Treatment of chronic homologous disease, induced in young adult (C57BL/6 X DBA/2)F1mice by injections of C57BL/6 spleen cells, was attempted with four agents: 6-mercaptopurine (6-MP), prednisone, methotrexate and total body X-radiation. 6-MP caused acceleration of the disease, regardless of dosage or timing of administration. Prednisone ameliorated homologous disease only when given at the same time as the parental cells. Methotrexate treatment suppressed the disease only when given after the parental cells, at a time when active disease is ordinarily expected. Total body X-radiation (400 r) led to extreme susceptibility to the graft-versus-host reaction, even if given 2 weeks before or 60 days after the parental cells. However, a period approximately 3 weeks after administration of parental cells was radioresistant in that no acceleration of the disease occurred. The accelerating effect of 6-mercaptopurine could not be reversed by administering isologous spleen cells. Amelioration of homologous disease by methotrexate was associated with the acquisition of durable specific immunological tolerance of host antigens by the injected parental cells.

The intrasplenic cloning method for bone marrow cells has been applied to produce populations of defined cell types, of bone marrow and spleen origin. The clones thus produced were found to consist of either erythroid, granuloid, or mixed (erythro- and granulo-) cell lines. No lymphoid elements were represented in the intrasplenic colonies. The immunological, competence of such “cloned” cell populations has been tested. No antibodies to red blood cell agglutinogens or toShigella dys. antigen were formed by “cloned” cells, when tested after inoculalation to X-irradiated recipients. The application of Simonsen's test for the graft-versus-host response has shown that “cloned” cell populations of either bone marrow or spleen origin are not capable of forming a detectable anti-host response. In view of their immunological deficiency, their capacity to confer protection on lethally X-irradiated animals across genetic barriers has been studied. It was demonstrated that “cloned” cells of parental strain origin can confer protection on irradiated F1hybrid recipients. The protective effect obtained—in contrast to lethal secondary disease produced by bone marrow cells of the same genetic origin—was attributed to the immunological incompetence of the “cloned” haemopoietic cells. The chimeras thus produced regained immunological reactivity, as manifested by testing them 3 months following treatment. Whether the cellular basis of this restoration is of host or donor origin has not been demonstrated. The haemopoietic system, however, was found to have reverted to host type between the 2nd and 5th month following treatment. The possibility of avoiding the lethal secondary disease in allogeneic radiation chimeras by the application of “cloned” haemopoietic cells, is indicated and discussed.

摘要:
To study the lymph from efferent vessels, 32 popliteal lymph nodes were drained in normal rabbits and in rabbits stimulated with guinea pig cancellous bone. It was found that under the influence of heterologous grafts the total number of cells in the lymph leaving the regional nodes rises nearly 9-fold. At the same time, a statistically significant increase was found in the percentage of blastic cells in the lymph. No changes were observed in the total proteins and protein fractions in either lymph or peripheral blood serum.
