AbstractThe hypothesis was tested that polyacrylamide gel which is presumably of constant fiber structure within a practical range of gel concentrations would yield a linear Ferguson plot in the gel electrophoresis of particles in the size range of 10–100 nm. Polystyrene particles ranging from 22 to 60 nm in radius were subjected to gel electrophoresis on 30% N,N′‐methylenebisacrylamide‐crosslinked polyacrylamide (2.0 to 3.2% gel concentration) containing 50 mM3‐[(3‐cholamidopropyl)‐dimethyl ammonio]‐1‐propane sulfonate (CHAPS). Visualization of the zones required transparent gels which were formed by polymerization both at 50 °C and in the presence of 6Murea. Ferguson plots [log(mobility)vs.gel concentration] were progressively curved with increasing particle size from 22.4 to 59.4 nm radius. As in agarose gel electrophoresis, this curvature comprises one or several sigmoidal curve elements, presumably indicative of particle compression [1, 2]. However, by contrast with agarose gel electrophoresis, the curves can be aligned along a linear, rather than a convex axis, and may therefore be designated as “overall‐linear” in essential agreement with the hypothesis underlying this study. AbstractThe plot of log(mobility)vs.gel concentration for polystyrene particles of 26.9 and 44.2 nm radius in 30% Bis‐crosslinked polyacrylamide below 2.2 %T, containing 22.5% Metrizamide and 6 M urea, is convex. The convex plots of the two particles are essentially parallel. This parallelism suggests a retardation by a mechanism other than sieving. The parallel plots cover a range of polyacrylamide concentrations characterized by progressive fluidity as %T is lowered below 2 %T. The convex curvature renders suspect the previous free mobility and valence determinations in polyacrylamide gel electrophoresis obtained by linear extrapolation of Ferguson plots to 0 %T. Nonlinear extrapolation to 0 % T of plots of log(mobility)vs.gel concentration in the concentration range below 2 %T of 30% Bis‐crosslinked polyacrylamide for the two polystyrene particles, corrected for the effects on mobilities of Metrizamide, urea and a lower ionic strength, provides free mobility values similar to those obtained under otherwise identical conditions on agarose gel electrophore

AbstractUrea‐Triton polyacrylamide gel electrophoresis (UT‐PAGE) has been used to separate, in one dimension, membrane proteins present in red blood cells and hemoglobin chains associated with cell ghosts. The membrane proteins are especially well resolved in the high molecular weight range. The protein fractions were further characterized by two‐dimensional gel electrophoresis with UT‐PAGE in the first dimension followed by sodium dodecyl sulfate (SDS)‐PAGE and phosphorylation in the presence or absence of cAMP. AbstractConventional agarose gel electrophoresis (AGE) in Tris/glycine/barbital buffer (TGVB) could classify the common allotype “F” of factor B (BF) into two kinds of subtypes (Fb1 and F). Analysis of mother‐child pairs in paternity cases confirmed inheritance of these suballeles in the same manner as other alleles at the BF locus. The gene frequencies were estimated at 0.0154 for BF*Fb 1 and 0.1492 for BF*F through the study of 325 unrelated individuals. After treatment of serum samples with zymosan, the polymorphic band of BFFb 1 appeared in the Ba fragment, migrating between those of BFF and BFS. AbstractThe genetic polymorphism of factor B (BF) in four Asian populations was studied by immunofixation after agarose gel electrophoresis (AGE) in both veronal buffer (VB) and Tris/glycine/veronal buffer (TGVB). A suballele of the BF*F and three rare alleles were detected in a Chinese population. The subtype and the rare variants were further characterized by using isoelectric focusing in polyacrylamide gels (PAGIF) and by zymosan treatment of the serum samples. The subtype was identified as BFFb1, first defined in the Japanese and found to occur at a polymorphic frequency. One of the rare variants exhibited an intermediate mobility between BFF and BFS and seemed to be similar to BFF025. PAGIF, however, could clearly discriminate the two variants that had a similar mobility on AGE. The results obtained in this study suggest that conventional electrophoresis is not enough to characterize a new variant and to discriminate it from previously reported variants. The findings obtained on immunofixation after AGE/TGVB and in PAGIF must be added to those obtained by the standard method.

AbstractThe electrophoretic patterns of extracted proteins from human head hair were compared to those of proteins from other human hair species (pubic, breast, beard, arm, leg. axillary and eyebrow hair). The respective patterns (keratin phenotypes) were shown to be uniform throughout one individual's hair. Keratin typing could be performed successfully subsequent to ABH blood group determination by mixed cell agglutination with the same hair material.

AbstractTwo rapid staining procedures using 8‐anilino‐1‐naphthalene sulphonate (ANS) and Nitro Blue Tetrazolium (NBT) were compared with a Coomassie Brilliant Blue R‐250 staining technique. The methods were applied to protein separations using both one‐ (1‐D) and two‐dimensional (2‐D) polyacrylamide gel electrophoresis. The ANS procedure worked well for both 1‐D and 2‐D gels. AbstractHuman serum amyloid A protein in inflammatory sera is easily identified by simple protein staining after sodium dodecyl sulfate polyacrylamide gel electrophoresis as the lowest molecular weight detected proteic band. Western blotting followed by immunoblot has been used to demonstrate this result. By contrast, serum amyloid A protein is not detectable by these combined techniques in normal sera.
