SUMMARYFour groups of children were tested for the distribution of C3 and BF phenotypes: HBV‐infected patients with Gianotti‐Crosti syndrome, with CAH and with glomerulonephritis, and healthy children. The frequency of the phenotype C3F was significantly higher in children with Gianotti‐Crosti syndrome in comparison with healthy children. The frequency of the phenotype BFS was significantly higher in patients with glomerulonephritis than in individuals with CAH. The difference between the frequency of the BFS phenotype in glomerulonephritis patients and that in healthy subjects neared significance. SUMMARYA VH gene segment that could not be assigned to any of the six known VH gene families of the channel catfish was identified in a genomic clone containing VH gene segments. This gene segment (designated VH7.1) exhibited the structural features characteristic of vertebrate VH genes, specifically potential upstream regulatory sequences, a leader sequence split by an intron, a reading frame that could be readily divided into framework and complementarity determining regions, and a 3′ recombination signal sequence. Two regions of nucleotide deletions coupled with degeneracy in the nonamer sequence indicate that this VH gene segment is a pseudogene. Genomic DN A restricted with different enzymes and hybridized under stringent conditions with probes derived from VH7.1 showed that 8‐10 bands were present in Southern blots. Reverse transcriptase PCR approaches were used to determine if any of these related sequences were expressed. Sequence analysis of cloned PCR products indicates that different VH gene segments exhibiting>80% similarity to germline VH7.1 are expressed. Multiple sequence alignments showed that the expressed cVH7a cDNA sequence shared less than 60% nucleotide similarity with representative cDNA sequences from the other known catfish VH gene families. These combined results thus fulfil the criteria for the definition of a new family of catfish VH gene segments. This newly defined, small VH family is designated

SUMMARYTransgenic mice expressingHLA‐DQA1*0301andHLA‐DQB1*0302genes (DQ8) were produced. The transgenes were then transferred into mouse (Ab°) class II negative mice: the only class II molecules expressed in these animals were therefore coded by theHLA‐DQ8genes. Good expression of HLA‐DQ molecules was found. Both CD4+ T cells and DQ8‐specific T‐cell receptor Vβ expressing cells were positively selected in these mice. The HLA‐DQ8 molecules expressed in these animals can present various foreign and self antigens and induce T‐cell proliferationin vitro.These mice will be invaluable in future studies of the structure and functi

SUMMARYSerological HLA types (A, B, C, DRandDQloci) were studied in five different Indian tribes (Cubeo, Tucano, Coreguaje, Embera and Noanama) belonging to two distinct linguistic families. For all the MHC loci, the range of variation among the five tribes was enormous. Two tribes, Cubeo and Tucano, showed a wide spectrum of antigenic specificities which seemed to be due to admixture from non‐tribal groups, while in the other three tribes the polymorphisms of variousHLAloci showed restricted distributions. SUMMARYTrichosanthin (Tk), a plant protein isolated from a Chinese medicinal herb, was shown in our previous experiments to suppress mitogenic, antigenic and allogeneic responses of human lymphocytes. Two non‐toxic pathways, the CD8‐dependent and the CD8‐independent, were identified as being involved in inducing the suppression. In this communication, we report that, in a subgroup of healthy subjects, only the CD8‐dependent pathway was found to operate. In cell cultures from the other subjects, removal of CD8 cells from the culture did not diminish the suppression, implying that the down‐regulatory function of the CD8 cells was not expressed in the presence of Tk. Two types of subject, CD8 mediators (M+) (CD8‐dependent pathway operating) and non‐mediators (M−) (CD8‐independent pathway operating), were thus distinguishable. A ‘mediation index’(MI) was calculated as MI (%) = RR of CD8‐depleted cell culture ‐ RR of non‐CD8‐depleted cell culture. Of 68 unrelated subjects tested, 21 (30.9%) could be classified as M+, with a mean ‘mediation index’ of 13.4±4.0, while 47 (69.1%) were M−, with a mean MI of −5.6±4.3 (t= 17.2,P<0.001). The M+and M−groups exhibited an essentially non‐overlapping bimodal distribution of MI. Among the 40 Caucasoids in the panel, nine of 14 M+subjects were HLA‐DR7, DQ2 (χ2= 14.652,Pc= 0.00084). The two DR7‐DQ3 panel members, however, were M−, suggesting that DQ2 might be associated with M+. Segregation patterns in two families revealed

SUMMARYWe describe a method for rapid matching of HLA‐A, ‐B and ‐C allotypes using simultaneous polymerase chain reaction‐single strand conformation polymorphism (PCR‐SSCP) and heteroduplex analysis. Electrophoresis is performed at ambient temperature without requirements for buffer cooling. SSCP and heteroduplexes are revealed as discrete spatially separated band clusters. UsingHLA‐A, ‐Band ‐C locus‐specific PCR primers, matching for alleles at these loci can be performed in 5h. We tested 17 serologically matched patient‐unrelated donor pairs and found considerable microheterogeneity at the DNA level. SUMMARYThe degree of matching ofHLAgenes between the selected donor and recipient is an important aspect of the selection of unrelated donors for allogeneic bone marrow transplantation (UBMT). The most sensitive methods currently used are serological typing ofHLAclass I genes, mixed lymphocyte culture (MLC), IEF and molecular genotyping ofHLAclass II genes by direct sequencing of PCR products. Serological typing of class I antigenes(A, BandC)fails to detect minor differences demonstrated by direct sequencing of DNA polymorphic regions. Molecular genotyping ofHLAclass I genes by DNA analysis is costly and work‐intensive. To improve compatibility between donor and recipient, we have set up a new rapid and non‐radioisotopic application of the ‘fingerprinting PCR’ technique for the analysis of the polymorphic second exon of theHLAclass IA, Band C genes. This technique is based on the formation of specific patterns (PCR fingerprints) of homoduplexes and heterodu‐plexes between heterologous amplified DNA sequences. After an electrophoretic run on non‐denaturing polyacrylamide gel, differentHLAclass I types give allele‐specific banding patterns.HLAclass I matching is performed, after the gel has been soaked in ethidium bromide or silver‐stained, by visual comparison of patients’ fingerprints with those of donors. Identity can be confirmed by mixing donor and recipient DNAs in an amplification cross‐match. To assess the technique, 10 normal samples, 22 related allogeneic bone marrow transplanted pairs and 10 unrelatedHLA‐AandHLA‐Bserologically matched patient‐donor pairs were analysed forHLAclass I polymorphic regions. SUMMARYThis study is the first to assess the prevalence of HLA B27 in Kuwaiti patients with ankylosing spondylitis (AS) or related spondyloarthropathies compared to healthy controls. Positive HLA B27 was found in nine (25.7%) of 35 patients, but only in 22 (4%) of 544 controls (P =0.000). A significant difference in Cw2 and Cw6 between patients and controls was also found (P<0.01 and 0.000, respectively), suggesting a linkage disequilibrium between B27 and Cw2 (P<0.000) in the Kuwaiti population. These findings suggest that B27‐positive Kuwaitis are at high risk of developing AS and spondyloarthropathies, especially if they carry Cw2 and Cw6.
