Runting was produced in newborn (C57BL/6 X DBA/2)F, hybrid mice by the injection of parental (C57BL/6) spleen cells. Intestinal bile salts were measured in runts and littermate controls to assess the physiological significance of the previously described ileal lesion. Bile salt depletion is an early manifestation of this type of graft-versus-host reaction, occurring as soon as 5 days following injection. Loss of bile salts into the colon and attenuation of the bile salt pool may contribute to the clinical syndrome.

Alloantisera that enhance survival of heart allografts in inbred rats were tested for their effect on mixed leukocyte reactions (MLR). Lewis (L) rats, administered 107Brown Norway (BN) rat bone marrow or spleen cells i.v. 7 days prior to transplantation, exhibited significant prolongation of BN graft survival. Passive administration to normal L rats of serum obtained from L rats 7 days after such i.v. immunization similarly prolonged BN graft survival. The unidirectional MLR of normal or presensitized L spleen cells with irradiated BN spleen cells was also suppressed by this serum in comparison with MLRs containing normal L serum. Twenty percent alloantiserum decreased the MLR of unsensitized L with irradiated BN as much as 6-fold. Less suppression was demonstrated with 10% antiserum and none with 5%. Alloantiserum suppression of MLR remained demonstrable in the absence of hemolytic complement. Immuno-logical specificity was demonstrated because “enhancing” serum suppressed the MLR of L to irradiated ACI rat leukocytes to a much smaller degree. When splenic leukocytes from presensitized L rats were washed free of enhancing serum and then incubated in normal L serum, they showed increased proliferative activity in MLR with irradiated BN spleen cells. Since these L cells, after removal of enhancing serum, were very active in the MLR, it indicates that enhancement by serum factors rather than central tolerance was the mechanism of graft prolongation. Thus, inhibition of MLR might be useful as an assay for the presence of enhancing alloantiserum.

The feasibility of grafting thyroid tissue as a test for histocompatibility or immunocompetence has been explored in rats. The viability of a graft was assessed by its ability to concentrate radioiodine. S.c. transplantation was successful in 90% of syngeneic or parental to F1strain combinations. Transplantation under the kidney capsule did not increase the proportion of successful grafts nor improve their functional capacity. The presence of an intact thyroid did not inhibit implantation of a grafted lobe but reduced the amount of radioiodine concentrated by it.

Graft-versus-host (GVH) reaction was induced by allogeneic blood lymphocytes in chick embryos and measured by the resulting splenomegaly. Prior to injection into embryos, lymphocytes were incubated in vitro in the presence of complement with rabbit antisera raised against chicken thymus (ATS), bursa cells (ABS), or immunoglobulin (AGS). Selective elimination of GVH-reactive lymphocytes was achieved by ATS but not by ABS. Cross absorption of ATS with bursal cells further increased its specificity. The antiserum concentration was critical: 10% was inhibitory but 1% enhanced the response. Treatment of lymphocytes with AGS under various conditions failed to inhibit the GVH reaction. Treatment of bone marrow cells with ATS eliminated GVH-reactive lymphocytes but preserved the erythropoietic cells. A possible application of this approach to clinical bone marrow transplantation is apparent.

Tooth germs (first molars) of 4–5-day-old mice were transplanted to the s.c. connective tissue of adult recipients in the following strain combinations: B10 B10. BY (H-1odifference), B10.BY B10 (H-1ddifference), B10 B10.LPa (H-3adifference), B10.LPa B10 (H-3bdifference), B10 B10.129(21M) (H-4adifference), B10.129 (21M) B10 (H-4bdifference), and A.BY B10 (multiple non-H-2difference). The recipients were killed at different intervals from 5 to 280 days after transplantation and the grafts were examined histologically after standard hematoxylin and eosin staining and marking with Procion dyes. In all the single non-H-2differences, the allografts behaved like syngeneic grafts and there was no evidence of immune allograft reaction. After a transient period of adjustment, during which a nonspecific inflammatory response took place, the tooth germs continued to develop and mature. The deviations from normal development could be attributed to the trauma caused by the surgical procedure and to the heterotopic environment of the grafts. Tooth germs transplanted across the multiple non-H-2barrier (A. BY B10), on the other hand, showed evidence of relatively strong immune reaction. They were heavily infiltrated with mononuclear cells and their further development was either completely arrested or seriously distorted.

A microcytotoxicity assay for murine H-2 antigens has been developed. The assay is performed in a microdroplet under mineral oil using 3,000 splenic lymphocytes as target cells, microliter quantities of antisera, and either rabbit or guinea pig complement. Cell death is determined by eosin uptake using phase contrast microscopy. The assay can be completed in 2–3 hr and uses minimal amounts of antisera. The method can also be used to detect microgram quantities of soluble H-2 antigens by their ability to inhibit cytotoxicity.

Preimmunization which occurs as a result of transfusion and pregnancy is postulated to be responsible for most early kidney transplant failures. A cell-mediated cytotoxicity test was evaluated as an assay of sensitization. Statistically significant higher levels of immunization could be shown in 29 patients who were in the process of rejecting, or who had already rejected, a transplant when compared to 23 patients with functioning kidney transplants, 53 patients on hemodialysis waiting for transplants, and 23 normal persons. The ability of lymphocytes to kill allogeneic target lines in culture may therefore be a method by which presensitization can be detected before transplantation and by which the state of immunity can be monitored after transplantation.
