SUMMARYMonoclonal antibodies (mAbs) which detect antigens on human red cells are also suitable for testing cells of other species. Such studies may reveal previously unrecognized heterogeneity in antibodies which apparently detect the same antigen on the human red cell surface. Information is also provided on specificities shared amongst several species. Here three anti‐LWaband a variety of Rh‐related antibodies have been tested against the red cells of various primates. One monoclonal anti‐LWabantibody, BS46, reacted with the red cells of gorillas and rhesus monkeys but not those of orang‐utans, baboons or marmosets. In contrast, BS56 and NIM‐M8 reacted with the cells of all these species. Chimpanzee cells, however, reacted only with NIM‐M8. Use of primate cells has shown that all three monoclonal anti‐LWabantibodies recognize different epitopes. These observations may explain early conflicting data concerning primate cells. The difference between the monoclonal anti‐D, D4, and three other anti‐D antibodies, 8G2, 8D6 and 7D10, has been confirmed. The D antigen is apparently confined to the red cells of apes and humans. D4 recognizes a polymorphism in chimpanzees and 8G2, 8D6 and 7D10 recognize a polymorphism in gorillas. Two Rh‐related mAbs, R6A and K70, were also investigated. R6A fails to react with Rhnullcells and reacts more weakly with homozygous ‐D‐ cells than with cells of common Rh phenotypes. K70 reacts weakly with Rhnulland ‐D‐/‐D‐ cells. The antigen detected by R6A is confined to the red cells of humans, gorillas and chimpanzees, while the antigen detected by K70 shows a wider species distribution. SUMMARYDQα and β DNA probes of the human major histocompatibility complex (MHC) were hybridized to restriction enzyme‐digested genomic DNA with the aim of establishing a correspondence between the polymorphisms recognized by classical serology and DNA restriction fragment length polymorphisms (RFLP). InDRhomozygous human cell lines, three distinctPstIfragments were recognized by the DQα probe and fourPstIfragments were recognized by the DQβprobe. Each fragment was associated with a different group of DR antigens. Three allelic forms of either DXα or β genes were identified, but none showed any strong association with DR or DQ. Family segregation analysis at the DNA level further confirmed theDRlinkage of the DQ alleles in estimations of gene frequencies of different alleles of DQα, DQβ, DXα and DXβ. Evidence was presented that the DQα and DQβ allelic forms described at the DNA level correspond to polymorphic determinants at the cell surface which can be defined serologically or in cellular assays. SUMMARYThe monoclonal antibody (mAb) NC1 recognizing a monomorphic DR determinant and the anti‐DQ mAbs PLM2, PLM9, and PLM12 recognizing polymorphic DQ determinants were produced. NCI reacted with the Hl Ll (DR) and H1L2 (DRw52, DRw53 and allelic products) complexes of a DR family, while the specificity of PLM12 was DQw3 (previously called TB21). Both PLM2 and PLM9 reacted with the allodeterminant of TAlO (a subtype Of DQw3). These three mAbs immunoprecipitated the same H2L3 complex of a DQ family. With the aid of these mAbs, DR molecules but not DQw3 molecules were detected on monocytes in significant quantities. The functional roles of these class II molecules in the T cell proliferative responses to three soluble antigens (PPD, mite,Candida) and alloantigens were examined by blocking assaysin vitro. NCI almost completely inhibited T cell proliferative responses against both soluble antigens and allogeneic antigens. In contrast, PLM2, PLM9 and PLM12 showed no significant inhibitory effects at all. These results indicate that DR and DQ antigens are different in their functional roles as well as their serological and immunochemical characteristics and tissue distributions; these three soluble antigens and the alloantigen were presented in association with determinants residing predominantly on two forms of NCl‐reactive molecule, namely the H1L1 and/or HlL2 complex of a DR family but not in association with the DQw3 allodeterminant on the H2L3 complex of a DQ fam

SUMMARYThe cosmid H3.5, containing genes mapping to the murineH‐2 Qaregion, was used to transfect L cells by the calcium phosphate co‐precipitation method. The resultant transfected cells expressed a Qa‐like determinant as detected by an immune serum raised against the transfectant cells and Qa specific monoclonal antibodies. Two‐dimensional gel analysis revealed the expression of a class I‐like heavy chain with a similar molecular mass to the Qa2 antigens of the positive strain B10 and B10.A but with a different isoelectric point. SUMMARYA graft‐versus‐host reaction (GVHR) directed against DBA/2 non‐H‐2 antigens alone can be induced by grafting B10.D2 bone marrow and spleen cells intravenously to heavily irradiated, H‐2dcompatible (DBA/2 x B10.D2)F1adult mice. Under the experimental conditions used, only 0‐l0% of recipients survive, but the survival is greatly increased by donor alloimmunization, a few days prior to grafting, against host‐specific (DBA/2) non‐H‐2 antigens and non‐specific (foreign) H‐2 antigens. The increased survival is mediated by alloimmunization‐activated suppressor cells which can decrease the intensity of the immune reaction developed by normal B10.D2 cells bothin vivo(GVHR) andin vitro(proliferative response measured in mixed lymphocyte culture, MLC). The present experiments were designed to explore the antigenic requirements for inducing suppression. The results showed that in GVHR the protective effect induced by donor alloimmunization against the specific non‐H‐2 antigens, which leads to 70‐80% survival, is due primarily, if not entirely, to immunization against Mls2antigens. Results of MLC experiments confirmed this conclusion, showing that immunization against Mlsaantigens is sufficient to account for the suppressive effect induced by the specific immunization. In addition, they indicated that the nonspecific protective effect induced by donor alloimmunization against foreign H‐2 antigens, which leads to 20‐30% survival, is due to immunization against antigens encoded by theKand/or I region(s) of theH‐2complex; immunization againstDregion e

SUMMARYControl by genes withinH‐2of natural resistance to fully virulent salmonellae in susceptible mice was studied by the typhoid relapse model. Susceptible (Itys), H‐2congenic C57BL/10 (B10) lines were infected with a lethal dose of the virulentS. typhimuriumC5 and rescued from death by ampicillin therapy, inducing a chronic infection. The response to therapy and its cessation, both early and late in the infection, varied in different strains. B10 (H‐Zb) and B10.D2 (H‐2d) responded less well to therapy, and were more prone to relapse on its removal, than B10.A (H‐2a) or B10.M (H‐2f) mice. This haplotype distribution is the same as that previously reported forH‐2linked resistance and susceptibility of similar mice to salmonellae of low virulence.
