Repeated parent-hybrid parabiosis was performed in mice across the H-2 barrier (DBA/2 and C57BL/6 × DBA/2)F1). In further groups of mice, parabiosis was combined with the i.v. administration of disrupted liver and spleen cell suspensions, azathioprine, and preliminary thymectomy in various combinations. Only minor modification of the rejection process, in the form of some attenuation of the anticipated second-set response following reparabiosis, was obtained. There was no true tolerance in the form of stable union. During repeated parabiosis, the anaemia-polycythaemia phenomenon was either entirely absent or appreciably reduced in magnitude, depending on the degree of sensitization produced by the initial parabiosis. A bilateral anaemia developed in several pairs late in the course of repeated parabiotic union, and prolonged anaemia developed frequently in both parental and hybrid strains following death and surgical separation of their partners. These findings in the parent strain mice suggest a hybrid-anti-parent reaction. Such a reaction may be explained by exceptions to the conventional rules governing parent-hybrid reactions, but indirect evidence suggests the involvement of the phenomenon of allogeneic inhibition.

Utilizing a new technique of in vitro arterial perfusion, renal allo-transplant rejection in the inbred rat has been studied. The earliest changes of rejection are detected on day 5 and these consist of scattered, nonperfused areas of cortex. This is consistent with physiological studies showing a normal Hippuran clearance at day 3 after allotransplantation and reduced at day 5 in this same model. By day 7, no cortical filling is observed; at, this time the Hippuran clearance is that of anephric rats. It is concluded that the earliest vascular lesion of functional significance which occurs during allotransplant rejection in this model is located in the preglomerular arterial supply of the cortex.

Recovery of the immune system of mouse radiation chimeras immunized with sheep red blood cells was estimated by enumeration of hemolysin-forming spleen cells and titration of serum hemolytic activity. Both estimates were found to attain normal values in isogenic chimeras. In allogenic chimeras the recovery of the hemolysin titer remained subnormal during the 5 months of observations, and it was attributed to insufficient number of hemolysin-forming cells in the spleen. The correlation between numbers of hemolysin-forming spleen cells and hemolysin titer was introduced as a new criterion to evaluate the recovery of the immune system of mouse radiation chimeras.

Three water-soluble preparations have been obtained from cell membrane fragments of rabbit spleen and lymph nodes and give serological evidence of containing rabbit histocompatibility antigens. These materials have been solubilized by the use of Triton X-100, and, from the water-insoluble fraction of the Triton extract, by snake venom phospholipase and by butanol. The evidence for soluble histo-compatibility antigens in these preparations is given by their reaction with antibodies present in rabbit antiserum to rabbit leucocytes, as indicated by their ability to agglutinate tanned sheep red blood cells (RBC) coated with specifically purified antibody from the antileucocyte serum, by their ability to render tanned RBC agglutinable by the antileucocyte serum, and by their inhibition of the agglutination of antigen-coated RBC by antileucocyte serum. The butanol-solubilized material has about 12 times the specific activity of the phospholipase-solubilized material; both yield approximately the same total amount of soluble antigens. These two materials are entirely excluded from Sephadex G-200, whereas the greater part of the Triton-solubilized is not excluded. On diethylaminoethyl cellulose chromatography of the butanol-solubilized preparation, almost all of the serological activity can be found within approximately half the total protein applied to the column.

Soluble antigen prepared by Triton and butanol from cell membrane fragments of rabbit lymph node and spleen has been shown to be associated with the suppression or rejection of allogeneic lymph node cells by its abilities (1) to inhibit or absorb the suppressive antibody in rabbit antileucocyte serum, (2) to induce actively in normal rabbits the rejection of transferred lymph node cells, and (3) to stimulate production of antibodies suppressive for allogeneic lymph node cells. This can be demonstrated in cell transfer or, in vitro, in the hemolytic antibody plaque test. Analysis by chromatography and sucrose density gradient ultracentrifugation shows that the rejection-associated antigen can be found within approximately half the total protein of the butanol extract, and suggests nonidentity of the antigen detectable by inhibition of suppressive antibody and that detectable serologically. Further evidence of nonidentity of the antigens and antibodies detected is given by substantial disproportionality of titers determined in suppressive and serological tests in primary and secondary antisera to whole allogeneic cells, respectively, and in secondary and tertiary antisera to the butanol-solubilized antigen.

Homograft tolerance was induced in C3H mice by neonatal injection of homologous spleen cells of CBA strain origin. Peritoneal cells, obtained from thioglycolate-treated donors, were able to induce homograft tolerance but were less effective than an equivalent number of spleen cells. Homograft tolerance in this strain combination is established for about 4 months, but then the mice undergo a spontaneous gradual escape from tolerance. Attempts to prolong the tolerant state by thymectomizing 1-month-old tolerant mice, were unsuccessful. Attempts to prematurely abolish homograft tolerance by injection of Freund's adjuvant were also unsuccessful. The hypothesis that the spontaneous escape from tolerance is due to an antigenic change on the part of the donor cells and subsequent failure of the donor cells to maintain tolerance, was disproven. Experimental evidence was obtained for the hypothesis, that skin test grafts are instrumental in initiating a local host-versus-graft reaction which eventually spreads and leads to the gradual loss of tolerance. This evidence consists of findings that when the test-grafting of tolerant mice was delayed, the symptoms signalling the onset of the host-vs-graft reaction was proportionally delayed.

Twenty-nine dogs received renal allografts with simultaneous bilateral nephrectomy. Five recipients were untreated, five were given daily s.c. injections of normal horse serum, 14 received daily injections of horse anti-dog antilymphocytic serum (ALS) at varying intervals before and after transplantation, and five were treated with daily injections of immune globulin prepared from the ALS by sodium sulphate precipitation and diethylaminoethanol cellulose chromatography.The mean survival times of the untreated control animals and those receiving normal horse serum were 9.6 days and 10 days respectively. No animal in these groups survived for more than 12 days.In the ALS-treated dogs, some prolongation of survival was achieved when the serum was started as late as 5 days postoperatively, but the best results were obtained with 5 days' pretreatment and continued postoperative administration, the dogs in this group showing a mean survival of 68.4 days even with a limitation of survival credit to 100 days.Immune globulin admistered on this schedule produced a mean survival of 28 days, so that some immunosuppressive activity was lost in the processing of the serum.

Thalidomide was found to prolong the survival of skin allografts in some strains of mice when administered daily i.p. as a suspension in 0.5 % earboxymethyl cellulose (in normal saline.) The doses employed were 12.5 and 25 mg/mouse/day. Both donors and recipients were treated, commencing 10 days before grafting. No toxicity was noted at these dose levels.
