ANTI‐IgA ANTIBODIES OF LIMITED SPECIFICITY IN HEALTHY IgA DEFICIENT SUBJECTS |

SUMMARYTo investigate the subclass and allotype specificity of anti‐IgA antibody synthesis, serum samples from 156 IgA deficient blood donors were screened for anti‐IgA antibodies by passive haemagglutination using IgA proteins of both subclasses (IgA1 and IgA2) as well as allotypes A2m(1) and A2m(2). Anti‐IgA activity was found in 25% (thirty‐nine). Antibodies were class‐specific in 19% (twenty‐nine) and of limited specificity in 6% (ten) of the samples. One unusual serum had anti‐IgA directed solely against IgA2. Its activity against A2m(2) was inhibited not only by A2m(2) protein but also by A2m(1) and by three IgA1 proteins. The anti‐A2m(1) activity of the same sample was inhibited only by A2m(1) proteins. The specific mechanism of IgA deficiency in this sample is discussed as well as the structural differences of the different allo‐ and subtypes of IgA and their relation to the antigenic properties of IgA. Inhibition studies could be performed with only one sample with anti‐IgA1 antibodies, but no allotypes of IgA1 were found. By haemagglutination inhibition, 15% (twenty‐three) of the samples contained minute amounts of IgA1, but no IgA2. None of the samples had only IgA2 or IgA1 and IgA2. Both findings, anti‐IgA antibodies with limited specificity in IgA deficient subjects and minute amounts of IgA of one subclass only in IgA deficient samples, are conceivable if IgA deficiency is caused by selective lack or defect of a subclass of B lymphocytes specifi

SUMMARYRabbit antiallotype sera raised against heavy chain markers sometimes show double precipitin lines with all or some of the corresponding antigens (double and single line phenotypes).In a number of cases the double line phenotypes behave as alleles of the single line phenotypes and this feature allows a genetic and immunochemical analysis of these systems.In three cases that have been analysed, the double line phenotype arises when a precipitatingalocus allotype and a non‐precipitatingdorelocus allotype are present on the same molecule (a1 and d14), (a1 and d11), (a3 and d11). This only happens when the corresponding genes are present on the same chromosome (cisconfiguration) of the diploid pair. SUMMARYAa and Ab allotype frequencies have been investigated in wild rabbit populations throughout Victoria and South‐east South Australia. The overall mean allele frequencies for thirty‐three populations are 0.53:0.12:0.36 for a1:a2:a3and 0.72:0.22:0.05 for b4:b5:b9. The b6allele was not found anywhere. The a1frequency has been found to increase in proportion to increases in mean annual rainfall.While considerable variations exist in allotype and allele frequencies from area to area, the relationships a1>a3>a2and b4>b5>b9are almost always observed. The results are discussed in terms of the historical origins of Australian wild rabbits and possible genetic mechanisms to explain the maintenance of allotype ‘hierarchies’ in large populations breeding at

SUMMARYFourteen species of higher plants were found to contain materials in their fruit or seed extracts which were heat stable, inhibitory for and capable of absorbing anti‐H, ‐A and ‐B agglutinins and also lectins. SUMMARYAn analysis of the HLA data of 100 myasthenia gravis (MG) patients and their first degree relatives and spouses is presented. The family studies confirmed the excess of B8 in the myasthenia gravis patients and showed that the excess was not limited to the A1, B8 haplotype. An excess of B8 homozygous patients was found from the matings of two B8 heterozygous parents, suggesting the existence of a dominant B8 linked MG susceptibility. B8 homozygous individuals run a 4.7 times higher risk of acquiring myasthenia gravis than B8 heterozygous individuals.

SUMMARYNeonatal red blood cells (RBC) of some mouse strains are agglutinable by H‐2 antibodies immediately after birth (early strains, E) while others only on day 3 (late strains, L). Previous genetic analysis led to the conclusion that there are at least three ‘temporal’ genes controlling the timing of agglutinability. In the present study, we attempted to throw some light on the way in which these genes function by defining the phenotypic difference(s) between H‐2 agglutinable (E) and non‐agglutinable (L) neonatal RBC. We found that the EvsL difference was not manifested in two other assays, for in agglutination with lectins (Con A and PHA) both cell types behaved as E while in formation of alloclusters with H‐2 specific lymphocytes both behaved as L. Unlike H‐2aE, H‐2aL (native) neonatal RBC had virtually no detectable antibody‐absorbing capacity, however, following digestion with trypsin or neuraminidase (NANAse) they acquired both antibody‐absorbing capacity and agglutinability. In contrast to it, adult RBC lost agglutinability following GA‐fixation, but this could not be counteracted by trypsinization. Neonatal H‐2aL RBC could be made agglutinable by the action of dimethylsulphoxide (DMSO) or by an admixture of adult H‐2aRBC. The results are thus compatible with a hypothesis that the H‐2 receptors on neonatally non‐agglutinable (L) RBC are not absent, but in a state interfering with agglutinability; a poor accessibility to antibody may be due to a rigid integration of H‐2 sites with the cell membrane which prevents their antibody‐mediated juxtaposition to similarly rigid receptors on other cells. SUMMARYThe presence of H‐2‐linked gene products on spermatozoa and their time of appearance during spermatogenesis was determined. Thymus leukaemia antigen specificities 1, 2 and 3 could not be detected on spermatozoa by absorption of the antisera. Immunofluorescent studies with anti‐Sip sera did not reveal any specific reactivity with target spermatozoa. In contrast, H‐2D antigens were present on somatic as well as germ line components in testes so the time of their first appearance during spermatogenesis could not be precisely specified. Cell separation experiments indicate that H‐2D antigens are present on pachytene spermatocytes and increased in quantity on spermatids.
