SUMMARYAntigens of the mouseH-2locus were studied on isolated liver cells by immunoferritin labeling. Cell suspensions were obtained by gentle homogenization of mouse livers that had been perfused with hypertonic sucrose to loosen cell junctions. Hepatocytes showed sparse labeling of H-2 antigens on the order of 1% of that present on peritoneal monocytes. The slight hepatocyte labeling appeared to be specific for theH-2locus, since there was essentially no label on congenic control hepatocytes. Other cells in the liver cell preparations were more heavily labeled. Bile duct epithelial cells showed heavy labeling on their lateral surface membranes, but none on their apical brush borders. The preparations contained another nonparen-chymal cell type that was densely labeled, but it was not positively identified. Spleen lymphocytes added to liver homogenates and carried through the cell isolation procedure with liver cells showed typically heavy ferritin labeling. These observations suggest that the expression of histocompatibility antigens by tissue cells is likely to be quite variable.

SUMMARYVein allografts were studied in the rat using the major his-tocompatibility complex-incompatible DA and Lewis inbred strains. Allografts were performed in the Lewis to DA and DA to Lewis combinations, with Lewis to Lewis isografts serving as controls. Vein grafts were performed by interposing a 1 cm length of fresh iliolumbar vein into a defect of the iliac artery, using microsurgical techniques. The grafts were under observation for 18 weeks for (1) patency, (2) gross structural changes, (3) histological changes, and (4) antibody responses. No immunosuppression was used. All grafts remained patent throughout the period of observation, although aneurysm formation was noted in some allografts toward the end of the observation period. Histologically, allografts and isografts were indistinguishable. In the first 2 weeks, they showed patchy areas of necrosis in the vein walls, with subsequent intimal hyperplasia and medial fibrosis. In both strain combinations a lymphocy-totoxin response was induced in most animals, the response being particularly strong in the DA to Lewis combination. All cytotoxic activity could be absorbed out using red blood cells, suggesting that the specificity of the antibody was mainly or entirely directed against SD antigens.

Lymphocyte proliferation induced in response to transplantation antigens and phytohaemagglutinin (PHA) was inhibited by this agent at doses that failed to inhibit mitogenisis in response to sheep anti-pig IgM, growth of kidney cell monolayers, and leukocyte migration.

SUMMARYMonocytes play a crucial role in the response of lymphocytes to phytomitogens. This role is evident during the very early events after exposure to the mitogen. Thus, significant lymphocyte transformation does occur when monocytes are removed from the culture within a few hours after exposure to mitogen, and the presence of monocytes after the initial period is no longer necessary. Monocytes appear to be required for the early increase in protein synthesis and burst in RNA synthesis following exposure to the mitogen. Monocytes may help in lymphocyte responses partly by longer range, by way of release of soluble humoral factors into culture supernatant fluids. Temperature dependence of the effect of pulse exposure of lymphocytes to these supernatants raises the possibility that membrane receptors on these cells to humoral factors released by monocytes may exist. Monocytes do not appear to act primarily by preserving the viability of lymphocytes in culture.

SUMMARYClinical experience with transplantation of kidneys infected prior to implantation led to a dog experiment designed to delineate the best mode of therapy. Five groups of dogs were homotransplanted and immunosuppressed. The donor kidneys were perfused for 4 hr. Dogs in group 1 were controls, received no gentamicin, and the perfusate for their transplant was sterile. The animals in group 2 received kidneys perfused with gentamicin-sensitiveEscherichia coli-contaminated perfusate and were not protected with gentamicin. Average death in this group was within 4 days, secondary to anastomotic rupture or generalized sepsis. The dogs in group 3 were treated preoperatively and thereafter with gentamicin and received a perfusatecontaminated kidney. They did as well as the controls, with no evidence of sepsis at autopsy 10 days post-transplantation. The animals in group 4 received a perfusate-contaminated kidney but were protected with gentamicin beginning 1 day post-transplantation. Average death was at 6 days; most showed evidence ofE. colisepsis at autopsy. The dogs in group 5 received a kidney, the perfusate of which containedE. coliplus gentamicin. They did as well as the controls. To evaluate the effect of gentamicin in the perfusate, two subgroups of five dogs each were autotransplanted after 4 hr of perfusion with and without gentamicin. Comparison showed no detectable differences in serial serum creatinine levels and in microscopic changes of the transplanted kidneys after 10 days.
