SUMMARYThe human Thy‐1 homologue (p25) was characterized biochemically for amino acid composition, sequence and carbohydrate content. Two other forms of the human Thy‐1 molecule were detected and partially characterized. A 40,000 mol.wt. molecule (p40) is the dimer of p25 and its formation is increased by the presence of sodium dodecyl sulphate (SDS). The second form of 16,000 mol.wt. (p16) appears to be a cryptic or breakdown form of p25. Comparison of the amino acid compositions of p25, p40 and p16 isolated from MOLT‐3 cells, with that deduced from the nucleotide sequence of the gene coding for part of the putative T cell antigen receptor, also from MOLT‐3 cells, shows that the Thy‐1 homologue is distinct from, but evolutionary related to, one of the putative T cell antigen receptor polypepti

SUMMARYNIM‐M8 is a monoclonla IgM antibody, specific for the LWabantigen as shown by its reaction with red cells of all donors except those lacking LWa, LWband LWab. Indirect immunofluorescent staining and cell sorter analyses have shown that LWabis present on a subpopulation of human lymphoctes. Cell fractionation studies indicate that subsets of both B and T cells express LWaband it may, therefore, provide a further marker for heterogeneity in these lymphocyte population

SUMMARYRg and Ch typing was performed, by serum inhibition, on 145 families that had been typed for HLA/C4/BF/C2 with a view to assessing partial inhibition (p.i.) of anti‐Rg/Ch and its haplotype associations. Rg p.i. was found predominatly with the C4A*3A*,2,B*QO homoduplicated C4 haplotype and BFF. The original type of Ch p.i. (Nordhagenet al., 1980) was closely associated with the allotype C4B 2, which also occasionally exhibited complete inhibition (c.i.), but this Ch p.i. was also found with the C4A*1,B*QO haplotype (Rittneret al., 1984a). The second type of Ch p.i. (Giles, 1984) was closely associated with the C4B 1 allotype most frequently in the haplotype C4A*6,B*1 but also with C4A*3,B*1. Both types of Ch p.i. are usually found with BF S. The present data indicate that the determinants of Rg and Ch are not directly related to any particular C4 allotype or extended haplotype.Further examples of C4A 1 with Ch and C4B 5 without Ch determinants have been detected and theoretical considerations are discussed as to how they might have arisen from unequal crossovers in homologous regions that result in hybrid protein molecule

SUMMARYMice of different inbred strains were injected intravenously on day O and 6 with autologous platelets incubated in a penicillin solution. After the second injection mice of most strains died immediately with anaphylactic symptoms. Both passive transfer experiments and indirect immunofluorescence studies suggest that a serum factor (probably IgM) is involved in this phenomenon. Only mice with a C3H background were unable to respond to autologous penicillintreated platelets, although C3H platelets did induce a lethal response in B10.A mice. SUMMARYRecently, this laboratory has developed a comprehensive strategy for the systematic localization of all the ‘continous’ antigenic (as well as other binding) sites of complex multivalent protein antigens involved in B and T cell recognition. The strategy depends on the syntheis of consecutive overlapping peptides that together account for the entire protein chain. This strategy was applied here for the localization of the ‘continuous’ T cell recognition sites of hen egg lysozyme. Eight overlapping peptides encompassing the entire protein chain of lysozyme were synthesized and examined for their ability to stimulatein vitroproliferation of T cells from several mouse strains (A/J, H‐2a; BALB/c and DBA/2, H‐2d; B10.BR, H‐2k; DBA/1, H‐2a; SJL, H‐2s) that had been primed with native lysozyme. SUMMARYA comprehensive strategy for the systematic localization of all continuous antigenic sites within a protein has previously been introduced by this laboratory. The strategy consists of studying the immunochemical activity of a series of consecutive synthetic peptides that encompass the entire protein chain and the are uniform in size and in overlap at their N and C‐terminals with neighbouring peptided. By application of this strategy to sperm whale myoglobin, we have been able to delineate the ontinuous sites of T cell recognition of myoglobin in three high responder mouse strains. Thirteen 17‐residue peptides that encompass the entire myoglobin chain and overlap by five residues at both ends were synthesized, purified and characterized. The peptides were examinedin vitrofor their ability to stimulate lymph node cells from myoglobin‐primed DBA/2 (H‐2d), BALB/c (H‐2d) and SJL (H‐2s) mice as well as long‐term cultures of myoglobin‐specific T cells. Several regions of the moleculr (T sites) were founnd to stimulate myoglobin‐primed lymph node cells and myoglobin‐specific long‐term T cell cultures. This strategy has enabled the localization of the full profile of dominant sites of T cell recognition in myoglobin for these mouse strains. Of these T sites, one region, residues 107‐125, was clearly immunodominant in these strains and was found to coincide with the antigenic (i.e. antibody binding) site 4 of myoglobin. Also, other regions stimulated T cells and appeared to coincide with previously known antigenic sites. SUMMARYThe T cell proliferative responses to GLA terpolymers among ofH‐2 a, b, d, k, q, r, andshaplotypes are heterogeneous. Following immunization with GLA20, GlA40and GLA60, mouse T cells reacted well with the three terpolymers. The cross‐reactions of the above T cells to GLA5decreased as the alanine content in the terpolymers (immunogens) increased, whereas the trend of the cross‐reaction to GA40did not change. The RIII strain showed no cross‐reaction with GLA5. Only mice ofH‐2haplotypesqandsfailed to show cross‐reactions to GA40. Mice of theH‐2qhaplotype are genetic non‐responders to GA40, wheras those of theH‐2shaplotype are reponders. Immunization of SJL mice with GA40mixed with GLA40led to reduction of the specific T cell reposne to GA40. This reduction of GA40specific T cell responses in SJL mice by GLA40occurred during the early stage of the immune resonse. The kinetic study showed that is was necessary to inject the GLA40intraperitoneally at least 7 days before GA40in order for the reducion to take place.
