paraffin sections contained cells that were stained by the antibody. the higher incidence of c‐erbB‐2 expression on smears seems to be due mainly to the better antigen preservation in the fresh cytological preparations. the correlation between c‐erbB‐2 oncogene expression and DNA ploidy assessment showed an increased incidence of oncogene expression in aneuploid tu

The product of c‐erbB‐2 gene is detected in a proportion of carcinomas from various sites and is generally associated with a high degree of malignancy. A series of 58 effusions containing malignant cells and 16 cytologically negative serous effusions was assessed by immunocyto‐chemical methods for c‐erbB‐2 expression using the monoclonal antibody NCL‐B11, which recognizes the internal domain of the c‐erbB‐2 oncoprotein. Both alcohol‐fixed smears and cell blocks from formalin‐fixed specimens were used. A crisp, clear cut membrane‐associated positive staining was evident in 51 % (30/58) malignant effusions and was restricted to metastatic adenocarcinomas. Breast and ovarian cancers showed the highest incidence of positivity. Mesotheliomas as well as non‐neoplastic effusions were consistently negative. Paraffin blocks from formalin‐fixed cells displayed a weak immunoreactivity when compared with their alcohol‐fixed counterparts. the study shows that the c‐erbB‐2 oncoprotein can be easily identified in standard cytological smears: it can be of assistance in differentiating adenocarcinomas from mesotheliomas, and in selected cases it can provide a further prognostic indicator, replac

The results of the determination of c‐erbB‐2 gene expression by immunocytochemical staining of cytological aspirates, prepared by cytocentrifugation, have been compared with paraffin‐embedded tissue sections from the same tumour. Our results show equivalent staining in 20/22 cases, with six cases being both scored positive and fourteen cases being both negative. Two samples gave conflicting results. One case was scored as being positive on the cytological aspirate, whereas in the tissue sections taken from the same tumour positive staining was only seen in areas of non‐invasive intraduct carcinoma. This sample was scored as being negative. In another case, cytoplasmic staining with less than 50% of the cells showing any positivity was observed in the cytospin sample, with negative staining in the corresponding tissue section. We conclude that expression of c‐erbB‐2 immunostaining is detectable on cytological preparations prepared by cytocentrifugation but must be interpreted with caution in tumours which may have a large intraduct component or which give predominant cytoplasm

Broncholaveolar lavage (BAL) specimens (n= 213) from AIDS and non‐HIV immunosuppressed patients were investigated for the presence ofPneumocystis cariniiinfection by fluorescence microscopy of Papanicolaou‐stained slides. Alveolar casts, extracellular pneumocysts and phagocytosed cysts and their degradation products in pulmonary alveolar macrophages were identified. the number of phagocytosed pneumocysts within human pulmonary alveolar macrophages was recorded and correlated with the number of extracellular cysts and alveolar casts, in both groups of patients. Both phagocytic and degradation capacity were depressed in AIDS patients. We present four cases of cytomegalovirus (CMV) infection detected by cervical cytology of asymptomatic young women. One patient who had undergone bone marrow transplantation was immunosuppressed, but no factors predisposing to CMV infection were identified in the other three patients. A cervical biopsy specimen from a fifth patient, who was also asymptomatic, demonstrated the locus of CMV infection to be in endocervical gland cells. Immunocytochemical and polymerase chain reaction studies on this biopsy specimen confirmed that the histological changes were caused by CMV. the finding of CMV‐infected cells in cervical cytological or biopsy specimens is a rarity.
